    Functional characterization of the rod visual pigment of the echidna (Tachyglossus aculeatus), a basal mammal

    Monotremes are the most basal egg-laying mammals comprised of two extant genera, which are largely nocturnal. Visual pigments, the first step in the sensory transduction cascade in photoreceptors of the eye, have been examined in a variety of vertebrates, but little work has been done to study the rhodopsin of monotremes. We isolated the rhodopsin gene of the nocturnal short-beaked echidna (Tachyglossus aculeatus) and expressed and functionally characterized the protein in vitro. Three mutants were also expressed and characterized: N83D, an important site for spectral tuning and metarhodopsin kinetics, and two sites with amino acids unique to the echidna (T158A and F169A). The λ max of echidna rhodopsin (497.9 ± 1.1 nm) did not vary significantly in either T158A (498.0 ± 1.3 nm) or F169A (499.4 ± 0.1 nm) but was redshifted in N83D (503.8 ± 1.5 nm). Unlike other mammalian rhodopsins, echidna rhodopsin did react when exposed to hydroxylamine, although not as fast as cone opsins. The retinal release rate of light-activated echidna rhodopsin, as measured by fluorescence spectroscopy, had a half-life of 9.5 ± 2.6 min−1, which is significantly shorter than that of bovine rhodopsin. The half-life of the N83D mutant was 5.1 ± 0.1 min−1, even shorter than wild type. Our results show that with respect to hydroxylamine sensitivity and retinal release, the wild-type echidna rhodopsin displays major differences to all previously characterized mammalian rhodopsins and appears more similar to other nonmammalian vertebrate rhodopsins such as chicken and anole. However, our N83D mutagenesis results suggest that this site may mediate adaptation in the echidna to dim light environments, possibly via increased stability of light-activated intermediates. This study is the first characterization of a rhodopsin from a most basal mammal and indicates that there might be more functional variation in mammalian rhodopsins than previously assume

    Two emerging phenotypes of atypical inclusion body myositis: illustrative cases

    OBJECTIVES: Sporadic inclusion body myositis (IBM) is the most common acquired myopathy in those aged above 50. It is classically heralded by weakness in the long finger flexors and quadriceps. The aim of this article is to describe five atypical cases of IBM, outlining two potential emerging clinical subsets of the disease. METHODS: We reviewed relevant clinical documentation and pertinent investigations for five patients with IBM. RESULTS: The first phenotype we describe is young-onset IBM in two patients who had symptoms since their early thirties. The literature supports that IBM can rarely present in this age range or younger. We describe a second phenotype in three middle-aged women who developed early bilateral facial weakness at presentation in tandem with dysphagia and bulbar impairment followed by respiratory failure requiring non-invasive ventilation (NIV). Within this group, two patients were noted to have macroglossia, another possible rare feature of IBM. CONCLUSIONS: Despite the classical phenotype described within the literature IBM can present in a heterogenous fashion. It is important to recognise IBM in younger patients and investigate for specific associations. The described pattern of facial diplegia, severe dysphagia, bulbar dysfunction and respiratory failure in female IBM patients requires further characterisation. Patients with this clinical pattern may require more complex and supportive management. Macroglossia is a potentially under recognised feature of IBM. The presence of macroglossia in IBM warrants further study, as its presence may lead to unnecessary investigations and delay diagnosis

    Ethical and methodological issues in engaging young people living in poverty with participatory research methods

    This paper discusses the methodological and ethical issues arising from a project that focused on conducting a qualitative study using participatory techniques with children and young people living in disadvantage. The main aim of the study was to explore the impact of poverty on children and young people's access to public and private services. The paper is based on the author's perspective of the first stage of the fieldwork from the project. It discusses the ethical implications of involving children and young people in the research process, in particular issues relating to access and recruitment, the role of young people's advisory groups, use of visual data and collection of data in young people's homes. The paper also identifies some strategies for addressing the difficulties encountered in relation to each of these aspects and it considers the benefits of adopting participatory methods when conducting research with children and young people

    Advances in Above- and In-Water Radiometry, Volume 3: Hybridspectral Next-Generation Optical Instruments

    This publication documents the scientific advances associated with new instrument systems and accessories built to improve above- and in-water observations of the apparent optical properties (AOPs) for a diversity of water masses, including optically complex waters. The principal objective is to be prepared for the launch of next-generation ocean color satellites with the most capable commercial off-the-shelf (COTS) instrumentation in the shortest time possible. The technologies described herein are entirely new hybrid sampling capabilities, so as to satisfy the requirements established for next-generation missions. Both above- and in-water instruments are documented with software options for autonomous control of data collection activities as applicable. The instruments were developed for the Hybridspectral Alternative for Remote Profiling of Optical Observations for NASA Satellites (HARPOONS) vicarious calibration project. The state-of-the-art accuracy required for vicarious calibration also led to the development of laboratory instruments to ensure the field observations were within uncertainty requirements. Separate detailed presentations of the individual instruments provide the hardware designs, accompanying software for data acquisition and processing, and examples of the results achieved

    Selection institutions and war aims

    We explore how the sizes of the winning coalition and selectorate influence the war aims of states. Leaders who answer to a small winning coalition are more likely to seek territorial gain as a way to increase state resources. Nonterritorial war aims produce a commitment problem in that after the war the defeated state may not comply with the victor's demands. States with large winning coalitions are more willing to continue the war to remove the enemy leader as a solution to this commitment problem. Separate detailed presentations of the individual instruments provide the hardware designs, accompanying software for data acquisition and processing, and examples of the results achieved. For the laboratory components, calibration and characterization procedures are described along with an estimation of the sources of uncertainty, which culminates in a full uncertainty budget for the radiometers deployed to the field
