    A Review of Common Medicinal Plants in Chin State, Myanmar:

    Promising sources of novel bioactive compounds include plants growing in several third-world countries where the local flora is still largely uninvestigated. A paradigmatic example is represented by medicinal plants growing in Myanmar, especially in Chin State, in northwestern Myanmar. This is one of the least developed areas of the country where the people still use natural remedies derived from a rich biodiversity. This review mainly covers the investigations done on phytochemical constituents and biological activities of 20 medicinal plants, namely Alangium chinense, Anemone obtusiloba, Anneslea fragrans, Antidesma bunius, Croton oblongifolius, Embelia tsjeriam-cottam, Ficus heterophylla, Gaultheria fragrantissima, Hydnocarpus kurzii, Leea macrophylla, Leucas cephalotes, Millingtonia hortensis, Myrica nagi, Olax scandens, Pimpinella heyneana, Pterospermum semisagittatum, Ruellia tuberosa, Smilax zeylanica, Stemona burkillii, and Tadehagi triquetrum, that have long been used in the Chin State for curing various diseases. These plants have been selected on the basis of their medicinal uses not only in Myanmar but also in the related Ayurvedic healing system. Moreover, besides their medicinal importance, most of them grow in the Chin State more abundantly than in other regions of Myanmar. Although the efficacy of some of these plants have been verified scientifically, the chemical constituents and biological activities of most of them still need to be investigated to confirm the claimed therapeutic effects

    Highly Cytotoxic Xanthones from Cratoxylum cochinchinense Collected in Myanmar

    Eight xanthones and one anthraquinone, together with four common triterpenoids, have been isolated from the barks of Cratoxylum cochinchinense, collected in Myanmar. The structures of the metabolites were elucidated by spectroscopic data analysis and their antiproliferative activities were measured against six human tumor cell lines, by using the MTT assay. Pruniflorone N (1) showed a significant cytotoxicity against all cancer cells with IC50 values in the range 3-9 μM, on average higher than the anticancer drug cisplatin. Instead, compounds 2 and 3 exhibited high antiproliferative activity against some specific cell lines

    Mitochondrial pseudogenes in the nuclear genome of Aedes aegypti mosquitoes: implications for past and future population genetic studies

    <p>Abstract</p> <p>Background</p> <p>Mitochondrial DNA (mtDNA) is widely used in population genetic and phylogenetic studies in animals. However, such studies can generate misleading results if the species concerned contain nuclear copies of mtDNA (Numts) as these may amplify in addition to, or even instead of, the authentic target mtDNA. The aim of this study was to determine if Numts are present in <it>Aedes aegypti </it>mosquitoes, to characterise any Numts detected, and to assess the utility of using mtDNA for population genetics studies in this species.</p> <p>Results</p> <p>BLAST searches revealed large numbers of Numts in the <it>Ae. aegypti </it>nuclear genome on 146 supercontigs. Although the majority are short (80% < 300 bp), some Numts are almost full length mtDNA copies. These long Numts are not due to misassembly of the nuclear genome sequence as the Numt-nuclear genome junctions could be recovered by amplification and sequencing. Numt evolution appears to be a complex process in <it>Ae. aegypti </it>with ongoing genomic integration, fragmentation and mutation and the secondary movement of Numts within the nuclear genome.</p> <p>The PCR amplification of the putative mtDNA nicotinamide adenine dinucleotide dehydrogenase subunit 4 (<it>ND4</it>) gene from 166 Southeast Asian <it>Ae. aegypti </it>mosquitoes generated a network with two highly divergent lineages (clade 1 and clade 2). Approximately 15% of the <it>ND4 </it>sequences were a composite of those from each clade indicating Numt amplification in addition to, or instead of, mtDNA. Clade 1 was shown to be composed at least partially of Numts by the removal of clade 1-specific bases from composite sequences following enrichment of the mtDNA. It is possible that all the clade 1 sequences in the network were Numts since the clade 2 sequences correspond to the known mitochondrial genome sequence and since all the individuals that produced clade 1 sequences were also found to contain clade 2 mtDNA-like sequences using clade 2-specific primers. However, either or both sets of clade sequences could have Numts since the BLAST searches revealed two long Numts that match clade 2 and one long Numt that matches clade 1. The substantial numbers of mutations in cloned <it>ND4 </it>PCR products also suggest there are both recently-derived clade 1 and clade 2 Numt sequences.</p> <p>Conclusion</p> <p>We conclude that Numts are prevalent in <it>Ae. aegypti </it>and that it is difficult to distinguish mtDNA sequences due to the presence of recently formed Numts. Given this, future population genetic or phylogenetic studies in <it>Ae. aegypti </it>should use nuclear, rather than mtDNA, markers.</p

    On the role of horizontal resolution over the Tibetan Plateau in the REMO regional climate model

    A number of studies have shown that added value is obtained by increasing the horizontal resolution of a regional climate model to capture additional fine-scale weather processes. However, the mechanisms leading to this added value are different over areas with complicated orographic features, such as the Tibetan Plateau (TP). To determine the role that horizontal resolution plays over the TP, a detailed comparison was made between the results from the REMO regional climate model at resolutions of 25 and 50 km for the period 1980–2007. The model was driven at the lateral boundaries by the European Centre for Medium-Range Weather Forecasts Interim Reanalysis data. The experiments differ only in representation of topography, all other land parameters (e.g., vegetation characteristics, soil texture) are the same. The results show that the high-resolution topography affects the regional air circulation near the ground surface around the edge of the TP, which leads to a redistribution of the transport of atmospheric water vapor, especially over the Brahmaputra and Irrawaddy valleys—the main water vapor paths for the southern TP—increasing the amount of atmospheric water vapor transported onto the TP by about 5. This, in turn, significantly decreases the temperature at 2 m by > 1.5 °C in winter in the high-resolution simulation of the southern TP. The impact of topography on the 2 m temperature over the TP is therefore by influencing the transport of atmospheric water vapor in the main water vapor paths. © 2018 Springer-Verlag GmbH Germany, part of Springer Natur

    The Assessment of Surface Air Temperature and Precipitation Simulated by Regional Climate Model REMO over China

    The ability of German regional climate Model (REMO) to simulate the near surface air temperature and total precipitation over China in 1989-2008 were assessed with the use of Taylor diagrams and bias analysis. Comparing the simulated near surface air temperature with a 20-year observational dataset from China, the spatial correlation coefficient was relatively high (0.94). However, the spatial correlation coefficient for total precipitation was relatively low (about 0.42). The near surface air temperature simulated by REMO was higher than the observed values in most part of China, showing a bias range within 4 C. Significant cold bias of about -4 to -2 C occurred over most of the Qinghai-Tibetan Plateau. In terms of total precipitation, the simulated values were higher than the observed ones, with biases evenly distributed. The annual mean bias in most part of China was within 300 mm. Except for the Qinghai-Tibetan Plateau, South China and Southwest China, REMO accurately reflected the distribution of near surface air temperature and total precipitation. REMO represented the temperature and total precipitation well in North China and Northeast China. REMO simulations were quite close to observations for near surface air temperature in summer and total precipitation in winter. REMO still needs to be improved in complex terrain areas

    Co-occurrence of Point Mutations in the Voltage-Gated Sodium Channel of Pyrethroid-Resistant Aedes aegypti Populations in Myanmar

    Background:Single amino acid substitutions in the voltage-gated sodium channel associated with pyrethroid resistance constitute one of the main causative factors of knockdown resistance in insects. The kdr gene has been observed in several mosquito species; however, point mutations in the para gene of Aedes aegypti populations in Myanmar have not been fully characterized. The aim of the present study was to determine the types and frequencies of mutations in the para gene of Aedes aegypti collected from used tires in Yangon City, Myanmar.Methodology/Principal Findings:We determined high pyrethroid resistance in Aedes aegypti larvae at all collection sites in Yangon City, by using a simplified knockdown bioassay. We showed that V1016G and S989P mutations were widely distributed, with high frequencies (84.4% and 78.8%, respectively). By contrast, we were unable to detect I1011M (or I1011V) or L1014F mutations. F1534C mutations were also widely distributed, but with a lower frequency than the V1016G mutation (21.2%). High percentage of co-occurrence of the homozygous V1016G/S989P mutations was detected (65.7%). Additionally, co-occurrence of homozygous V1016G/F1534C mutations (2.9%) and homozygous V1016G/F1534C/S989P mutations (0.98%) were detected in the present study.Conclusions/Significance:Pyrethroid insecticides were first used for malaria control in 1992, and have since been constantly used in Myanmar. This intensive use may explain the strong selection pressure toward Aedes aegypti, because this mosquito is generally a domestic and endophagic species with a preference for indoor breeding. Extensive use of DDT for malaria control before the use of this chemical was banned may also explain the development of pyrethroid resistance in Aedes aegypti

    Breast cancer management pathways during the COVID-19 pandemic: outcomes from the UK ‘Alert Level 4’ phase of the B-MaP-C study

    Abstract: Background: The B-MaP-C study aimed to determine alterations to breast cancer (BC) management during the peak transmission period of the UK COVID-19 pandemic and the potential impact of these treatment decisions. Methods: This was a national cohort study of patients with early BC undergoing multidisciplinary team (MDT)-guided treatment recommendations during the pandemic, designated ‘standard’ or ‘COVID-altered’, in the preoperative, operative and post-operative setting. Findings: Of 3776 patients (from 64 UK units) in the study, 2246 (59%) had ‘COVID-altered’ management. ‘Bridging’ endocrine therapy was used (n = 951) where theatre capacity was reduced. There was increasing access to COVID-19 low-risk theatres during the study period (59%). In line with national guidance, immediate breast reconstruction was avoided (n = 299). Where adjuvant chemotherapy was omitted (n = 81), the median benefit was only 3% (IQR 2–9%) using ‘NHS Predict’. There was the rapid adoption of new evidence-based hypofractionated radiotherapy (n = 781, from 46 units). Only 14 patients (1%) tested positive for SARS-CoV-2 during their treatment journey. Conclusions: The majority of ‘COVID-altered’ management decisions were largely in line with pre-COVID evidence-based guidelines, implying that breast cancer survival outcomes are unlikely to be negatively impacted by the pandemic. However, in this study, the potential impact of delays to BC presentation or diagnosis remains unknown
