    The Morphological Effects of Two Antimicrobial Peptides, Hecate-1 and Melittin, on Escherichia Coli

    The effects of the 26 amino acid, cationic, amphipathic, antibacterial peptide melittin and hecate-1, a 23 amino acid analog of it, on the gram negative bacterium Escherichia coli were investigated using scanning electron microscopy (SEM), transmission electron micros-copy (TEM), and freeze-fracture. Both peptides killed virtually all bacteria at the peptide concentration and cell density used. TEM and SEM revealed aggregates of bacteria entangled with material extruded from the bacterial surfaces. SEM revealed irregular bacterial surfaces with bleb-like projections. TEM and freeze-fracture indicate that the bacterial inner and outer membranes, as well as the peptidoglycan layer between, were extensively damaged. The cytoplasmic contents of the cells, however, did not appear radically disturbed, providing little evidence for osmotically induced cytolysis

    Individual differences in the shape of the nasal visual field

    AbstractBetween-subject differences in the shape of the nasal visual field were assessed for 103 volunteers 21–85years of age and free of visual disorder. Perimetry was conducted with a stimulus for which contrast sensitivity is minimally affected by peripheral defocus and decreased retinal illumination. One eye each was tested for 103 volunteers free of eye disease in a multi-center prospective longitudinal study. A peripheral deviation index was computed as the difference in log contrast sensitivity at outer (25–29° nasal) and inner (8° from fixation) locations. Values for this index ranged from 0.01 (outer sensitivity slightly greater than inner sensitivity) to −0.7 log unit (outer sensitivity much lower than inner sensitivity). Mean sensitivity for the inner locations was independent of the deviation index (R2<1%), while mean sensitivity for the outer locations was not (R2=38%, p<0.0005). Age was only modestly related to the index, with a decline by 0.017 log unit per decade (R2=10%). Test-retest data for 21 volunteers who completed 7–10 visits yielded standard deviations for the index from 0.04 to 0.17 log unit, with a mean of 0.09 log unit. Between-subject differences in peripheral deviation persisted over two years of longitudinal testing. Peripheral deviation indices were correlated with indices for three other perimetric stimuli used in a subset of 24 volunteers (R2 from 20% to 49%). Between-subject variability in shape of the visual field raises concerns about current clinical visual field indices, and further studies are needed to develop improved indices

Faced with a fact pattern that did not embody an apparent anticompetitive intent, a classic conspiracy between labor and non-labor entities, or activity clearly unrelated to the legitimate union interest in achieving better wages and working conditions, the Court was forced to abandon the clear showing test of Pennington, the intimately related test of Jewel Tea\u27 and, perhaps, even the Allen Bradley doctrine. ============================= William G. Scott By extending federal jurisdiction to encompass all robbery and extortion potentially affecting interstate commerce, the instant decision not only reflects, but substantially contributes to, the increasing federalization of intrastate crime under the commerce clause. In light of the decision\u27s broad rationale, the case may portend virtually unlimited expansion of federal jurisdiction into the field of crime control. At the very least, the decision constitutes authority for extending the jurisdictional range of other affecting commerce statutes to encompass all conduct potentially affecting commerce. It is difficult to conceive of any criminal activity, no matter how localized, that remains beyond the scope of the instant rationale. =========================== Mitchell M. Purvis The instant decision, relying on one side of conflicting precedent from other circuits, does little to reconcile the divergent answers to the issue raised by the Hayden caveat: what limits, if any, on searches and seizures should be developed to replace the discredited categorizations of the mere evidence rule. The Bennett decision,considering whether an item that possessed the requisite characteristics for protection under the privilege against self-incrimination consequently was proscribed as an object of a reasonable search and seizure, began a series of opinions obscuring the focus of this issue by failing to recognize that the amendments jointly protect overlapping substantive values through procedurally distinct mechanisms. =========================== George M. Kryder, III The instant court attempted to resolve the tension between these interests by permitting rejection of the entire agreement only after a substantial showing that continued operations would lead to collapse of the business. The court then would require a debtor-in-possession to bargain with the incumbent employees. A better approach would be to permit rejection of only those portions of the collective bargaining agreement that the court finds onerous and burdensome, while leaving in force the remaining portions of the agreement upon which the employees have relied. Such an analysis would afford employees greater protection than merely imposing an obligation to bargain, while simultaneously allowing the debtor-in-possession to renegotiate the burdensome provisions of the old agreement. ================================ Richard Michael Pitt The instant court recognized at the outset that the proper extraterritorial application of the securities laws was not to be found in the language of the acts.     Non-Invasive Prenatal Detection of Trisomy 21 Using Tandem Single Nucleotide Polymorphisms

    BACKGROUND: Screening tests for Trisomy 21 (T21), also known as Down syndrome, are routinely performed for the majority of pregnant women. However, current tests rely on either evaluating non-specific markers, which lead to false negative and false positive results, or on invasive tests, which while highly accurate, are expensive and carry a risk of fetal loss. We outline a novel, rapid, highly sensitive, and targeted approach to non-invasively detect fetal T21 using maternal plasma DNA. METHODS AND FINDINGS: Highly heterozygous tandem Single Nucleotide Polymorphism (SNP) sequences on chromosome 21 were analyzed using High-Fidelity PCR and Cycling Temperature Capillary Electrophoresis (CTCE). This approach was used to blindly analyze plasma DNA obtained from peripheral blood from 40 high risk pregnant women, in adherence to a Medical College of Wisconsin Institutional Review Board approved protocol. Tandem SNP sequences were informative when the mother was heterozygous and a third paternal haplotype was present, permitting a quantitative comparison between the maternally inherited haplotype and the paternally inherited haplotype to infer fetal chromosomal dosage by calculating a Haplotype Ratio (HR). 27 subjects were assessable; 13 subjects were not informative due to either low DNA yield or were not informative at the tandem SNP sequences examined. All results were confirmed by a procedure (amniocentesis/CVS) or at postnatal follow-up. Twenty subjects were identified as carrying a disomy 21 fetus (with two copies of chromosome 21) and seven subjects were identified as carrying a T21 fetus. The sensitivity and the specificity of the assay was 100% when HR values lying between 3/5 and 5/3 were used as a threshold for normal subjects. CONCLUSIONS: In summary, a targeted approach, based on calculation of Haplotype Ratios from tandem SNP sequences combined with a sensitive and quantitative DNA measurement technology can be used to accurately detect fetal T21 in maternal plasma when sufficient fetal DNA is present in maternal plasma

    Pre- and Post Impoundment Ichthyoparasite Succession in a New Arkansas Reservoir

    Helminth and crustacean parasites from 2,387 Micropterus dolomieui, M. punctulatus, and M. salmoides were utilized to monitor annual pre- and postimpoundment succession patterns spanning eight con- tinuous years in Beaver Reservoir, Arkansas. Incidence of infection by ichthyoparasites with direct life cycles (monogenetic trematodes, leeches, and crustaceans) generally increased following impoundment, although leeches remained relatively constant. Exceptions to this general pattern occurred. Incidence of ichthyoparasites with indirect life cycles (digenetic trematodes, cestodes, acanthocephalans, and nematodes) decreased immediately following impoundment with subsequent increases to a point equal or above that of preimpoundment, although exceptions occurred. Time for species adaptation to the reservoir environment varied, with some species disappearing and others occurring for the first time. Diversity indices indicated that a moderate parasite community was maintained in the White River two years prior to its impoundment to form Beaver Reservoir. During the first impoundment year the parasite community declined to the lowest postimpoundment level with the abrupt change in habitat. Throughout the following four post- impoundment years the parasite community gradually increased to become much larger and more complex than it was during preimpoundment. Parasite community succession stabilization occurred in the fifth postimpoundment year and continued the following year indicating the establishment of a climax ichthyoparasite community

    Statement of the Third International Exercise-Associated Hyponatremia Consensus Development Conference, Carlsbad, California, 2015

    The third International Exercise-Associated Hyponatremia (EAH) Consensus Development Conference convened in Carlsbad, California in February 2015 with a panel of 17 international experts. The delegates represented 4 countries and 9 medical and scientific sub-specialties pertaining to athletic training, exercise physiology, sports medicine, water/sodium metabolism, and body fluid homeostasis. The primary goal of the panel was to review the existing data on EAH and update the 2008 Consensus Statement.1 This document serves to replace the second International EAH Consensus Development Conference Statement and launch an educational campaign designed to address the morbidity and mortality associated with a preventable and treatable fluid imbalance. The following statement is a summary of the data synthesized by the 2015 EAH Consensus Panel and represents an evolution of the most current knowledge on EAH. This document will summarize the most current information on the prevalence, etiology, diagnosis, treatment and prevention of EAH for medical personnel, athletes, athletic trainers, and the greater public. The EAH Consensus Panel strove to clearly articulate what we agreed upon, did not agree upon, and did not know, including minority viewpoints that were supported by clinical experience and experimental data. Further updates will be necessary to both: (1) remain current with our understanding and (2) critically assess the effectiveness of our present recommendations. Suggestions for future research and educational strategies to reduce the incidence and prevalence of EAH are provided at the end of the document as well as areas of controversy that remain in this topic.
