Estimation of Diversity and Community Structure Through Restriction-Fragment-Length-Polymorphism Distribution Analysis of Bacterial 16s Ribosomal-RNA Genes from a Microbial Mat at an Active, Hydrothermal Vent System, Loihi Seamount, Hawaii
PCR was used to amplify (eu)bacterial small-subunit (16S) rRNA genes from total-community genomic DNA. The source of total-community genomic DNA used for this culture-independent analysis was the microbial mats from a deep-sea, hydrothermal vent system, Pele\u27s Vents, located at Loihi Seamount, Hawaii. Oligonucleotides complementary to conserved regions in the 16S rRNA-encoding DNA (rDNA) of bacteria were used to direct the synthesis of PCR products, which were then subcloned by blunt-end ligation into phagemid vector pBluescript II. Restriction fragment length polymorphism patterns, created by using tandem tetrameric restriction endonucleases, revealed the presence of 12 groups of 16S rRNA genes representing discrete operational taxonomic units (OTUs). The rank order abundance of these putative OTUs was measured, and the two most abundant OTUs accounted for 72.9% of all of the 16S rDNA clones. Among the remaining 27.1% of the 16S rDNA clones, none of the 10 OTUs was represented by more than three individual clones. The cumulative OTU distribution for 48 bacterial 16S rDNA clones demonstrated that the majority of taxa represented in the clone library were detected, a result which we assume to be an estimate of the diversity of bacteria in the native hydrothermal vent habitat. 16S rDNA fingerprinting of individual clones belonging to particular OTUs by using an oligonucleotide probe that binds to a universally conserved region of the 16S rDNA fragments was conducted to confirm OTU specificity and 16S rDNA identity

Assessing rotation-invariant feature classification for automated wildebeest population counts
Accurate and on-demand animal population counts are the holy grail for wildlife conservation organizations throughout the world because they enable fast and responsive adaptive management policies. While the collection of image data from camera traps, satellites, and manned or unmanned aircraft has advanced significantly, the detection and identification of animals within images remains a major bottleneck since counting is primarily conducted by dedicated enumerators or citizen scientists. Recent developments in the field of computer vision suggest a potential resolution to this issue through the use of rotation-invariant object descriptors combined with machine learning algorithms. Here we implement an algorithm to detect and count wildebeest from aerial images collected in the Serengeti National Park in 2009 as part of the biennial wildebeest count. We find that the per image error rates are greater than, but comparable to, two separate human counts. For the total count, the algorithm is more accurate than both manual counts, suggesting that human counters have a tendency to systematically over or under count images. While the accuracy of the algorithm is not yet at an acceptable level for fully automatic counts, our results show this method is a promising avenue for further research and we highlight specific areas where future research should focus in order to develop fast and accurate enumeration of aerial count data. If combined with a bespoke image collection protocol, this approach may yield a fully automated wildebeest count in the near future
Notes on distribution and behaviour of the Rufous-winged Sunbird Cinnyris rufipennis
Rufous-winged Sunbird Cinnyris rufipennis is known only from Udzungwa Mountains in central Tanzania where it occurs in nine montane forests. Based on field surveys between 2011 and 2014, supplemented by observations made by all authors since the early 1990s, we provide a map showing its distribution and relative abundance. We estimate the size of the core areas where Rufous-winged Sunbird is common to c. 120 km2 and the total range to about 200 km2. We also provide data that suggests that breeding takes place during the rainy season from November to February with the breeding area usually centred on a forest glade or tree-fall gap. Finally, we describe observations of presumed lekking courtship display with up to three males assembling in a clearing and excitedly hopping from branch to branch, singing aggressively at each other with a single female observing
Bird species richness in the montane evergreen forests of the Udzungwa Mountains, Tanzania
Species richness and relative abundance of montane forest birds in the Udzungwa Mountains are presented for the 11 forests larger than 1 km2. A high positive correlation between the number of montane bird species and the size of the forest is found with the highest species richness recorded in the largest forest. A few small (< 5 km2) forest fragments also support a high richness of forest birds. Their isolation from larger forest tracts is probably relatively recent (within the last 100–200 years) and their high bird species numbers may be partly due to delayed extirpations. Twenty-three restricted range montane forest species were recorded, and many of these were widespread in the Udzungwas. The largest populations of White-winged Apalis Apalis chariessa, Dapple-throat Arcanator orostruthus, Iringa Akalat Sheppardia lowei and Usambara Weaver Ploceus nicolli are most likely in Udzungwa forests.

De novo design of immunoglobulin-like domains
Antibodies, and antibody derivatives such as nanobodies, contain immunoglobulin-like (Ig) β-sandwich scaffolds which anchor the hypervariable antigen-binding loops and constitute the largest growing class of drugs. Current engineering strategies for this class of compounds rely on naturally existing Ig frameworks, which can be hard to modify and have limitations in manufacturability, designability and range of action. Here, we develop design rules for the central feature of the Ig fold architecture—the non-local cross-β structure connecting the two β-sheets—and use these to design highly stable Ig domains de novo, confirm their structures through X-ray crystallography, and show they can correctly scaffold functional loops. Pre-service and in-service teachers’ perceptions on the integration of children’s literature in mathematics teaching and learning in Ireland
The beneficial role that children’s literature plays in facilitating the meaningful integration and advancement of literacy and numeracy in the primary mathematics classroom has been well validated by research findings internationally. In Ireland, supporting the development of literacy and numeracy is a key educational priority. Consequently, a myriad of policy initiatives such as the Literacy and Numeracy for Learning and Life strategy have been introduced. All aim to address concerns about young people’s lack of basic literacy and numeracy skills and to consider new teaching and learning modalities to enhance same. Despite this, no official emphasis is given to incorporating literature in the Irish primary school mathematics curriculum. Therefore, it is pertinent and timely that this study seeks to ascertain pre-service and in-service teachers’ views on the use of literature to support mathematics teaching and learning and to investigate perceived barriers to and enablers for the integration of children’ literature in the mathematics classroom in Ireland. The analysis of the findings will be framed using Ajzen (1991)’s Theory of Planned Behavior (TPB) model. Distribution of lithostratigraphic units within the central block of Yucca Mountain, Nevada: A three-dimensional computer-based model, Version YMP.R2.0
Yucca Mountain, Nevada is underlain by 14.0 to 11.6 Ma volcanic rocks tilted eastward 3{degree} to 20{degree} and cut by faults that were primarily active between 12.7 and 11.6 Ma. A three-dimensional computer-based model of the central block of the mountain consists of seven structural subblocks composed of six formations and the interstratified-bedded tuffaceous deposits. Rocks from the 12.7 Ma Tiva Canyon Tuff, which forms most of the exposed rocks on the mountain, to the 13.1 Ma Prow Pass Tuff are modeled with 13 surfaces. Modeled units represent single formations such as the Pah Canyon Tuff, grouped units such as the combination of the Yucca Mountain Tuff with the superjacent bedded tuff, and divisions of the Topopah Spring Tuff such as the crystal-poor vitrophyre interval. The model is based on data from 75 boreholes from which a structure contour map at the base of the Tiva Canyon Tuff and isochore maps for each unit are constructed to serve as primary input. Modeling consists of an iterative cycle that begins with the primary structure-contour map from which isochore values of the subjacent model unit are subtracted to produce the structure contour map on the base of the unit. This new structure contour map forms the input for another cycle of isochore subtraction to produce the next structure contour map. In this method of solids modeling, the model units are presented by surfaces (structure contour maps), and all surfaces are stored in the model. Surfaces can be converted to form volumes of model units with additional effort. This lithostratigraphic and structural model can be used for (1) storing data from, and planning future, site characterization activities, (2) preliminary geometry of units for design of Exploratory Studies Facility and potential repository, and (3) performance assessment evaluations
