O conceito de educação da atenção foi elaborado pelo antropólogo Tim Ingold e remete a sintonia fina do nosso sistema perceptivo e o meio ambiente. Os participantes foram crianças que frequentavam a Escola de Artes Gasparzinho, localizada no município de Palotina-Paraná. As atividades ocorreram em forma de oficinas didáticas e foram desenvolvidas em cinco encontros. Como instrumento de produção de dados recorremos à observação participante, o diário de bordo e a elaboração de desenhos. A análise de dados foi realizada a partir da triangulação de dados e o embasamento teórico da pesquisa. Com base na análise durante os encontros, descritas em um diário de bordo foi possível identificar as experiências compartilhadas, sendo elas, a alegria, a curiosidade, a afetividade e o fluxo de vida no sol

    Messung von Ressourceneffizienz mit der ESSENZ-Methode

    Eine belastbare Methode zu entwickeln, um den Ressourceneinsatz zu bewerten, ist das Ziel dieses Buches. Notwendig ist dies, da das starke Wirtschaftswachstum der letzten Jahrzehnte zu einer intensiven Beanspruchung natürlicher Ressourcen geführt hat. Mit ihrer steigenden Nutzung gehen auch zusätzliche Belastungen der Umwelt sowie Restriktionen der Verfügbarkeit von Ressourcen einher. Aus diesen Gründen ist ein effizienter Einsatz von Ressourcen als wichtiger Beitrag zu einer nachhaltigen Entwicklung erforderlich. Die ESSENZ-Methode (Integrierte Methode zur ganzheitlichen Berechnung/Messung von Ressourceneffizienz) wurde in einer Kooperation der Technischen Universität Berlin mit den Industriepartnern Daimler AG, Deutsches Kupferinstitut Berufsverband e. V., Evonik Industries AG, Siemens AG, ThyssenKrupp Steel Europe AG und Wissenschaftlicher Gerätebau Dr. Ing. Herbert Knauer GmbH entwickelt. Sie unterstützt die umfassende Messung und Bewertung von Ressourceneffizienz innerhalb der vier Dimensionen „Verfügbarkeit“, „Gesellschaftliche Akzeptanz“, „Umweltauswirkungen“ und „Nutzen“. Um diese vier Dimensionen wissenschaftlich abzubilden, werden 21 anwendbare Indikatoren entwickelt und vorgestellt. Die ESSENZ-Methode kann sowohl für die Analyse und Optimierung eines einzelnen Produktes als auch für den Vergleich mehrerer Produktalternativen verwendet werden.     Integrated method to assess resource efficiency – ESSENZ

    With increasing demand of abiotic resources also the pollution of natural resources like water and soil has risen in the last decades due to global industrial and technological development. Thus, enhancing resource efficiency is a key goal of national and international strategies. For a comprehensive assessment of all related impacts of resource extraction and use all three sustainability dimensions have to be taken into account: economic, environmental and social aspects. Furthermore, to avoid burden shifting life cycle based methods should be applied. As companies need operational tools and approaches, a comprehensive method has been developed to measure resource efficiency of products, processes and services in the context of sustainable development (ESSENZ). Overall 21 categories are established to measure impacts on the environment, physical and socio-economic availability of the used resources as well as their societal acceptance. For the categories socio-economic availability and societal acceptance new approaches are developed and characterization factors are provided for a portfolio of 36 metals and four fossil raw materials.     Disturbance of intestinal wound closure leads to insufficient anastomotic healing and is associated with considerable morbidity following colorectal resections. Matrix metalloproteinases (MMPs) play a crucial role in regulation of wound closure. Here fluorescence endoscopy was evaluated for assessment of MMP-2/-9 expression during failed intestinal anastomotic healing.Distal colonic anastomoses were performed as a model for disturbed healing in 36 Balb/c mice. Healing was evaluated endoscopically, macroscopically, and histologically after 1, 3 and 5 days. For detection of MMP-2/-9 expression fluorescence endoscopy (FE) was used following i.v.-administration of a Cy5.5-labeled MMP-2/-9 specific tracer. FE was complemented by quantification of the fluorescence signal using the MS-FX PRO Optical Imaging System. An overall leakage score was calculated and correlated with the results of FE.With increasing incidence of anastomotic leakage from POD1 (17%) to POD5 (83%) the uptake of the MMP tracer gradually increased (signal-to-noise ratio (SNR), POD1: 17.91 ± 1.251 vs. POD3: 30.56 ± 3.03 vs. POD5: 44.8 ± 4.473, P<0.0001). Mice with defective anastomotic healing showed significantly higher uptake compared to non-defective (SNR: 37.37± 3.63 vs. 26.16± 3.635, P = 0.0369). White light endoscopy and FE allowed evaluation of anastomotic healing and visualization of mucosal MMPs in vivo. Using FE based detection of MMPs in the anastomosis, an overall positive predictive value of 71.4% and negative predictive value of 66.6% was calculated for detection of anastomotic leakage.During disturbed anastomotic healing increased expression of MMP-2/-9 was observed in the anastomotic tissue. Fluorescence endoscopy for detection of MMP-2/-9 during the healing process might be a promising tool for early identification of anastomotic leakage

    Evidence for nucleotide excision repair as a modifying factor of O6-methylguanine-DNA methyltransferase-mediated innate chloroethylnitrosourea resistance in human tumor cell lines

    SUMMARY We examined the O 6 -methylguanine-DNA methyltransferase (MGMT) protein as well as MGMT activity levels and the excision repair cross-complementing rodent repair deficiency gene, ERCC2 (XPD), protein levels in 14 human tumor cell lines not selected for chloroethylnitrosourea (CENU) resistance. These results were compared with 1,3-bis-(2-chloroethyl)-1-nitrosourea (BCNU) cytotoxicity and UV light sensitivity. MGMT protein correlated significantly with MGMT activity (r ϭ 0.9497, p ϭ 0.0001). There was no significant linear correlation between BCNU cytotoxicity and MGMT content as determined by both Western analysis (r ϭ 0.139, p ϭ 0.6348) and activity assay (r ϭ 0.131, p ϭ 0.6515). However, MGMT-rich cell lines were found to be more resistant than MGMT-poor cell lines to BCNU (t ϭ 2.2375, p ϭ 0.0225) but not to UV (t ϭ 1.1734, p ϭ 0.1317). Furthermore, the most BCNU-sensitive cell lines were all MGMT-poor. UV sensitivity was significantly correlated to BCNU cytotoxicity (r ϭ 0.858, p ϭ 0.0001). Significant correlations were found between ERCC2 protein levels and BCNU cytotoxicity (r ϭ 0.786, p ϭ 0.0009) or UV sensitivity (r ϭ 0.874, p ϭ 0.0001). Our results confirm that MGMT plays an important role in CENU resistance, but not in UV resistance. The correlation of UV sensitivity with BCNU cytotoxicity suggests that nucleotide excision repair is an important modifying factor of MGMT-mediated innate CENU resistance in human tumor cell lines, especially in highly resistant cell lines. ERCC2 may be implicated in this process

    Spatially controlled cell adhesion on three-dimensional substrates

    The microenvironment of cells in vivo is defined by spatiotemporal patterns of chemical and biophysical cues. Therefore, one important goal of tissue engineering is the generation of scaffolds with defined biofunctionalization in order to control processes like cell adhesion and differentiation. Mimicking extrinsic factors like integrin ligands presented by the extracellular matrix is one of the key elements to study cellular adhesion on biocompatible scaffolds. By using special thermoformable polymer films with anchored biomolecules micro structured scaffolds, e.g. curved and µ-patterned substrates, can be fabricated. Here, we present a novel strategy for the fabrication of µ-patterned scaffolds based on the “Substrate Modification and Replication by Thermoforming” (SMART) technology: The surface of a poly lactic acid membrane, having a low forming temperature of 60°C and being initially very cell attractive, was coated with a photopatterned layer of poly(L-lysine) (PLL) and hyaluronic acid (VAHyal) to gain spatial control over cell adhesion. Subsequently, this modified polymer membrane was thermoformed to create an array of spherical microcavities with diameters of 300 µm for 3D cell culture. Human hepatoma cells (HepG2) and mouse fibroblasts (L929) were used to demonstrate guided cell adhesion. HepG2 cells adhered and aggregated exclusively within these cavities without attaching to the passivated surfaces between the cavities. Also L929 cells adhering very strongly on the pristine substrate polymer were effectively patterned by the cell repellent properties of the hyaluronic acid based hydrogel. This is the first time cell adhesion was controlled by patterned functionalization of a polymeric substrate with UV curable PLL-VAHyal in thermoformed 3D microstructures
