Role of EPAC in axon determination

Het vermogen van een neuron om ruimtelijke asymmetrie te induceren wordt neuronale polariteit genoemd. Neuronale polariteit is een proces dat zeer cruciaal is bij neuronale ontwikkeling en de vorming van zenuwbanen, en fundamenteel voor de hersenfunctie. In dit proefschrift wordt ingegaan op de rol van EPAC bij neuronale polariteit, een eiwit die onder directe controle staat van cyclisch AMP. Hierbij maakten we voornamelijk gebruik van vers in kweek gebrachte hippocampale neuronen afkomstig van muizen hersenen. Farmacologische inhibitie van EPAC en genetische activatie induceren tegenovergestelde effecten in de morfologie van het neuron. Inhibitie leidt tot neuronen met meerdere axonen, terwijl activatie juist leidt tot verkorte axonen alsmede tot een verminderd aantal gepolariseerde neuronen. De gevonden resultaten zijn daarmee overeenstemmend met andere eiwitten die zijn beschreven bij polariteit. Verder onderzoek naar de onderliggende mechanismen leert ons dat Rap1B een belangrijk doeleiwit van EPAC is. We bevestigen hiermee eerder gedane experimenten die zich richten tot Rap1B, een GTPase die vanuit de literatuur al bekend stond om haar rol bij neuronale polariteit. De robuustheid van EPAC in dit proces hebben we verder aangetoond door te kijken naar neuroblastoma cellijnen. Ook in deze cellijn kan activatie van EPAC leiden tot meerdere axon-achtige structuren, een proces wat voorheen enkel werd toegewezen aan PKA, een ander doeleiwit van cyclisch AMP. Het werk wat wordt beschreven in dit proefschrift biedt een beter begrip in de mechanismen die betrokken zijn bij neuronale polariteit en de ontwikkeling van het axon. De centrale rol van cyclisch AMP in dit proces beperkt zich niet alleen tot PKA. EPAC, tot voor kort nog onbekend in dit proces, speelt een cruciale rol bij cyclisch AMP-gedreven neuronale polariteit

Modeling Digital Twin Data and Architecture: A Building Guide with FIWARE as Enabling Technology

The use of Digital Twins in the industry has become a growing trend in recent years, allowing to improve the lifecycle of any process by taking advantage of the relationship between the physical and the virtual world. Existing literature formulates several challenges for building Digital Twins, as well as some proposals for overcoming them. However, in the vast majority of the cases, the architectures and technologies presented are strongly bounded to the domain where the Digital Twins are applied. This article proposes the FIWARE Ecosystem, combining its catalog of components and its Smart Data Models, as a solution for the development of any Digital Twin. We also provide a use case to showcase how to use FIWARE for building Digital Twins through a complete example of a Parking Digital Twin. We conclude that the FIWARE Ecosystem constitutes a real reference option for developing DTs in any domain.Comment: 7 pages, 3 figure

Social networks and traditional metrics of impact in pulmonary medicine journals: a correlation study

Introduction: The Scimago Journal Rank (SJR) impact factor is extensively used. However, as the Internet has become widely available, new metrics are coming into play. Our research aims to determine whether a correlation  between the SJR impact factor and metrics reflecting social media activity does exist.Materials and methods: We have used pulmonary medicine journals indexed in the SJR. Variables of social network usage have been extracted from verified accounts. Bivariate analyses have been performed with the Mann-Whitney U tests, the correlation between social media-derived variables and the SJR impact factor have been assessed with the Spearman correlation coefficient. Results are presented before and after adjustment for the years since the creation of the accounts.Results: From 130 journals, 38 had at least one social network account, Twitter being the most commonly used (22.85%). The H index was higher in journals with social network accounts (Median 60 vs 17; p < 0.01). The global correlation between the SJR and the number of followers on Twitter revealed moderate agreement (r = 0.46; p < 0.01), which was excellent in open access journals (rs = 0.90; p < 0.05).Conclusions: The use of social networks is directly correlated with traditional indicators of scientific impact. The joint use of alternative and traditional metrics may be useful for journals in order to generate strategies aiming to increase their audience, as well as for researchers when deciding about the best option of disseminating their articles
