    Interaction between GroES and Proteasomes: structural and functional characterization\ud

    Interactome is the whole set of molecular interactions in cells. When referred to in terms of proteomics, it concerns protein-protein interaction.\ud These activities are very important to fully understand properties and operations of cellular systems. Recently, together with the application of experimental techniques for the determination of protein interaction networks and protein complexes, a number of computational methods\ud based on sequence and genomic information have emerged. Among these, orthology-based methods assume that two proteins may interact if each one has at least an ancestor in its phylogenetic tree interacting with at least an ancestor of the other.\ud The purpose of this work was the evaluation of the interaction between two potential binding partners, namely bovine MPC and GroES, and presented to main phases: a preliminary in silico study, aimed to the assessment of feasibility of the interaction, and an in vitro study, based on fluorimetric, chromatographic and SPR approaches, directed to the confirmation andcharacterization of the interaction.\u

    Financiarización del régimen de acumulación en Argentina. Un análisis desde la Teoría de la Regulación para el período 2016-2018

    El presente trabajo analiza el cambio del modo de regulación en la economía Argentina desde diciembre de 2015 hasta el mes de julio de 2018 utilizando el marco teórico elaborado por la Teoría de la Regulación Francesa. Mediante el estudio de las formas institucionales régimen monetario y sistema financiero, relación salarial y forma del Estado, se comprueba que a partir de la asunción del gobierno de la alianza Cambiemos tuvo lugar en la Argentina un cambio en el modo de regulación con respecto al del régimen de acumulación de la post convertibilidad, y que las características que adoptó colocan a las finanzas en el lugar jerárquico de la nueva configuración institucional, subordinando a las restantes formas institucionales. Asimismo, el análisis empírico permite afirmar que el nuevo modo de regulación implementado adquiere las características propias que la literatura especializada describe para el modo de regulación de los regímenes de acumulación financiarizados, pero en el caso particular de la Argentina da lugar a una financiarización del régimen de acumulación subordinada al flujo positivo de divisas.Fil: Montecchia, Agustín Alan.     El proceso de desarrollo con igualdad implica, entre otras cuestiones, al cambio estructural (CEPAL, 2012), que conlleva una mayor diversificación productiva, generación de valor e intensificación tecnológica (Katz,2001). El análisis de las relaciones económicas a través de la Matriz Insumo Producto (MIP) permite conocer la estructura productiva y a partir de allí la formulación de políticas públicas tendientes a alcanzar el desarrollo económico (Leontief, 1977).     Financiarización del régimen de acumulación en Argentina. Un análisis del período 2016-2018

    This paper analyzes, from a regulationist approach, the emerging accumulation regime in the argentine economy from the assumption of Cambiemos to the national government since December 2015. It is intended to argue that the government change effectively produced a breakdown in the mode of regulation and that the direction it assumed presented its own characteristics of a finance-led regime of accumulation. Certainly, the institutional form of money and the financial system returned to occupy the hierarchical place of the institutional configuration, as in the stage of the financial rent regime of the last quarter of the 20th century, subordinating the wage relationship, which had regained a leading role during the post-convertibility regime. Thus, by the form and operating logic adopted by the financial system and the wage relationship, the specificities of the financialization process that took place in Argentina are described, concluding that it was a subordinated financialization to the entry of foreign exchange.Este trabajo analiza, desde una perspectiva regulacionista, las características del régimen de acumulación emergente en la economía argentina a partir de la asunción del gobierno nacional por parte de la alianza Cambiemos desde diciembre de 2015. Mediante su análisis, se argumentar que con el cambio de gobierno efectivamente se produjo un cambio en el modo de regulación, que presentó características propias de lo que la literatura especializada denomina régimen de acumulación financiarizado o impulsado por las finanzas. Ciertamente, la forma institucional moneda y el sistema financiero volvieron a ocupar el lugar jerárquico de la configuración institucional, como en la etapa del régimen rentístico financiero del último cuarto del siglo XX, subordinando a la relación salarial, que había recobrado un rol protagónico durante el régimen de la post convertibilidad. El trabajo profundiza en la forma y en la lógica de funcionamiento que adoptaron el sistema financiero y la relación salarial que se describen, las especificidades del proceso de financiarización en Argentina, y concluye que se trató de una financiarización subordinada al ingreso de divisas

    Nasopharyngeal tubes in paediatric anaesthesia: is the flow-dependent pressure drop across the tube suitable for calculating oropharyngeal pressure?

    Background: Nasopharyngeal tubes (NPTs) are useful in paediatric anaesthesia for insufflating oxygen and anaesthetics. During NPT-anaesthesia, gas insufflation provides some positive oropharyngeal pressure that differs from the proximal airway pressure (Paw) owing to the flow-dependent pressure drop across the NPT (ΔPNPT ). Aims: This study aimed to investigate whether ΔPNPT could be used for calculating oropharyngeal pressure during NPT-assisted anaesthesia. Methods: In a physical model of NPT-anaesthesia, using Rohrer's equation, we calculated ΔPNPT for three NPTs (3.5, 4.0, and 5.0, mm inner diameter) under oxygen and several sevoflurane in oxygen combinations in two ventilatory scenarios (continuous positive airway pressure and intermittent positive pressure ventilation). We then calculated oropharyngeal pressure as Paw minus ΔPNPT . Calculated and measured oropharyngeal pressure couples of values were compared with the root mean square deviation (RMSD) to assess accuracy. We also investigated whether oropharyngeal pressure accuracy depends on the NPT diameter, flow rate, gas composition, and leak size. Using ΔPNPT charts, we tested whether ΔPNPT calculation was feasible in clinical practice. Results: When we tested small-diameter NPTs at high flow or high peak inspiratory pressure, Paw measurements markedly overestimated oropharyngeal pressure. Comparing measured and calculated maximum and minimum oropharyngeal pressure couples yielded RMSDs less than 0.5 cmH2 O regardless of ventilatory modality, NPT diameter, flow rate, gas composition, and leak size. Conclusion: During NPT-assisted anaesthesia, Paw readings on the anaesthetic monitoring machine overestimate oropharyngeal pressure especially for smaller-diameter NPTs and higher flow, and to a lesser extent for large leaks. Given the importance of calculating oropharyngeal pressure in guiding NPT-ventilation in clinical practice, we propose an accurate calculation using Rohrer's equation method, or approximating oropharyngeal pressure from flow and pressure readings on the anaesthetic machine using the ΔPNPT charts

    The DAMA/NaI set-up of the DAMA experiment has been operative during seven annual cycles and has investigated several rare processes. In particular, it has been realised in order to investigate the model independent annual modulation signature for Dark Matter particles in the galactic halo. With the total exposure collected in the seven annual cycles (107731 kg day) a model independent evidence for the presence of a Dark Matter particle component in the galactic halo has been pointed out at 6.3 sigma C.L..     DAMA/LIBRA results and perspectives

    The DAMA/LIBRA experiment, running at the Gran Sasso National Laboratory of the I.N.F.N. in Italy, has a sensitive mass of about 250 kg highly radiopure NaI(Tl). It is mainly devoted to the investigation of Dark Matter (DM) particles in the Galactic halo by exploiting the model independent DM annual modulation signature. The present DAMA/LIBRA experiment and the former DAMA/NaI one (the first generation experiment having an exposed mass of about 100 kg) have released so far results corresponding to a total exposure of 1.17 ton yr over 13 annual cycles. They provide a model independent evidence of the presence of DM particles in the galactic halo at 8.9 sigma C.L..     Peroxynitrite-mediated oxidation of the C85S/C152E mutant of dihydrofolate reductase from Escherichia coli: functional and structural effects

    Peroxynitrite is a potent reactive oxygen species that is believed to mediate deleterious protein modifications in a wide variety of neurodegenerative disorders. In this study, we have analysed the effects of oxidative damage induced by peroxynitrite on a cysteine-free mutant of dihydrofolate reductase (SE-DHFR), from a functional and a structural point of view. The peroxynitrite-mediated oxidation results in the inhibition, concentration-dependent, of the catalytic activity. This effect is strongly influenced by the HCO(3)(-)/CO(2) buffering system, that we observed to significantly affect the yield of protein oxidation by modulating the peroxynitrite-induced modification of aromatic residues. Because of this effect, in presence of bicarbonate system, we have observed a protection of enzymatic activity of SE-DHFR with regard to peroxynitrite. The thermodynamic stability of the oxidized protein has been studied in comparison with the non-oxidized protein by differential scanning calorimetry. The thermodynamic parameters obtained showed a decrease of stability of SE-DHFR upon oxidation, evaluated in terms of Gibbs free energy of about 1.25 kcal/mol at 25 degrees C, with respect to the non-oxidized protein. Together, these data indicate that structural and functional alterations induced by peroxynitrite may play a direct role in compromising DHFR function in multiple pathological conditions
