    Concentration and Bioactivity of Condensed Tannins and Total Phenolics of \u3cem\u3eLespedeza\u3c/em\u3e Species From a Germplasm Collection

    Interest in ‘AU-Grazer’ sericea lespedeza (Lespedeza cuneata) as an anti-parasitic bioactive forage has been growing in the United States, but the concentration and bioactivity of tannins and other phenolics from additional accessions of L. cuneata, as well as other Lespedeza species, have not been evaluated. A study was completed to determine the concentration of extractable condensed tannins (ECT), total CT (TCT), total phenolics (TP), and protein-precipitable phenolics (PPP) of 32 accessions of L. cuneata and 16 additional Lespedeza species from a germplasm collection. The plants were established in small pots in a greenhouse and then transplanted into small field plots at the Fort Valley State University Agricultural Research Station in Georgia. Once established in the field, samples from each plot were collected, freeze-dried, and ground for analysis. The ECT and TCT for L. cuneata accessions averaged 6.6 ± 1.4% and 9.1 ± 1.8%, respectively, while TP were 114.8 ± 33.2 mg/g plant material and PPP averaged 81.5 ± 25.3 mg binding CT/g plant material. For other Lespedeza species, ECT ranged from 3.7 ± 1.3 (L. striate) to 8.8 ± 1.3 (L. frutescens) and TCT from 6.0 ± 0.5 (L. japonica) to 10.8 ± 1.3 (L. frutescens). Total phenolics ranged from 45.3 ± 24.0 (L. striate) to 185.8 ± 43.9 (L. virgata), and PPP from 22.1 ± 71 (L. tomentosa) to 89.0 ± 23.6 (L. virginica). All Lespedeza species evaluated in this study had relatively high levels of CT, with several species as high or higher in TP and PPP (bioactivity) than L. cuneata, suggesting potential increased use of these plants as nutraceutical forages for animals

    Sustainable intensification of cultivated pastures is needed in ruminant production if we are to feed a growing world population expected to exceed 9 billion by 2050. Planting pastures of diverse, and therefore more productive and resilient, plant species has been proposed and researched. Despite illustrative examples from wild grasslands (Hofmann, 1989) and rangelands (Glimp, 1988), very little research and even less application of multiple herbivore species (MHS) in cultivated pastures has followed. We review the specific mechanics of divergent domesticated ruminants and theorize how these could best be combined to sustainably intensify meat, milk and fiber production from cultivated pastures around the world

    If ruminant production from cultivated and natural grasslands is to depend less on petroleum-based products, forage legumes must serve as protein sources. Commercially available legumes for warm-dry climate grasslands are, however, very limited and resources available for developing such legumes are inadequate. Indeterminate flowering and dehiscent seed pods combined with the need for specialized seed harvesting equipment are major impediments (Butler and Muir 2012). Warm climates often present environmental challenges such as poor rainfall distribution, extended dry seasons, temperature extremes and aggressive grass species (Muir et al. 2011). Erosion of indigenous knowledge and replacement with inappropriate land management approaches from moist-temperate regions compound the challenges

    Forage legumes could enhance ruminant production in Latin America far more than they currently do. With a few instructive exceptions, decades invested in domesticating, testing and divulgating pasture and rangeland species have had limited impact. Reasons include lack of end-user involvement in research and development, inadequate commercial seed sources, low persistence under grazing, and substitution with industrial nitrogen fertilizer. Current efforts to improve legume adoption include research on domestication of new species especially natives, grass-legume mixtures, silvopasture, protein banks, and mitigating anti-nutritive components. Future challenges might include a greater focus on economical seed production, establishment in multi-species plantings, persistence under grazing, sustainable intensification, domesticating local germplasm, ecosystems services, multiple uses, harnessing condensed tannins, and greater crop-livestock integration of legumes.     On the relevance of animal behavior to the management and conservation of fishes and fisheries

    There are many syntheses on the role of animal behavior in understanding and mitigating conservation threats for wildlife. That body of work has inspired the development of a new discipline called conservation behavior. Yet, the majority of those synthetic papers focus on non-fish taxa such as birds and mammals. Many fish populations are subject to intensive exploitation and management and for decades researchers have used concepts and knowledge from animal behavior to support management and conservation actions. Dr. David L. G. Noakes is an influential ethologist who did much foundational work related to illustrating how behavior was relevant to the management and conservation of wild fish. We pay tribute to the late Dr. Noakes by summarizing the relevance of animal behavior to fisheries management and conservation. To do so, we first consider what behavior has revealed about how fish respond to key threats such as habitat alteration and loss, invasive species, climate change, pollution, and exploitation. We then consider how behavior has informed the application of common management interventions such as protected areas and spatial planning, stock enhancement, and restoration of habitat and connectivity. Our synthesis focuses on the totality of the field but includes reflections on the specific contributions of Dr. Noakes. Themes emerging from his approach include the value of fundamental research, management-scale experiments, and bridging behavior, physiology, and ecology. Animal behavior plays a key role in understanding and mitigating threats to wild fish populations and will become more important with the increasing pressures facing aquatic ecosystems. Fortunately, the toolbox for studying behavior is expanding, with technological and analytical advances revolutionizing our understanding of wild fish and generating new knowledge for fisheries managers and conservation practitioners.publishedVersio

    Pellino-1 Regulates the Responses of the Airway to Viral Infection

    Exposure to respiratory pathogens is a leading cause of exacerbations of airway diseases such as asthma and chronic obstructive pulmonary disease (COPD). Pellino-1 is an E3 ubiquitin ligase known to regulate virally-induced inflammation. We wished to determine the role of Pellino-1 in the host response to respiratory viruses in health and disease. Pellino-1 expression was examined in bronchial sections from patients with GOLD stage two COPD and healthy controls. Primary bronchial epithelial cells (PBECs) in which Pellino-1 expression had been knocked down were extracellularly challenged with the TLR3 agonist poly(I:C). C57BL/6 Peli1-/- mice and wild type littermates were subjected to intranasal infection with clinically-relevant respiratory viruses: rhinovirus (RV1B) and influenza A. We found that Pellino-1 is expressed in the airways of normal subjects and those with COPD, and that Pellino-1 regulates TLR3 signaling and responses to airways viruses. In particular we observed that knockout of Pellino-1 in the murine lung resulted in increased production of proinflammatory cytokines IL-6 and TNFα upon viral infection, accompanied by enhanced recruitment of immune cells to the airways, without any change in viral replication. Pellino-1 therefore regulates inflammatory airway responses without altering replication of respiratory viruses.</p

    Safety and efficacy of sonothrombolysis for acute ischaemic stroke: a multicentre, double-blind, phase 3, randomised controlled trial

    Background: Pulsed-wave ultrasound increases the exposure of an intracranial thrombus to alteplase (recombinant tissue plasminogen activator), potentially facilitating early reperfusion. We aimed to ascertain if a novel operator-independent transcranial ultrasound device delivering low-power high-frequency ultrasound could improve functional outcome in patients treated with alteplase after acute ischaemic stroke. Methods: We did a multicentre, double-blind, phase 3, randomised controlled trial (CLOTBUST-ER) at 76 medical centres in 14 countries. We included patients with acute ischaemic stroke (National Institutes of Health Stroke Scale score ≄10) who received intravenous thrombolysis (alteplase bolus) within 3 h of symptom onset in North America and within 4·5 h of symptom onset in all other countries. Participants were randomly allocated (1:1) via an interactive web response system to either active ultrasound (2 MHz pulsed-wave ultrasound for 120 min [sonothrombolysis]; intervention group) or sham ultrasound (control group). Ultrasound was delivered using an operator-independent device, which had to be activated within 30 min of the alteplase bolus. Participants, investigators, and those assessing outcomes were unaware of group assignments. The primary outcome was improvement in the modified Rankin Scale score at 90 days in patients enrolled within 3 h of symptom onset, assessed in the intention-to-treat population as a common odds ratio (cOR) using ordinal logistic regression shift analysis. This trial is registered with ClinicalTrials.gov, number NCT01098981. The trial was stopped early by the funder after the second interim analysis because of futility. Findings: Between August, 2013, and April, 2015, 335 patients were randomly allocated to the intervention group and 341 patients to the control group. Compared with the control group, the adjusted cOR for an improvement in modified Rankin Scale score at 90 days in the intervention group was 1·05 (95% CI 0·77–1·45; p=0·74). 51 (16%) of 317 patients in the intervention group and 44 (13%) of 329 patients in the control group died (unadjusted OR 1·24, 95% CI 0·80–1·92; p=0·37) and 83 (26%) and 79 (24%), respectively, had serious adverse events (1·12, 0·79–1·60; p=0·53). Interpretation: Sonothrombolysis delivered by an operator-independent device to patients treated with alteplase after acute ischaemic stroke was feasible and most likely safe, but no clinical benefit was seen at 90 days. Sonothrombolysis could be further investigated either in randomised trials undertaken in stroke centres that are dependent on patient transfer for endovascular reperfusion therapies or in countries where these treatments cannot yet be offered as the standard of care

    First narrow-band search for continuous gravitational waves from known pulsars in advanced detector data

    Spinning neutron stars asymmetric with respect to their rotation axis are potential sources of continuous gravitational waves for ground-based interferometric detectors. In the case of known pulsars a fully coherent search, based on matched filtering, which uses the position and rotational parameters obtained from electromagnetic observations, can be carried out. Matched filtering maximizes the signalto- noise (SNR) ratio, but a large sensitivity loss is expected in case of even a very small mismatch between the assumed and the true signal parameters. For this reason, narrow-band analysis methods have been developed, allowing a fully coherent search for gravitational waves from known pulsars over a fraction of a hertz and several spin-down values. In this paper we describe a narrow-band search of 11 pulsars using data from Advanced LIGO’s first observing run. Although we have found several initial outliers, further studies show no significant evidence for the presence of a gravitational wave signal. Finally, we have placed upper limits on the signal strain amplitude lower than the spin-down limit for 5 of the 11 targets over the bands searched; in the case of J1813-1749 the spin-down limit has been beaten for the first time. For an additional 3 targets, the median upper limit across the search bands is below the spin-down limit. This is the most sensitive narrow-band search for continuous gravitational waves carried out so far
