An improved fungal mounting technique for Nomarski microscopy
The 'sellotape technique' of Butler and Mann (1959)
with lactophenol cotton blue stain is widely used in
these laboratories for the production of non-permanent
fungal mounts for class examination using bright field microscopy. The technique allows very little disturbance of fungal structure for identification and taxonomic work. The major drawbacks of this technique for permanent mounts are the fact that the tape itself is not inert and the adhesive reacts with the lactophenol forming droplets which cover the field of view (Onions et al., l98l). The stressed nature of the tape also gives rise to problems due to interference patterns during Nomarski microscopy. The increasing use of diflerential interference contrast(Nomarski) microscopy in mycological studies prompted a re-evaluation of techniques used in the preparation of fungal mounts. Other types of nonstressed tapes were found but these proved not to be sticky enough to pick up fungal structures from agar media cleanly and without undue damage. The technique described below uses a silicone based adhesive in conjunction with the slightly modified polyvinyl
alcohol mountant of Salmon (1954), updated by Omar et al. (1918), and represents a modification and improvement of the technique described by Bretz and Berry (1964)
Psychiatric Symptomatology among Individuals in Alcohol Detoxification Treatment
The coexistence of psychiatric symptomatology among individuals receiving longer-term treatment for alcohol use disorders has been well-established; however, less is known about comorbidity among individuals receiving alcohol detoxification. Using the Brief Symptom Inventory (BSI; Derogatis, 1992), we compared psychiatric symptomatology among 815 individuals receiving short-term detoxification services with normative data from nonpatients, psychiatric patients, and out-of-treatment individuals using street drugs. Findings revealed that individuals in the current sample reported a wide range of psychiatric symptoms with over 80% meeting BSI criteria for diagnosable mental illness. These BSI scores were significantly more severe than those reported by out-of-treatment individuals using street drugs and most closely resembled BSI scores reported for adult psychiatric inpatients. Findings suggest that routine screening for severe mental health symptoms appears warranted in detoxification units. Such screening would greatly increase the chance that coexistence of substance use and other psychiatric disorders would be properly addressed in ongoing treatment
The effects of inspiratory muscle training in older adults
Purpose: Declining inspiratory muscle function and structure and systemic low-level inflammation and oxidative stress may contribute to morbidity and mortality during normal ageing. Therefore, we examined the effects of inspiratory muscle training (IMT) in older adults on inspiratory muscle function and structure and systemic inflammation and oxidative stress, and re-examined the reported positive effects of IMT on respiratory muscle strength, inspiratory muscle endurance, spirometry, exercise performance, physical activity levels (PAL) and quality of life (QoL). Methods: Thirty-four healthy older adults (68 ± 3 years) with normal spirometry, respiratory muscle strength and physical fitness were divided equally into a pressure-threshold IMT or sham-hypoxic placebo group. Before and after an 8 week intervention, measurements were taken for dynamic inspiratory muscle function and inspiratory muscle endurance using a weighted plunger pressure-threshold loading device, diaphragm thickness using B-mode ultrasonography, plasma cytokine concentrations using immunoassays, DNA damage levels in peripheral blood mononuclear cells (PBMC) using Comet Assays, spirometry, maximal mouth pressures, exercise performance using a six minute walk test, PAL using a questionnaire and accelerometry, and QoL using a questionnaire
Phytophthora root and stem rot of soybean
Phytophthora sojae is a soil borne pathogen that in the past has caused very large economic losses. During the late 1970s, 300,000 soybean acres (approximately 10% of total soybean production in Ohio) were lost due to P. sojae. This disease has since been effectively managed predominately through the incorporation of single-gene mediated resistance but quantitative or partial resistance has been used as well. In fact, today, we can repeat 100% loss by planting soybean cultivars that were popular during earlier epidemics. Without high levels of resistance to this pathogen, many soybean acres would be lost each year to this disease. Phytophthora doesn’t forget and it doesn’t go away

No Negative Priming Effect in the Manual Stroop Task.
The negative priming effect is an increase in interference when the response to the target on the current trial corresponds to the response to the distractor word on a preceding trial. Contrary to the commonly held belief that the negative priming effect is ubiquitous in the Stroop task, in the original study by Neill (1977), negative priming was found only in the oral, and not the manual Stroop task. The present paper makes three empirical observations. First, we replicate the discrepancy in the finding of the negative priming effect in the oral versus manual Stroop tasks tested under identical conditions, where response mode could be the only the causal factor. Second, we point out that previous manual Stroop experiments reporting the negative priming effect confounded the effect of response repetition. Third, we report the analysis of the negative priming effect at the level of whole RT distribution, which revealed that the effect was absent throughout the RT distribution in the manual task, and it was of constant size across the RT distribution in the oral task. Creaky knees: Is there a reason for concern? A qualitative study of the perspectives of people with knee crepitus
Objective: Crepitus is a feature of osteoarthritis that may affect one's participation in exercise. An informed understanding is required of the perceptions that people have of their knee crepitus and how it affects their exercise behaviours. This study aims to investigate the role that crepitus may play in beliefs about exercise and knee health. Methods: Focus group and individual interviews were conducted online with participants who had knee crepitus. The transcripts were thematically analysed through an inductive approach. Results: Five main themes were identified from 24 participants: (1) individual variation of, (2) occurrence of, (3) meaning of knee crepitus, (4) attitudes and exercise behaviours regarding crepitus, and (5) knowledge deficits and needs concerning crepitus during exercise. The variety of crepitus sounds described occurred with a range of exercises or after inactivity. For those already with osteoarthritis or other symptoms, crepitus was of less concern than symptoms such as pain. Most participants had not ceased exercise but may have modified movement due to crepitus and associated symptoms; some had increased intentional strength training to try alleviating it. Participants agreed that more understanding about the processes causing crepitus and what exercise was safe for knee health would be beneficial. Conclusion: Crepitus does not appear to be a major cause of concern for people who experience it. However, it is a factor that influences exercise behaviours as is pain. If health professionals could guide people with concerns about their crepitus, they may be more confident in exercising to benefit their joint health
Visual similarity effects on masked priming.
We investigated the role of the visual similarity of masked primes to targets in a lexical decision experiment. In the primes, some letters in the target (e.g., A in ABANDON) had either visually similar letters (e.g., H), dissimilar letters (D), visually similar digits (4), or dissimilar digits (6) substituted for them. The similarities of the digits and letters to the base letter were equated and verified in a two-alternative forced choice (2AFC) perceptual identification task. Using targets presented in lowercase (e.g., abandon) and primes presented in uppercase, visually similar digit primes (e.g., 484NDON) produced more priming than did visually dissimilar digit primes (676NDON), but little difference was found between the visually similar and dissimilar letter primes (HRHNDON vs. DWDNDON). These results were explained in terms of task-driven competition between the target letter and the visually similar letter
