    Effect of Calyx Extract of Hibiscus sabdariffa against Cadmium-Induced Liver Damage

    This study was conducted to elucidate the protective properties of calyx extract of Hibiscus sabdariffa against cadmium toxicity. A total of 16 male rats grouped into 4 were used for this study that lasted for 2 weeks. Group A animals received water and served as the control. Animals in groups B, C and D received cadmium sulfate 10mg/kg body weight orally, once daily throughout the study period. Group C and D animals additionally received calyx extract of Hibiscus sabdariffa twice daily orally at 2.5 and 4.6g/kg body weight respectively. Animals in group B were observed to have continuous loss of hair and were less active compared with the control. Animals in groups C and D were relatively active. Serum activities of alkaline phosphatase (ALP), alanine transaminase (ALT) and aspartate transaminase (AST) showed significant increase (P<0.05) in group B compared with the control group; indicating the hepatotoxic effect of cadmium. Serum ALT, AST and ALP activities of animals in group C were 24.53±2.80, 53.25±9.46 and 234.60±35.63 respectively. Serum ALT, AST and ALP activities of animals in group D were respectively 22.09±2.05, 46.75±4.50 and 165.60±22.54. Even though the activities of the serum enzymes in groups C and D showed significant increase (P<0.05) as compared with the control, they are significantly lower (P<0.05) compared with group B. There is no significant difference (P<0.05) in serum bilirubin levels for groups C, D and B as compared with the control. The decrease in the activities of serum ALT, AST and ALP of groups C and D that received the calyx extract as compared with group B indicates the potential protective action of the extract against cadmium toxicity.     Photosynthetic gas exchange and chlorophyll in Bambara groundnut (Vigna subterranea l. Verdc.) subjected to water deficit

    The study evaluated the response of two Bambara groundnut (Vigna subterranea L. Verdc.) landraces sourced  from contrasting environments to periods of water deficit initiated at vegetative stage. The two landraces  AHM-753 and Bogor, from Namibia and Indonesia, respectively, were grown in pots and irrigation was withheld 30 days after sowing in stressed group while the control plants were fully watered. The responses were studied  by measurements of relative water content (RWC), photosynthetic gas exchange, and SPAD  chlorophyll. RWCand gas exchange parameters were found to decrease in stressed plants. Significant  (P<0.001) decrease in SPAD chlorophyll concentration was seen only in Bogor landrace. The results showed a possible drought avoidance strategy in Bambara  groundnut in response to water deficit. The results pointed a greater tolerance to the imposed drought in the landrace AHM-753 compared to Bogor suggesting that the origin of the landrace and the environment it is adapted to determine its response to water deficit.Keywords:Bambara groundnut;Chlorophyll; Gas exchange; Photosynthesis, Water defici

    Effect of homogenates of avocado pear (Persea americana) seeds and fluted pumpkin (Telfairia occidentalis) leaves coadministered with anti-tuberculosis drugs on liver enzymes of albino rats

    This study evaluated the effects of aqueous homogenates of avocado pear (Persea americana) seeds and fluted pumpkin (Telfairia occidentalis) leaves co-administered with anti-tuberculosis drug on liver enzymes of albino rats. Twenty (20) albino rats were divided into five (5) groups (of four rats each) designated as I, II, III, IV and V (which is the normal control). A dose of 5.14mg/kg body weight of a first line anti-tuberculosis drug (a combination of isoniazid, rifampicin, pyrazinamide and ethambutol) was administered orally to two (2) rats in group I, II, III, and IV for seven (7) days and the remaining two rats in each group for fourteen (14) days. The homogenates of avocado pear seeds (at a dose of 250mg/kg body weight), fluted pumpkin leaves (at a dose of 250mg/kg BW) and a mixture of avocado pear seeds and fluted pumpkin leaves (at a dose of 250mg/kg BW, and 125mg/kg BW respectively) were co-administered with the anti-TB drug to groups II, III and IV respectively. Group I rats served as the positive control (i.e. treated with only 5.14mg/kg BW of anti-TB drug). The results indicated that the activities of serum AST, ALT and ALP were significantly higher (P<0.05) in rats administered with 5.14 mg/kg BW first line anti-TB drugs only when compared with that of the rats co-administered with the same anti-TB drug and 250mg/kg BW plant homogenates (avocado pear seeds, fluted pumpkin leave, and a combination of the two homogenates). The mean serum AST, ALT and ALP activities were found to have decrease (P<0.05) in groups II, III, and IV rats from the beginning to the end of the treatment periods. Similarly, mean serum activities of AST, ALT and ALP in group III rats decreased (P<0.05) significantly when compared with that of group II rats. Moreover, the serum activities of these enzymes were significantly lower (P<0.05) in group V rats compared to that of groups II and III rats. The rise in the mean serum activities of liver enzymes in group I rats when compared to that of group V rats indicated that the anti-TB drug is hepatotoxic. The findings of this research work predicts that the aqueous homogenates of both avocado pear seeds and fluted pumpkin leaves have hepatoprotective abilities but, the combination of these plant parts shows more hepatoprotective function (i. e. exhibits synergy).Key words: Avocado pear, fluted pumpkin, homogenates, anti-tuberculosis, isoniazid

    Concept Discovery and Argument Bundles in the Experience Web

    In this paper we focus on a particular interesting web user-generated content: people¿s experiences. We extend our previous work on aspect extraction and sentiment analysis and propose a novel approach to create a vocabulary of basic level concepts with the appropriate granularity to characterize a set of products. This concept vocabulary is created by analyzing the usage of the aspects over a set of reviews, and allows us to find those features with a clear positive and negative polarity to create the bundles of arguments.     The information needs of people living with ankylosing spondylitis: a questionnaire survey

    <p>BACKGROUND:Today, health care is patient-centred with patients more involved in medical decision making and taking an active role in managing their disease. It is important that patients are appropriately informed about their condition and that their health care needs are met. We examine the information utilisation, sources and needs of people with ankylosing spondylitis (AS).</p> <p>METHODS: Participants in an existing AS cohort study were asked to complete a postal or online questionnaire containing closed and open-ended questions, regarding their information access and needs. Participants were stratified by age and descriptive statistics were performed using STATA 11, while thematic analysis was performed on open-ended question narratives. Qualitative data was handled in Microsoft Access and explored for emerging themes and patterns of experiences.</p> <p>RESULTS: Despite 73% of respondents having internet access, only 49% used the internet to access information regarding AS. Even then, this was only infrequently. Only 50% of respondents reported accessing written information about AS, which was obtained mainly in specialist clinics. Women were more likely than men to access information (63% (women) 46% (men)) regardless of the source, while younger patients were more likely to use online sources. The main source of non-written information was the rheumatologist. Overall, the respondents felt there was sufficient information available, but there was a perception that the tone was often too negative. The majority (95%) of people would like to receive a regular newsletter about AS, containing positive practical and local information. Suggestions were also made for more information about AS to be made available to non-specialist medical professionals and the general public.</p> <p>CONCLUSIONS: There appears to be sufficient information available for people with AS in the UK and this is mostly accessed by younger AS patients. Many patients, particularly men, choose not to access AS information and concerns were raised about its negative tone. Patients still rely on written and verbal information from their specialists. Future initiatives should focus on the delivery of more positive information, targeting younger participants in particular and increasing the awareness in the general population and wider non-specialist medical community.</p&gt

    Improving ductal carcinoma in situ classification by convolutional neural network with exponential linear unit and rank-based weighted pooling

    Ductal carcinoma in situ (DCIS) is a pre-cancerous lesion in the ducts of the breast, and early diagnosis is crucial for optimal therapeutic intervention. Thermography imaging is a non-invasive imaging tool that can be utilized for detection of DCIS and although it has high accuracy (~88%), it is sensitivity can still be improved. Hence, we aimed to develop an automated artificial intelligence-based system for improved detection of DCIS in thermographs. This study proposed a novel artificial intelligence based system based on convolutional neural network (CNN) termed CNN-BDER on a multisource dataset containing 240 DCIS images and 240 healthy breast images. Based on CNN, batch normalization, dropout, exponential linear unit and rank-based weighted pooling were integrated, along with L-way data augmentation. Ten runs of tenfold cross validation were chosen to report the unbiased performances. Our proposed method achieved a sensitivity of 94.08±1.22%, a specificity of 93.58±1.49 and an accuracy of 93.83±0.96. The proposed method gives superior performance than eight state-of-theart approaches and manual diagnosis.     Genetic determinants of co-accessible chromatin regions in activated T cells across humans.

    Over 90% of genetic variants associated with complex human traits map to non-coding regions, but little is understood about how they modulate gene regulation in health and disease. One possible mechanism is that genetic variants affect the activity of one or more cis-regulatory elements leading to gene expression variation in specific cell types. To identify such cases, we analyzed ATAC-seq and RNA-seq profiles from stimulated primary CD4+ T cells in up to 105 healthy donors. We found that regions of accessible chromatin (ATAC-peaks) are co-accessible at kilobase and megabase resolution, consistent with the three-dimensional chromatin organization measured by in situ Hi-C in T cells. Fifteen percent of genetic variants located within ATAC-peaks affected the accessibility of the corresponding peak (local-ATAC-QTLs). Local-ATAC-QTLs have the largest effects on co-accessible peaks, are associated with gene expression and are enriched for autoimmune disease variants. Our results provide insights into how natural genetic variants modulate cis-regulatory elements, in isolation or in concert, to influence gene expression

    Quantitative Analysis of the Effect of Cancer Invasiveness and Collagen Concentration on 3D Matrix Remodeling

    Extracellular matrix (ECM) remodeling is a key component of cell migration and tumor metastasis, and has been associated with cancer progression. Despite the importance of matrix remodeling, systematic and quantitative studies on the process have largely been lacking. Furthermore, it remains unclear if the disrupted tensional homeostasis characteristic of malignancy is due to initially altered ECM and tissue properties, or to the alteration of the tissue by tumor cells. To explore these questions, we studied matrix remodeling by two different prostate cancer cell lines in a three-dimensional collagen system. Over one week, we monitored structural changes in gels of varying collagen content using confocal reflection microscopy and quantitative image analysis, tracking metrics of fibril fraction, pore size, and fiber length and diameter. Gels that were seeded with no cells (control), LNCaP cells, and DU-145 cells were quantitatively compared. Gels with higher collagen content initially had smaller pore sizes and higher fibril fractions, as expected. However, over time, LNCaP- and DU-145-populated matrices showed different structural properties compared both to each other and to the control gels, with LNCaP cells appearing to favor microenvironments with lower collagen fiber fractions and larger pores than DU-145 cells. We posit that the DU-145 cells' preference for denser matrices is due to their higher invasiveness and proteolytic capabilities. Inhibition of matrix proteases resulted in reduced fibril fractions for high concentration gels seeded with either cell type, supporting our hypothesis. Our novel quantitative results probe the dynamics of gel remodeling in three dimensions and suggest that prostate cancer cells remodel their ECM in a synergistic manner that is dependent on both initial matrix properties as well as their invasiveness

The primary cilium functions as a dual sensor, as mechanosensors and chemosensors. The primary cilia coordinate several essential cell signaling pathways that are mainly involved in cell division and differentiation. A primary cilium malfunction can result in several human diseases. Mechanical loading is sense by mechanosensitive cells in nearly all tissues and organs. With this sensation, the mechanical signal is further transduced into biochemical signals involving pathways such as Akt, PKA, FAK, ERK, and MAPK.     Basic science of osteoarthritis

    Osteoarthritis (OA) is a prevalent, disabling disorder of the joints that affects a large population worldwide and for which there is no definitive cure. This review provides critical insights into the basic knowledge on OA that may lead to innovative end efficient new therapeutic regimens. While degradation of the articular cartilage is the hallmark of OA, with altered interactions between chondrocytes and compounds of the extracellular matrix, the subchondral bone has been also described as a key component of the disease, involving specific pathomechanisms controlling its initiation and progression. The identification of such events (and thus of possible targets for therapy) has been made possible by the availability of a number of animal models that aim at reproducing the human pathology, in particular large models of high tibial osteotomy (HTO). From a therapeutic point of view, mesenchymal stem cells (MSCs) represent a promising option for the treatment of OA and may be used concomitantly with functional substitutes integrating scaffolds and drugs/growth factors in tissue engineering setups. Altogether, these advances in the fundamental and experimental knowledge on OA may allow for the generation of improved, adapted therapeutic regimens to treat human OA.(undefined
