    Enhanced production of tropane alkaloids in transgenic Scopolia parviflora hairy root cultures over-expressing putrescine N-methyl transferase (PMT) and hyoscyamine-6β-hydroxylase (H6H)

    Scopolia parviflora adventitious roots were metabolically engineered by co-expression of the two gene putrescine N-methyl transferase (PMT) and hyoscyamine-6β-hydroxylase (H6H) cDNAs with the aid of Agrobacterium rhizogenes. The transformed roots developed into morphologically distinct S. parviflora PMT1 (SpPMT1), S. parviflora PMT1 (SpPMT2), and S. parviflora H6H (SpH6H) transgenic hairy root lines. Consequent to the introduction of these key enzyme genes, the production of the alkaloids hyoscyamine and scopolamine was enhanced. Among the transgenic hairy root lines, SpPMT2 line possessed the highest growth index. The treatment of transgenic hairy roots with growth regulators further enhanced the production of scopolamine. Thus, the results suggest that PMT1, PMT2, and H6H genes may not only be involved in the metabolic regulation of alkaloid production but also that these genes may play a role in the root development

    Synthesis and pinning properties of the infinite-layer superconductor Sr0.9La0.1CuO

    We report the high-pressure synthesis of the electron-doped infinite-layer superconductor Sr0.9La0.1CuO2 and its superconducting properties. A Rietveld analysis of X-ray powder diffraction data showed that, within the resolution of the measurement, the sample had purely an infinite-layer structure without any discernible impurities. The superconducting volume fraction and the transition width were greatly improved compared to those in previous reports. The irreversibility field line and the intragranular critical current density were much higher than those of La1.85Sr0.15CuO4 and Nd1.85Ce0.15CuO4. The stronger pinning behaviors are consistent with the strong interlayer coupling due to the short distance between CuO2 planes.Comment: Physica C (in press) 5 pages, 4 figur

    Selection of high berberine yielding phellodendron insulare nak. lines and the antimicrobial activity of their extracts

    High berberine yielding Phellodendron insulare Nak. lines were selected by aggregate cloning method and the antimicrobial activity of their extracts was assessed. The berberine producing cork tree lines were selected by adopting a colorimetric method. In all 300 high berberine producing lines were selected with a colorimetric reagent containing 5M HCl and H2O2 and established from dissociated cell aggregates. The crude extracts from these lines showed antibacterial activities against tested Escherichia coli, Staphylococcus aureus, Salmonella typhimulium, and Listeria monocytogenes. The cork tree extracts were found to be inhibitory to these test organisms. Further the antimicrobial activity of plant extracts was on par with the berberine isolated from the extracts from native cork trees. These results have potential for developing alternative plant products as antimicrobial substances for application in agriculture and food industry

    Electronic Structure and Optical Properties of the Co-doped Anatase TiO2_{2} Studied from First Principles

    The Co-doped anatase TiO2_{2}, a recently discovered room-temperature ferromagnetic insulator, has been studied by the first-principles calculations in the pseudo-potential plane-wave formalism within the local-spin-density approximation (LSDA), supplemented by the full-potential linear augmented plane wave (FP-LAPW) method. Emphasis is placed on the dependence of its electronic structures and linear optical properties on the Co-doping concentration and oxygen vacancy in the system in order to pursue the origin of its ferromagnetism. In the case of substitutional doping of Co for Ti, our calculated results are well consistent with the experimental data, showing that Co is in its low spin state. Also, it is shown that the oxygen vacancy enhances the ferromagnetism and has larger effect on both the electronic structure and optical properties than the Co-doping concentration only.Comment: 12 pages, 4 figure

    Immune-Related Gene Expression in Two B-Complex Disparate Genetically Inbred Fayoumi Chicken Lines Following Eimeria maxima Infection

    To investigate the influence of genetic differences in the MHC on susceptibility to avian coccidiosis, M5.1 and M15.2 B-haplotype-disparate Fayoumi chickens were orally infected with live Eimeria maxima oocysts, and BW gain, fecal oocyst production, and expression of 14 immune-related genes were determined as parameters of protective immunity. Weight loss was reduced and fecal parasite numbers were lower in birds of the M5.1 line compared with M15.2 line birds. Intestinal intraepithelial lymphocytes from M5.1 chickens expressed greater levels of transcripts encoding interferon-γ (IFN-γ), interleukin-1β (IL-1β), IL-6, IL-8, IL-12, IL-15, IL-17A, inducible nitric oxide synthase, and lipopolysaccharide-induced tumor necrosis factor-α factor and lower levels of mRNA for IFN-α, IL-10, IL-17D, NK-lysin, and tumor necrosis factor superfamily 15 compared with the M15.2 line. In the spleen, E. maxima infection was associated with greater expression levels of IFN-γ, IL-15, and IL-8 and lower levels of IL-6, IL-17D, and IL-12 in M5.1 vs. M15.2 birds. These results suggest that genetic determinants within the chicken MHC influence resistance to E. maxima infection by controlling the local and systemic expression of immune-related cytokine and chemokine genes

    Search for sterile neutrino oscillation using RENO and NEOS data

    We present a reactor model independent search for sterile neutrino oscillation using 2\,509\,days of RENO near detector data and 180 days of NEOS data. The reactor related systematic uncertainties are significantly suppressed as both detectors are located at the same reactor complex of Hanbit Nuclear Power Plant. The search is performed by electron antineutrino\,(νe\overline{\nu}_e) disappearance between six reactors and two detectors with baselines of 294\,m\,(RENO) and 24\,m\,(NEOS). A spectral comparison of the NEOS prompt-energy spectrum with a no-oscillation prediction from the RENO measurement can explore reactor νe\overline{\nu}_e oscillations to sterile neutrino. Based on the comparison, we obtain a 95\% C.L. excluded region of 0.1<Δm412<70.1<|\Delta m_{41}^2|<7\,eV2^2. We also obtain a 68\% C.L. allowed region with the best fit of Δm412=2.41±0.03|\Delta m_{41}^2|=2.41\,\pm\,0.03\,\,eV2^2 and sin22θ14\sin^2 2\theta_{14}=0.08±\,\pm\,0.03 with a p-value of 8.2\%. Comparisons of obtained reactor antineutrino spectra at reactor sources are made among RENO, NEOS, and Daya Bay to find a possible spectral variation.Comment: 6 pages, 5 figures: This manuscript has been significantly revised by the joint reanalysis by RENO and NEOS Collaborations.
