    Exploring Yoga as a Leisure-Stress Coping Strategy Among Adults with Intellectual and Developmental Disabilities

    Individuals with intellectual and developmental disabilities (IDD) are at a greater risk for experiencing stress, particularly stress associated with negative interpersonal relations, than individuals without disabilities. Research has found that individuals with IDD often use maladaptive coping strategies to manage stress. The purpose of this study was to explore whether participation in a seven-and-a-half week yoga intervention served as a leisure-stress coping strategy for adults with IDD. Using a multi-method research design, six participants completed pre- and post-Lifestress Inventory assessments, and nine participants completed post-intervention semi-structured interviews focused on yoga and coping. Qualitative results indicated that yoga served as an emotion-focused coping strategy for participants, as it enhanced their mood, and provided them a sense of social support. Qualitative data did not suggest that yoga served as a problem-focused coping strategy for participants. While results suggest that yoga has the potential to serve as emotion-focused coping strategy for adults with IDD, it is recommended that yoga be offered in conjunction with other stress management interventions when teaching stress-coping techniques to adults with IDD. Future research should continue to explore adults with IDD’s stress experiences, and the types of coping strategies they find most effective when managing stress

    Organizational Design for Spill Containment in Deepwater Drilling Operations in the Gulf of Mexico: Assessment of the Marine Well Containment Company (MWCC)

    The Deepwater Horizon oil spill in the Gulf of Mexico in April 2010 led to the deaths of 11 workers, a six-month moratorium on deepwater drilling in the Gulf, and nearly three months of massive engineering and logistics efforts to stop the spill. The series of failures before the well was finally capped and the spill contained revealed an inability to deal effectively with a well in deepwater and ultradeepwater. Ensuring that containment capabilities are adequate for drilling operations at these depths is therefore a salient challenge for government and industry. In this paper we assess the Marine Well Containment Company (MWCC), a consortium aimed at designing and building a system capable of containing future deepwater spills in the Gulf. We also consider alternatives for long-term readiness for deepwater spill containment. We focus on the roles of liability and regulation as determinants of readiness and the adequacy of incentives for technological innovation in oil spill containment technology to keep pace with advances in deepwater drilling capability. Liability and regulation can significantly influence the strength of these incentives.     Hydraulic Fracturing on Federal and Indian Lands: An Analysis of the Bureau of Land Management\u27s Revised Proposed Rule

    The federal government controls 700 million acres of subsurface rights (plus 56 million subsurface acres of Indian mineral estate) across 24 states, making it the largest landowner in the nation, and therefore in a position to negotiate lease terms and shape regulations of oil and gas development. The federal Bureau of Land Management’s (BLM) rules on how drilling activity can take place on federal lands essentially dictate terms, making BLM the largest “regulator” of drilling activity in the country. BLM last revised its oil and gas regulations (the Onshore Orders) in the 1980s and early 1990s, well before the recent rapid expansion of shale gas development. To date there are two rounds of proposed revisions, the first issued in 2012 and the most recent issued in May 2013, after BLM received 177,000 comments on the first round. This paper examines the 2013 proposal in several key respects, including the scope and requirements of the new proposal, the substantial changes from the 2012 proposal, and a comparison of BLM’s proposed rules with rules in states with shale gas development and significant federal land holdings, based on earlier work. We find that BLM’s proposal addresses some apparent gaps in state-level regulation and that, generally, BLM rules do not appear to impose significant requirements beyond existing state regulations, at least across the regulatory elements we analyzed and in those states with large federal land holdings

    Locally Enhanced Angiogenesis Promotes Transplanted Cell Survival

    A developing therapy for complete or partial loss of function in various tissues and organs involves transplanting an appropriate cell population, capable of compensating for the existing deficiencies. Clinical application of this type of strategy is currently limited by the death or dedifferentiation of the transplanted cells after delivery to the recipient. A delay in thorough vascularization of the implant area creates an environment low in oxygen and other nutrients, and likely contributes to the initial death of transplanted cells. We have addressed this problem by sustained delivery of vascular endothelial growth factor (VEGF), an initiator of angiogenesis, from a porous polymer matrix utilized simultaneously for cell delivery. As expected from previous studies, VEGF delivered from these constructs elicited an enhanced angiogenic response over a 2-week period when implanted subcutaneously in SCID mice. Hepatocytes implanted using VEGF-containing matrices demonstrated significantly greater survival after 1 week in vivo as compared with cells implanted on matrices without growth factor. The results of this study therefore indicate that enhancing vascularization in the location of transplanted cells promotes their survival.     Branching coral growth and visual health during bleaching and recovery on the central Great Barrier Reef

    Coral reefs are under threat from cumulative impacts such as cyclones, crown-of-thorns starfish (COTS) outbreaks and climate-driven coral bleaching events. Branching corals are more severely impacted by these events than other coral morphologies due to their sensitivity to heat stress and weaker skeletons and COTS preferred prey. The central Great Barrier Reef experienced unprecedented back-to-back bleaching events in 2016 and 2017. This study commenced in 2017 at the peak of heat stress and examined the impact of the heatwave on the survival and recovery of corals by assessing the growth, health (based on the visual health index) and physiological parameters (chlorophyll a, zooxanthellae density, lipid and protein content) of two species, Acropora millepora and Pocillopora acuta (N = 60 colonies for each species). It was conducted across a gradient of turbidity at three reefs, Pandora, Orpheus and Rib, that experienced in April 2017, degree heating weeks (DHW) of 9, 8 and 7, respectively. Orpheus experienced the worst bleaching, based on visual health score, followed by Rib and Pandora. Rib experienced the greatest mortality (78% by Nov 2017); however, this was attributed to the presence of actively feeding crown-of-thorns starfish. Growth rates of A. millepora were almost twice the rate of P. acuta. Both species showed significant seasonal variation with growth of A. millepora and P. acuta 35–40% and 23–33% significantly greater in the summer, respectively. Differences in growth rates were best explained by indicators of energy acquisition. For example, the most important predictor variable in determining higher growth rates and visual health score in A. millepora was chlorophyll a content. For P. acuta, visual health score was the best predictor variable for higher growth rates. This study highlights the important role that chlorophyll a and associated symbionts play in growth and survival in these corals during and after a heat stress event

    d-Tubocurarine and Berbamine: Alkaloids That Are Permeant Blockers of the Hair Cell's Mechano-Electrical Transducer Channel and Protect from Aminoglycoside Toxicity

    Aminoglycoside antibiotics are widely used for the treatment of life-threatening bacterial infections, but cause permanent hearing loss in a substantial proportion of treated patients. The sensory hair cells of the inner ear are damaged following entry of these antibiotics via the mechano-electrical transducer (MET) channels located at the tips of the hair cell’s stereocilia. d-Tubocurarine (dTC) is a MET channel blocker that reduces the loading of gentamicin-Texas Red (GTTR) into rat cochlear hair cells and protects them from gentamicin treatment. Berbamine is a structurally related alkaloid that reduces GTTR labeling of zebrafish lateral-line hair cells and protects them from aminoglycoside-induced cell death. Both compounds are thought to reduce aminoglycoside entry into hair cells through the MET channels. Here we show that dTC (≥6.25 µM) or berbamine (≥1.55 µM) protect zebrafish hair cells in vivo from neomycin (6.25 µM, 1 h). Protection of zebrafish hair cells against gentamicin (10 µM, 6 h) was provided by ≥25 µM dTC or ≥12.5 µM berbamine. Hair cells in mouse cochlear cultures are protected from longer-term exposure to gentamicin (5 µM, 48 h) by 20 µM berbamine or 25 µM dTC. Berbamine is, however, highly toxic to mouse cochlear hair cells at higher concentrations (≥30 µM) whilst dTC is not. The absence of toxicity in the zebrafish assays prompts caution in extrapolating results from zebrafish neuromasts to mammalian cochlear hair cells. MET current recordings from mouse outer hair cells (OHCs) show that both compounds are permeant open-channel blockers, rapidly and reversibly blocking the MET channel with half-blocking concentrations of 2.2 µM (dTC) and 2.8 µM (berbamine) in the presence of 1.3 mM Ca2+ at −104 mV. Berbamine, but not dTC, also blocks the hair cell’s basolateral K + current, IK,neo, and modeling studies indicate that berbamine permeates the MET channel more readily than dTC. These studies reveal key properties of MET-channel blockers required for the future design of successful otoprotectants

    Design, synthesis and biological evaluation of a new series of carvedilol derivatives that protect sensory hair cells from aminoglycoside-induced damage by blocking the mechanoelectrical transducer channel

    Aminoglycosides (AGs) are broad-spectrum antibiotics used for the treatment of serious bacterial infections but have use-limiting side effects including irreversible hearing loss. Here, we assessed the otoprotective profile of carvedilol in mouse cochlear cultures and in vivo zebrafish assays and investigated its mechanism of protection which, we found, may be mediated by a block of the hair cell’s mechanoelectrical transducer (MET) channel, the major entry route for the AGs. To understand the full otoprotective potential of carvedilol, a series of 18 analogues were prepared and evaluated for their effect against AG-induced damage as well as their affinity for the MET channel. One derivative was found to confer greater protection than carvedilol itself in cochlear cultures and also to bind more tightly to the MET channel. At higher concentrations, both carvedilol and this derivative were toxic in cochlear cultures but not in zebrafish, suggesting a good therapeutic window under in vivo conditions

    LOOC UP: Locating and observing optical counterparts to gravitational wave bursts

    Gravitational wave (GW) bursts (short duration signals) are expected to be associated with highly energetic astrophysical processes. With such high energies present, it is likely these astrophysical events will have signatures in the EM spectrum as well as in gravitational radiation. We have initiated a program, "Locating and Observing Optical Counterparts to Unmodeled Pulses in Gravitational Waves" (LOOC UP) to promptly search for counterparts to GW burst candidates. The proposed method analyzes near real-time data from the LIGO-Virgo network, and then uses a telescope network to seek optical-transient counterparts to candidate GW signals. We carried out a pilot study using S5/VSR1 data from the LIGO-Virgo network to develop methods and software tools for such a search.     Rapamycin-mediated mouse lifespan extension: Late-life dosage regimes with sex-specific effects.

    To see if variations in timing of rapamycin (Rapa), administered to middle aged mice starting at 20 months, would lead to different survival outcomes, we compared three dosing regimens. Initiation of Rapa at 42 ppm increased survival significantly in both male and female mice. Exposure to Rapa for a 3-month period led to significant longevity benefit in males only. Protocols in which each month of Rapa treatment was followed by a month without Rapa exposure were also effective in both sexes, though this approach was less effective than continuous exposure in female mice. Interpretation of these results is made more complicated by unanticipated variation in patterns of weight gain, prior to the initiation of the Rapa treatment, presumably due to the use of drug-free food from two different suppliers. The experimental design included tests of four other drugs, minocycline, β-guanidinopropionic acid, MitoQ, and 17-dimethylaminoethylamino-17-demethoxygeldanamycin (17-DMAG), but none of these led to a change in survival in either sex
