Stability of human plasma lipidome depending on the storage conditions and sample manipulation
Diplomová práca je zameraná na štúdium stability vybraných tried lipidov v ľudskej plazme spojením ultravysokoúčinnej superkritickej fluidnej chromatografie a hmotnostnej spektrometrie. V teoretickej časti je priblížená štruktúra, rozdelenie a funkcie lipidov. Popísaný je odber biologického materiálu, v tomto prípade najmä ľudskej plazmy a jeho následné spracovanie pre lipidomickú analýzu. Charakterizované sú tiež separačné techniky využívané pre analýzu lipidov rovnako ako techniky hmotnostnej spektrometrie. Cieľom práce bolo stanovenie koncentračných zmien vybraných tried lipidov vo vzorkách zmesi interných štandardov a plazmy pri rôznych skladovacích podmienkach a počte rozmrazovacích cyklov.The diploma thesis is focused on the stability study of chosen lipid classes of human plasma using ultra-high performance supercritical fluid chromatography coupled to the mass spectrometry. In the theoretical part, the structure, classification and functions of lipids are elucidated. Biological material sampling, in this case primarily human plasma and its treatment for lipidomic analysis is desribed. Characterised are also separation techniques and techniques of mass spectrometry used for lipid analysis. Collection, processing and analysis of samples of hair and nails
V úvodnej časti tejto bakalárskej práce je popísaná anatómia kože a jej derivátov s cestami inkorporácie substancií do vlasov a nechtov. Práca je zameraná na ich analýzu za účelom stanovenia látok pochádzajúcich zo životného prostredia, drog a liečiv. Popísané sú kroky odberu vzoriek, ich dekontaminácie a jednotlivých preanalytických postupov. Pozornosť je venovaná tiež segmentálnej analýze odhaľujúcej čas požitia sledovaných substancií.In the initial part of this bachelor thesis is described anatomy of skin and its derivatives with ways of incorporation of substances to hair and nails. Thesis is aimed at their analysis for the purpose of determinating substances originating in environment, drugs and pharmaceuticals. Steps of sample collection, decontamination and analytical pretreatment are described. An upwind-based scheme for solving the oblique derivative boundary-value problem related to physical geodesy
The paper presents a novel original upwindbased
approach for solving the oblique derivative boundary
value problem by the finite volume method. In this approach,
the oblique derivative boundary condition is interpreted
as a stationary advection equation for the unknown
disturbing potential. Its approximation is then performed
by using the first order upwind scheme taking into account
information from inflow parts of the finite volume boundary
only. When the numerical scheme is derived, numerical
simulations in 2D and 3D domains are performed and
the experimental order of convergence of the proposed algorithm
is studied. Moreover a comparison with a solution
by the central scheme previously used for this kind
of problem is performed. Finally we present numerical experiments
dealing with the global and local gravity field
