    Absence of Effective Vesicular-Arbuscular Mycorrhizae on Lecythidaceae in a Primary Forest Area of Central Amazonia.

    Lecythidaceae comprise one of the four most common tree families in the Amazon forest, but there have been few studies of vesicular-arbuscular mycorrhizae in this taxon. In this study roots of 31 species in six genera of Lecythidaceae were collected in a primary forest on an acid oxisol. No vesicular-arbuscular mycorrhizae, as indicated by the presence of vesicles or arbuscules were detected. Only some hyfas were found on some species roots, but not enough for increasing the nutrient absorption from the soil. This indicated that under the edaphic condiction and the time which the samples were collected, these species worked as no mycor-rhizal plants. These informations confirm datas found for other researches, and may be important for exploring more efficiently and by a not predatory way, the species of Lecythidaceae on agroforestry systems in the Amazon.As Lecythidaceae compreendem uma das quatro famĂ­lias de ĂĄrvores mais abundantes na floresta amazĂŽnica, mas foram pouco estudadas no que sc refere Ă  simbiose com fungos micorrĂ­zicos dos solos. Em algumas espĂ©cies observou-se uma ausĂȘncia total de fungos nas raĂ­zes, enquanto que em outras, apenas algumas hifas de fungos, mas sem vesĂ­culas ou arbĂșsculos, estruturas que serviriam para confirmar a presença de micorrizas vesĂ­culo-arbusculares efetivas nas mesmas. As poucas bifas encontradas nĂŁo seriam suficientes para aumentar a absorção de nutrientes do solo, indicando que nas condiçÔes edĂĄficas e Ă©pocas de coletas do presente estudo, as plantas se comportaram como nĂŁo micorrizicas.     Slow freezing versus vitrification for the cryopreservation of zebrafish (Danio rerio) ovarian tissue.

    The aim of the present study was to compare the efficiency of vitrification and slow freezing techniques for the cryopreservation of zebrafish ovarian tissue containing immature follicles. In Experiment 1, assessment of cell membrane integrity by trypan blue exclusion staining was used to select the best cryoprotectant solution for each cryopreservation method. Primary growth (PG) oocytes showed the best percentage of membrane integrity (63.5 ± 2.99%) when SF4 solution (2 M methanol + 0.1 M trehalose + 10% egg yolk solution) was employed. The vitrification solution, which presented the highest membrane integrity (V2; 1.5 M methanol + 5.5 M Me2SO + 0.5 M sucrose + 10% egg yolk solution) was selected for Experiment 2. Experiment 2 aimed to compare the vitrification and slow freezing techniques in the following parameters: morphology, oxidative stress, mitochondrial activity, and DNA damage. Frozen ovarian tissue showed higher ROS levels and lower mitochondrial activity than vitrified ovarian tissue. Ultrastructural observations of frozen PG oocytes showed rupture of the plasma membrane, loss of intracellular contents and a large number of damaged mitochondria, while vitrified PG oocytes had intact mitochondria and cell plasma membranes. We conclude that vitrification may be more effective than slow freezing for the cryopreservation of zebrafish ovarian tissue

    Cytotoxicity and DNA damage in the neutrophils of patients with sickle cell anaemia treated with hydroxyurea

    Hydroxyurea (HU) is the most important advance in the treatment of sickle cell anaemia (SCA) for preventing complications and improving quality of life for patients. However, some aspects of treatment with HU remain unclear, including their effect on and potential toxicity to other blood cells such as neutrophils. This study used the measurement of Lactate Dehydrogenase (LDH) and Methyl ThiazolTetrazolium (MTT) and the comet assay to investigate the cytotoxicity and damage index (DI) of the DNA in the neutrophils of patients with SCA using HU.In the LDH and MTT assays, a cytoprotective effect was observed in the group of patients treated, as well as an absence of toxicity. When compared to patients without the treatment, the SS group (n=20, 13 women and 07 men, aged 18-69 years), and the group of healthy individuals (AA) used as a control group (n=52, 28 women and 24 men, aged 19-60 years), The SSHU group (n=21, 11 women and 10 men, aged 19-63 years) showed a significant reduction (p20 months), demonstrating that despite the cytoprotective effects in terms of cell viability, the use of HU can induce DNA damage in neutrophils

    The evolving SARS-CoV-2 epidemic in Africa: Insights from rapidly expanding genomic surveillance

    INTRODUCTION Investment in Africa over the past year with regard to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) sequencing has led to a massive increase in the number of sequences, which, to date, exceeds 100,000 sequences generated to track the pandemic on the continent. These sequences have profoundly affected how public health officials in Africa have navigated the COVID-19 pandemic. RATIONALE We demonstrate how the first 100,000 SARS-CoV-2 sequences from Africa have helped monitor the epidemic on the continent, how genomic surveillance expanded over the course of the pandemic, and how we adapted our sequencing methods to deal with an evolving virus. Finally, we also examine how viral lineages have spread across the continent in a phylogeographic framework to gain insights into the underlying temporal and spatial transmission dynamics for several variants of concern (VOCs). RESULTS Our results indicate that the number of countries in Africa that can sequence the virus within their own borders is growing and that this is coupled with a shorter turnaround time from the time of sampling to sequence submission. Ongoing evolution necessitated the continual updating of primer sets, and, as a result, eight primer sets were designed in tandem with viral evolution and used to ensure effective sequencing of the virus. The pandemic unfolded through multiple waves of infection that were each driven by distinct genetic lineages, with B.1-like ancestral strains associated with the first pandemic wave of infections in 2020. Successive waves on the continent were fueled by different VOCs, with Alpha and Beta cocirculating in distinct spatial patterns during the second wave and Delta and Omicron affecting the whole continent during the third and fourth waves, respectively. Phylogeographic reconstruction points toward distinct differences in viral importation and exportation patterns associated with the Alpha, Beta, Delta, and Omicron variants and subvariants, when considering both Africa versus the rest of the world and viral dissemination within the continent. Our epidemiological and phylogenetic inferences therefore underscore the heterogeneous nature of the pandemic on the continent and highlight key insights and challenges, for instance, recognizing the limitations of low testing proportions. We also highlight the early warning capacity that genomic surveillance in Africa has had for the rest of the world with the detection of new lineages and variants, the most recent being the characterization of various Omicron subvariants. CONCLUSION Sustained investment for diagnostics and genomic surveillance in Africa is needed as the virus continues to evolve. This is important not only to help combat SARS-CoV-2 on the continent but also because it can be used as a platform to help address the many emerging and reemerging infectious disease threats in Africa. In particular, capacity building for local sequencing within countries or within the continent should be prioritized because this is generally associated with shorter turnaround times, providing the most benefit to local public health authorities tasked with pandemic response and mitigation and allowing for the fastest reaction to localized outbreaks. These investments are crucial for pandemic preparedness and response and will serve the health of the continent well into the 21st century

    As Lecythidaceae compreendem uma das quatro famílias de árvores mais abundantes na floresta amazônica, mas foram pouco estudadas no que sc refere à simbiose com fungos micorrízicos dos solos. Foram coletadas raízes de 31 espécies de seis gêneros desta família numa floresta de terra firme da Amazônia Central, próxima a Manaus, sob um latossolo amarelo. Em algumas espécies observou-se uma ausência total de fungos nas raízes, enquanto que em outras, apenas algumas hifas de fungos, mas sem vesículas ou arbúsculos, estruturas que serviriam para confirmar a presença de micorrizas vesículo-arbusculares efetivas nas mesmas. As poucas bifas encontradas não seriam suficientes para aumentar a absorção de nutrientes do solo, indicando que nas condições edáficas e épocas de coletas do presente estudo, as plantas se comportaram como não micorrizicas.
