Novel processes for extraction and fractionation of fatty acids from microbial cell mass
In this thesis a novel integrated process for extraction and fractionation of long chain polyunsaturated fatty acids (LC-PUFA), particularly the high-value docosahexaenoic acid (DHA), from wet microbial cell mass was investigated. The research was focused in two main aspects: (i) selection of enzymes for stepwise hydrolysis of triacylglycerols present the cell mass, and (ii) evaluation of a biphasic membrane contactor system, comprising an asymmetric membrane, for extraction of released fatty acids.
The kinetics of hydrolysis of triacylglycerols to diacylglycerols, monoacylglycerols and free fatty acids for specific and non-specific lipases was characterised. It was demonstrated that the enzymatic hydrolysis of triacylglycerols can be effectively performed directly in the cell mass suspension, i.e. without previous isolation of the lipid fraction. New analytical methods were developed for the analyses of acylglycerols and fatty acids by LC-ESI-MS. The access to the concentration profiles of tri-, di-, and monoacylglycerols throughout the reaction enabled a better evaluation of the performance of enzymes tested in terms of kinetics and selectivity. An effective selective hydrolysis can be attained by employing specific and non-specific lipases in the enzymatic hydrolysis of triacylglycerols. Based on the results obtained, optimum combinations of enzymes for process applications can be proposed.
Asymmetric polyimide membranes were prepared and tested for the separation of fatty acids in the biphasic membrane contactor system. It was found that the type of polymer, the membrane molecular weight cut-off, and the composition of the aqueous phase all had considerable effect on mass transfer across the membrane. Significantly higher mass transfer coefficients of long-chain fatty acids than previously reported were achieved. The solute transport mechanism through asymmetric polyimide porous membranes in the biphasic system was also explored. However, the membrane mass transfer coefficients calculated based on the solute transport model proposed, did not explain experimental observations well.
The feasibility of the proposed integrated process for sequential enzymatic hydrolysis of triacylglycerols and simultaneous separation of released fatty acids was demonstrated. As expected, the presence of an emulsion in the aqueous phase limited the mass transfer through the membrane. However, promising results were observed, and mass transfer rates may be improved by further optimisation of the operating conditions. It is then concluded that the objective of the present work was accomplished as a partial fractionation of the main fatty acids was obtained.Open Acces
Global behavior of the Ricci flow on generalized flag manifolds with two isotropy summands
AbstractIn this paper, we study the global behavior of the Ricci flow equation for two classes of homogeneous manifolds with two isotropy summands. Using methods of the qualitative theory of differential equations, we present the global phase portrait of such systems and derive some geometrical consequences on the structure of such manifolds under the action of the Ricci flow
The Ricci flow of left invariant metrics on full flag manifold SU(3)/T from a dynamical systems point of view
In this paper we study the behavior of the Ricci flow at infinity for the
full flag manifold using techniques of the qualitative theory of
differential equations, in special the Poincar\'e Compactification and Lyapunov
exponents. We prove that there are four invariant lines for the Ricci flow
equation, each one associated with a singularity corresponding to a Einstein
metric. In such manifold, the bi-invariant normal metric is Einstein. Moreover,
around each invariant line there is a cylinder of initial conditions such that
the limit metric under the Ricci flow is the corresponding Einstein metric; in
particular we obtain the convergence of left-invariant metrics to a

Rescuing emotional intelligence from the curse of fragmentation: Towards an integrative framework
A evolução das publicações sobre Inteligência Emocional (IE) mostra um interesse crescente
neste tópico, tanto na literatura científica, como popular. No entanto, a ausência de uma
abordagem consensual de conceptualização e medição traduz-se em três pressupostos que
prejudicam a construção do conhecimento. O primeiro é o de que as abordagens existentes
são mutuamente exclusivas; o segundo assenta na ideia de que é inevitável este tipo de
constructo ser demasiado inclusivo; e o último assume que a IE é redundante face a outras
variáveis na predição de resultados importantes para os indivíduos e as organizações. O
presente trabalho desafia estas suposições (a) propondo uma estrutura integrativa que inclui as
principais abordagens de estudo da IE numa estrutura multicamadas; (b) propondo e testando
empiricamente os componentes-chave que medem os aspectos essenciais da IE; e finalmente
(c) usando modelos não-lineares para testar empiricamente o valor acrescentado da IE
enquanto mediadora entre a personalidade e resultados relevantes, como o sucesso académico,
empenhamento no trabalho e comprometimento organizacional. Os resultados indicam que é
possível encontrar um modelo válido de componentes-chave para conceptualizar a IE de uma
forma parcimoniosa (estudos 1 e 2) e que, além de estar significativamente relacionada com a
saúde numa revisão meta-analítica (estudo 3), a IE prediz o sucesso académico (estudo 4), o
empenhamento no trabalho e o comprometimento organizacional (estudo 5) acima da
personalidade. Globalmente, o presente trabalho destaca a necessidade de construir
conhecimento convergente neste campo de pesquisa e explorar modelos não-lineares para
melhor compreender a sua natureza e dinâmica.The evolution of publications on Emotional Intelligence (EI) shows an increasing interest in
this topic, both in the popular and scientific literatures. However, the absence of a consensual
conceptualization and measurement approach translates into three assumptions that are
hampering knowledge building. The first is that the extant approaches are mutually exclusive;
the second is based on the idea that over inclusiveness is inevitable in this sort of constructs;
and the last assumes that EI is redundant with other variables in the prediction of important
outcomes for individuals and organizations. The present work challenges these issues by (a)
proposing an integrative framework that includes the main approaches to the study of EI in a
multi-layer structure; (b) proposing and empirically testing the core components that measure
the essential aspects of EI; and finally (c) using nonlinear models to empirically test the added
value of EI as a mediator between personality and relevant outcomes such as academic
success, work engagement and organizational commitment. Results indicate that it is possible
to find a valid core components model to conceptualize EI in a more parsimonious way
(studies 1 and 2); and that besides being significantly associated with health in a meta-analytic
review (study 3), EI predicts academic success (study 4), work engagement and
organizational commitment (study 5) over and above personality. Overall, the present work
highlights the need to build convergent knowledge in this research field and to explore
nonlinear models to better grasp its nature and dynamics
Rotulagem de géneros alimentícios pré-embalados numa empresa de restauração
Mestrado em Engenharia Alimentar - Processamento de Alimentos - Instituto Superior de Agronomia - ULA rotulagem de géneros alimentícios constitui o principal meio de comunicação entre
as empresas do setor alimentar e os consumidores. A rotulagem fornece informações
importantes sobre os géneros alimentícios, permitindo aos consumidores efetuarem escolhas
alimentares mais saudáveis e ajustadas às suas necessidades nutricionais, enquanto garante
a qualidade e a segurança alimentar.
O Regulamento (UE) nº 1169/2011 relativo à prestação de informação aos
consumidores sobre os géneros alimentícios constitui o mais recente diploma da União
Europeia. Este Regulamento surge da necessidade de fornecer uma rotulagem mais
informativa, garantindo um elevado nível de defesa do consumidor e assegurando o bom
funcionamento do mercado interno.
O principal objetivo deste trabalho consistiu em avaliar o grau de não conformidades
dos rótulos de géneros alimentícios refrigerados e congelados pré-embalados, face à
legislação em vigor, por grupo de produtos e por fornecedor, através do levantamento das
não conformidades detetadas. Esta análise foi precedida por uma compilação dos requisitos
legais dos rótulos deste tipo de produtos.
Foram assim analisados 311 rótulos de produtos de quatro setores distintos: produtos
de carne, produtos da pesca, pastelaria e produtos pré-confecionados. Verificou-se que
apenas 14% dos rótulos analisados estavam totalmente conformes e que em 86% dos rótulos
foram identificados diferentes tipos de não conformidades. Este elevado grau de não
conformidade encontrado nos rótulos dos fornecedores da empresa em estudo poderá estar
relacionado com a dificuldade de compreensão da legislação e com a dispersão dos requisitos
de rotulagem por diversos diplomas.
Na sequência desta avaliação, foi sugerida a disponibilização de rótulos modelo, aos
fornecedores, bem como de uma lista de verificação para uso na análise dos rótulos, à
empresa. Esta lista de verificação permitirá facilitar a deteção de não conformidades e a sua
comunicação aos fornecedores. As medidas aqui propostas pretendem prevenir a ocorrência
