    Evaluation of the Physicochemical, Antioxidant, and Antibacterial Properties of Tunichrome Released from Phallusia nigra Persian Gulf Marine Tunicate

    The aim of this study was to evaluate the physicochemical, nutraceutical, antioxidant, and antibacterial properties of tunichrome released from Persian Gulf tunicate (Phallusia nigra). For this purpose, molecular weight (SDS-PAGE), amino acid profile, chemical composition (GC-MS), mineral composition, functional groups (FTIR), total phenol content (TPC), total flavonoid content (TFC), antioxidant activity, and antimicrobial properties were investigated. The results showed that tunichrome contained a high amount of essential amino acids (i.e., Lys = 32.24 mg/100 g) and essential minerals. According to GC-MS results, tunichrome had different antioxidant and antimicrobial components. The TPC and TFC of tunichrome were 0.55 mg GA/g and 0.21 mg quercetin/100 g, respectively. Tunichrome showed higher antioxidant activity than ascorbic acid, and its radical scavenging activity values were increased from 30.28 to 82.08% by increasing concentration from 50 to 200 ppm. Inhibition zones of Staphylococcus aureus, Bacillus cereus, Salmonella enterica, and Escherichia coli O157:H7 were 14, 18, 17, and 15 mm, respectively. Moreover, the minimum inhibitory concentration values of tunichrome for S. aureus, Bacillus cereus, S. enterica, and E. coli O157:H7 were 1.17, 0.59, 0.59, and 1.17 mg/ml, respectively. The minimum bacterial concentrations were 2.34, 1.17, 1.17, and 2.34 mg/ml for S. aureus, Bacillus cereus, S. enterica, and E. coli O157:H7, respectively. These results showed that tunichrome of Phallusia nigra has excellent biological effects as a bioactive compound for food fortification

    Synthesis, Structural Studies, and α-Glucosidase Inhibitory, Antidiabetic, and Antioxidant Activities of 2,3-Dihydroquinazolin-4(1 H)-ones Derived from Pyrazol-4-carbaldehyde and Anilines

    A series of new quinazoline derivatives were designed and synthesized via a one-pot condensation reaction between isatoic anhydride and aromatic aldehydes with anilines using aluminum sulfate as a catalyst in refluxing ethanol. Their structures were confirmed by their physical, IR, 1H NMR, 13C NMR, and mass spectroscopy data and evaluated for some biological effects, including the antioxidant and α-glucosidase inhibitory activities as well as some in vivo hematological parameters. The ability of synthesized compounds in the inhibition of α-glucosidase was also investigated through the in silico study. The significant and important changes in some hematological tests were perceived. Notably, compound 4h showed more reducing effects on cholesterol and triglyceride levels. This molecule certainly has the potential to be developed as the antihyperlipemic compound. The tested compounds, in particular, compounds 4j and 4l, were found to be uniquely reducing blood sugar levels. The entire synthesized compounds showed the potent α-glucosidase inhibitory activity compared with acarbose as a standard material. Amongst, the compounds 4h and 4i showed the strongest enzyme inhibitory potentials than the standard drug acarbose. There was a good correlation between in vitro and in silico studies for ligands 4i and 4l. The majority of compounds presented a good radical scavenging activity, though the compound 4j exhibited the strongest activity, even to the standard of ascorbic acid. Further studies are required to determine whether these main compounds could be a potential treatment for diabetes and hyperlipidemia diseases

The thermal conductivity of a heat conducting body depends on the temperature distribution over the body. In this study, the simulated measured temperature distribution on part of the boundary is related to the variable heat flux imposed on a different part of the boundary through incorporating the variable thermal conductivity components into the sensitivity coefficients. To do so, a body-fitted grid generation technique is used to mesh the two-dimensional irregular body and solve the direct heat conduction problem. An efficient, accurate, robust, and easy to implement method is presented to compute the sensitivity coefficients through derived expressions. Novelty of the study is twofold: (1) Boundary-fitted grid-based sensitivity analysis in which all sensitivities can be obtained in only one direct solution (at each iteration), irrespective of the number of unknown parameters, and (2) the way the measured temperatures on part of boundary are related to a variable heat flux applied on another part of boundary through components of a variable thermal conductivity. The conjugate gradient method along with the discrepancy principle is used in the inverse analysis to minimize the objective function and achieve the desired solution

    Assessing Google Flu Trends Performance in the United States during the 2009 Influenza Virus A (H1N1) Pandemic

    Google Flu Trends (GFT) uses anonymized, aggregated internet search activity to provide near-real time estimates of influenza activity. GFT estimates have shown a strong correlation with official influenza surveillance data. The 2009 influenza virus A (H1N1) pandemic [pH1N1] provided the first opportunity to evaluate GFT during a non-seasonal influenza outbreak. In September 2009, an updated United States GFT model was developed using data from the beginning of pH1N1.We evaluated the accuracy of each U.S. GFT model by comparing weekly estimates of ILI (influenza-like illness) activity with the U.S. Outpatient Influenza-like Illness Surveillance Network (ILINet). For each GFT model we calculated the correlation and RMSE (root mean square error) between model estimates and ILINet for four time periods: pre-H1N1, Summer H1N1, Winter H1N1, and H1N1 overall (Mar 2009–Dec 2009). We also compared the number of queries, query volume, and types of queries (e.g., influenza symptoms, influenza complications) in each model. Both models' estimates were highly correlated with ILINet pre-H1N1 and over the entire surveillance period, although the original model underestimated the magnitude of ILI activity during pH1N1. The updated model was more correlated with ILINet than the original model during Summer H1N1 (r = 0.95 and 0.29, respectively). The updated model included more search query terms than the original model, with more queries directly related to influenza infection, whereas the original model contained more queries related to influenza complications.Internet search behavior changed during pH1N1, particularly in the categories “influenza complications” and “term for influenza.” The complications associated with pH1N1, the fact that pH1N1 began in the summer rather than winter, and changes in health-seeking behavior each may have played a part. Both GFT models performed well prior to and during pH1N1, although the updated model performed better during pH1N1, especially during the summer months

    A systematic review of gut microbiota composition in observational studies of major depressive disorder, bipolar disorder and schizophrenia

    The emerging understanding of gut microbiota as ‘metabolic machinery’ influencing many aspects of physiology has gained substantial attention in the field of psychiatry. This is largely due to the many overlapping pathophysiological mechanisms associated with both the potential functionality of the gut microbiota and the biological mechanisms thought to be underpinning mental disorders. In this systematic review, we synthesised the current literature investigating differences in gut microbiota composition in people with the major psychiatric disorders, major depressive disorder (MDD), bipolar disorder (BD) and schizophrenia (SZ), compared to ‘healthy’ controls. We also explored gut microbiota composition across disorders in an attempt to elucidate potential commonalities in the microbial signatures associated with these mental disorders. Following the PRISMA guidelines, databases were searched from inception through to December 2021. We identified 44 studies (including a total of 2510 psychiatric cases and 2407 controls) that met inclusion criteria, of which 24 investigated gut microbiota composition in MDD, seven investigated gut microbiota composition in BD, and 15 investigated gut microbiota composition in SZ. Our syntheses provide no strong evidence for a difference in the number or distribution (α-diversity) of bacteria in those with a mental disorder compared to controls. However, studies were relatively consistent in reporting differences in overall community composition (β-diversity) in people with and without mental disorders. Our syntheses also identified specific bacterial taxa commonly associated with mental disorders, including lower levels of bacterial genera that produce short-chain fatty acids (e.g. butyrate), higher levels of lactic acid-producing bacteria, and higher levels of bacteria associated with glutamate and GABA metabolism. We also observed substantial heterogeneity across studies with regards to methodologies and reporting. Further prospective and experimental research using new tools and robust guidelines hold promise for improving our understanding of the role of the gut microbiota in mental and brain health and the development of interventions based on modification of gut microbiota

    Evaluation of Embodied Carbon Emissions in UK Supermarket Constructions: A Study on Steel, Brick, and Timber Frameworks with Consideration of End-of-Life Processes

    Buildings and the construction sector as a whole are among the chief emitters of carbon, and the structural system of a building contributes substantially to its embodied carbon emissions. Whereas extensive studies exist into carbon missions, a detailed evaluation of real multipart building systems in brick, steel, and timber (glulam) substitutes is lacking. This paper employs whole-life-embedded carbon as a sustainability metric to compare a current UK supermarket building system of steel, brick, and timber. Four construction systems by the supermarket, referred to as CS1, CS2, CS3, and CS4, are used in the investigation. Comparisons are also made between two end-of-life treatment methods (recycle and landfill) along with the benefits that can be realised in future construction projects. The outcome from the comparative assessment reveals that there are minor variations in the embodied carbon of building systems used by the supermarket. CS4, while currently presenting marginal gains (approximately 148,960.68 kgCO2eq.) compared to CS1, loses its advantages when recycled contents for future construction projects are considered. The result indicates that CS4 generates about 18% less carbon emission reduction potential than CS1, whilst CS3 generates approximately 16% less than CS1. The findings of this article can enhance the knowledge of embodied carbon estimation and reduction capabilities of timber, steel, and brick buildings. Also, the detailed method for quantifying embodied carbon used in this article can be adopted in similar projects around the world

    Differences in the food consumption between kidney stone formers and non-formers in the Swiss Kidney Stone Cohort.

    OBJECTIVE Diet has a major influence on the formation and management of kidney stones. However, kidney stone formers' diet is difficult to capture in a large population. Our objective was to describe the dietary intake of kidney stone formers in Switzerland and to compare it to non-stone formers. METHODS We used data from the Swiss Kidney Stone Cohort (n=261), a multicentric cohort of recurrent or incident kidney stone formers with additional risk factors, and a control group of CT-scan proven non-stone formers (n=197). Dieticians conducted two consecutive 24-h dietary recalls, using structured interviews and validated software (GloboDiet). We took the mean consumption per participant of the two 24-h dietary recalls to describe the dietary intake and used two-part models to compare the two groups. RESULTS The dietary intake was overall similar between stone and non-stone formers. However, we identified that kidney stone formers had a higher probability of consuming cakes and biscuits (odds ratio, OR[95% CI] =1.56[1.03; 2.37]) and soft drinks (OR=1.66[1.08; 2.55]). Kidney stone formers had a lower probability of consuming nuts and seeds (OR =0.53[0.35; 0.82]), fresh cheese (OR=0.54[0.30; 0.96]), teas (OR=0.50[0.3; 0.84]), and alcoholic beverages (OR=0.35[0.23; 0.54]), especially wine (OR=0.42[0.27; 0.65]). Furthermore, among consumers, stone formers reported smaller quantities of vegetables (β coeff[95% CI]= - 0.23[- 0.41; - 0.06]), coffee (β coeff= - 0.21[- 0.37; - 0.05]), teas (β coeff= - 0.52[- 0.92; - 0.11]) and alcoholic beverages (β coeff= - 0.34[- 0.63; - 0.06]). CONCLUSION Stone formers reported lower intakes of vegetables, tea, coffee, and alcoholic beverages, more specifically wine, but reported drinking more frequently soft drinks than non-stone formers. For the other food groups, stone formers and non-formers reported similar dietary intakes. Further research is needed to better understand the links between diet and kidney stone formation and develop dietary recommendations adapted to the local settings and cultural habits
