    Seroprevalence of Toxoplasma gondii infection in household and stray cats in Lanzhou, northwest China

    <p>Abstract</p> <p>Background</p> <p><it>Toxoplasma gondii </it>is an important protozoan parasite infecting humans and almost all warm-blooded animals. As the only definitive host, cats play a crucial role in the transmission of <it>T. gondii </it>infection by shedding parasite oocysts in their feces. However, little information on <it>T. gondii </it>infection in cats was available in Lanzhou, northwest China. This study was performed to determine the seroprevalence of <it>T. gondii </it>infection in household and stray cats in Lanzhou, northwest China.</p> <p>Results</p> <p>A total of 221 (179 households and 42 strays) blood samples were collected from clinically healthy cats admitted to several pet hospitals located in Lanzhou City, between November 2010 and July 2011 for the serological detection of <it>T. gondii </it>infection. The majority (207) of these cats represented Chinese Lihua cats. 47 of 221 (21.3%) examined cats were seropositive for <it>T. gondii </it>infection using the modified agglutination test (MAT) at the cut-off of 1:25. The seroprevalence in household and stray cats was assessed to be 15.6% and 45.2%, respectively, and the difference was statistically significant (<it>P <</it>0.05). The seroprevalence ranged from 15.1% to 25.8% among different age groups, but the differences were not statistically significant (<it>P ></it>0.05). Studies showed that there was no relationship between seroprevalence and the gender (<it>P ></it>0.05).</p> <p>Conclusions</p> <p>The present survey indicated the high seroprevalence of <it>T. gondii </it>in cats in Lanzhou, northwest China, which poses a threat to animal and human health. Therefore, measures should be taken to control and prevent toxoplasmosis of cats in this area.</p

    Qi-Shen-Yi-Qi Dripping Pills for the Secondary Prevention of Myocardial Infarction: A Randomised Clinical Trial

    Background. Several types of drugs have been recommended for the secondary prevention of myocardial infarction (MI). However, these conventional strategies have several limitations, such as low adherence, high cost, and side effects during long time use. Novel approaches to this problem are still needed. This trial aimed to test the effectiveness and safety of Qi-Shen-Yi-Qi Dripping Pills (QSYQ), a multi-ingredient Chinese patent medicine, for the secondary prevention of MI. Methods and Findings. A total of 3505 eligible patients were randomly assigned to QSYQ group (1746 patients) or aspirin group (1759). Patients took their treatments for 12 months. The final follow-up visit took place 6 months after the end of the trial drugs. The 12-month and 18-month estimated incidences of the primary outcome were 2.98% and 3.67%, respectively, in the QSYQ group. The figures were 2.96% and 3.81% in the aspirin group. No significant difference was identified between the groups. Conclusions. This trial did not show significant difference of primary and secondary outcomes between aspirin and QSYQ in patients who have had an MI. Though inconclusive, the result suggests that QSYQ has similar effects to aspirin in the secondary prevention of MI

    Acceleration and radiation of externally injected electrons in laser plasma wakefield driven by a Laguerre-Gaussian pulse

    By using three-dimensional particle-in-cell simulations, externally injected electron beam acceleration and radiation in donut-like wake fields driven by a LaguerreGaussian pulse are investigated. Studies show that in the acceleration process the total charge and azimuthal momenta of electrons can be stably maintained at a distance of a few hundreds of micrometers. Electrons experience low-frequency spiral rotation and high-frequency betatron oscillation, which leads to a synchrotron-like radiation. The radiation spectrum is mainly determined by the betatron motion of electrons. The far field distribution of radiation intensity shows axial symmetry due to the uniform transverse injection and spiral rotation of electrons. Our studies suggest a new way to simultaneously generate hollow electron beam and radiation source from a compact laser plasma accelerator

    Seroprevalence of Toxoplasma gondii infection in pet dogs in Lanzhou, Northwest China

    <p>Abstract</p> <p>Background</p> <p>In recent years, surveys of <it>Toxoplasma gondii </it>infection in dogs have been reported worldwide, including China. However, little is known about the prevalence of <it>T. gondii </it>in pet dogs in Northwest China. In the present study, the prevalence of <it>T. gondii </it>in pet dogs in Lanzhou, China was investigated using the modified agglutination test (MAT).</p> <p>Results</p> <p>In this survey, antibodies to <it>T. gondii </it>were found in 28 of 259 (10.81%) pet dogs, with MAT titers of 1:20 in 14 dogs, 1:40 in nine, 1:80 in four, and 1:160 or higher in one dog. The prevalence ranged from 6.67% to 16.67% among dogs of different ages, with low rates in young pet dogs, and high rates in older pet dogs. The seroprevalence in dogs >3 years old was higher than that in dogs ≤1 years old, but the difference was not statistically significant (<it>P ></it>0.05). The seroprevalence in male dogs was 12.50% (17 of 136), and in female dogs it was 8.94% (11 of 123), but the difference was not statistically significant (<it>P ></it>0.05).</p> <p>Conclusions</p> <p>A high prevalence of <it>T. gondii </it>infection was found in pet dogs in Lanzhou, Northwest China, which has implications for public health in this region. In order to reduce the risk of exposure to <it>T. gondii</it>, further measures and essential control strategies should be carried out rationally in this region.</p

    Ultraviolet C Irradiation Induces Different Expression of Cyclooxygenase 2 in NIH 3T3 Cells and A431 Cells: The Roles of COX-2 Are Different in Various Cell Lines

    Ultraviolet C (UVC) is a DNA damage inducer, and 20 J/m2 of UVC irradiation caused cell growth inhibition and induced cell death after exposure for 24–36 h. The growth of NIH 3T3 cells was significantly suppressed at 24 h after UVC irradiation whereas the proliferation of A431 cells was inhibited until 36 h after UVC irradiation. UVC irradiation increased COX-2 expression and such up-regulation reached a maximum during 3–6 h in NIH 3T3 cells. In contrast, UVC-induced COX-2 reached a maximum after 24–36 h in A431 cells. Measuring prostaglandin E2 (PGE2) level showed a biphasic profile that PGE2 release was rapidly elevated in 1–12 h after UVC irradiation and increased again at 24 h in both cell lines. Treatment with the selective COX-2 inhibitor, SC-791, during maximum expression of COX-2 induction, attenuated the UVC induced-growth inhibition in NIH 3T3 cells. In contrast, SC-791 treatment after UVC irradiation enhanced death of A431 cells. These data showed that the patterns of UVC-induced PGE2 secretion from NIH 3T3 cells and A431 cells were similar despite the differential profile in UVC-induced COX-2 up-regulation. Besides, COX-2 might play different roles in cellular response to UVC irradiation in various cell lines

    Role of Te on selective oxidation of propane to acrolein over MoBiTeO/SiO2 catalysts

    A series of MoBiTeO/SiO2 catalysts with different Te contents was prepared by impregnation method. Effect of Te on the catalysts'structure, acidity and catalytic performance for selective oxidation of propane to acrolem was investigated by XRD, Raman, XPS, NH3-TPD, FIF-IR, and performance evaluation techniques. The results indicated that by forming Mo-O-Te bond with the Mo species, tellurium component in the catalyst improved the dispersion of Mo oxide species that would be favorable to the formation of isolate active sites on the catalysts. Besides, Te could be the centers for alpha-H abstraction of propene and O-insertion to allyl species. So, the addition of Te to MoBiO/SiO2 catalyst promoted the acrolem selectivity greatly. However, the addition of tellurium also could contribute the formation of B acid sites in MoBiTeO/SiO2 catalysts that had been proved to be harmful to the generation of acrolein. Therefore, there existed an optimal scale for the amount of Te-content in the MoBiTeO/SiO, catalysts. When the n(Te)/n(Mb) ratio was 0.05-0.1, the catalyst showed the best catalytic performance

    The Ratio of Plasma and Urinary 8-oxo-Gsn Could Be a Novel Evaluation Index for Patients with Chronic Kidney Disease

    Nucleic acid oxidation plays an important role in the pathophysiology progress of a variety of diseases. 8-oxo-7,8-dihydro-2′-deoxyguanosine (8-oxo-dGsn) and 8-oxo-7,8-dihydroguanosine (8-oxo-Gsn), which originate from DNA and RNA oxidation, were the most widely used indicators for oxidative stress. The study investigated the relation between 8-oxo-dGsn, 8-oxo-Gsn, and CKD. 146 patients with CKD were divided into five disease stages, and their fasting blood and morning urine were collected. The levels of 8-oxo-dGsn and 8-oxo-Gsn in plasma and urine were quantified by LC-MS/MS. The ratio of urinary 8-oxo-Gsn to creatinine increased from stages 1 to 4 corresponding to the increased severity of CKD, but it decreased in stage 5. And plasma 8-oxo-Gsn gradually increased with the decline of renal function. In particular, the increased ratio of plasma and urine 8-oxo-Gsn in stage 5 exceeded the concentration of creatinine. This trend was similar to the estimated glomerular filtration rate (eGFR), which indicates that 8-oxo-Gsn could be an appropriate indicator for renal function. Our finding indicates that as the disease progresses, RNA oxidation is increased. The significant increase in the ratio of plasma and urinary 8-oxo-Gsn is a novel evaluation index of end-stage renal disease

    Antimetastatic Effects of Norcantharidin on Hepatocellular Carcinoma by Transcriptional Inhibition of MMP-9 through Modulation of NF-kB Activity

    The rate of morbidity and mortality of hepatocellular carcinoma (HCC) in Taiwan has not lessened because of difficulty in treating tumor metastasis. Norcantharidin (NCTD) is currently used as an anticancer drug for hepatoma, breast cancer, and colorectal adenocarcinoma. NCTD possesses various biological anticancer activities, including apoptosis. However, detailed effects and molecular mechanisms of NCTD on metastasis are unclear. Thus, HCC cells were subjected to treatment with NCTD and then analyzed to determine the effects of NCTD on cell metastasis.Modified Boyden chamber assays revealed that NCTD treatment inhibited cell migration and invasion capacities of HCC cells substantially. Results of zymography and western blotting showed that activities and protein levels of matrix metalloproteinase-9 (MMP-9) and urokinase plasminogen activator (u-PA) were inhibited by NCTD. Western blot analysis showed that NCTD inhibits phosphorylation of ERK1/2. Testing of mRNA level, quantitative real-time PCR, and promoter assays evaluated the inhibitory effects of NCTD on MMP-9 and u-PA expression in HCC cells. The chromatin immunoprecipitation (ChIP) assay for analyzing the genomic DNA sequences bound to these proteins was reactive to the transcription protein nuclear factor (NF)-kappaB, which was inhibited by NCTD. The expression of NF-kappa B was measured by western blot analysis, which revealed decreased nuclear-factor DNA-binding activity after NCTD treatment.NCTD inhibited MMP-9 and u-PA expression through the phosphorylation of ERK1/2 and NF-kappaB signaling pathway which serves as a powerful chemopreventive agent in HCC cell metastasis
