    VISUAL DETECTION OF RESPONSE TIME IN ATHLETICS: A “GOLD STANDARD”?

    The response time (RT) in the sprint start in athletics is determined automatically from sensors on the blocks but the final decision on whether an athlete is disqualified is determined by visual inspection of the sensor data. This study explored the level of uncertainty of the visual detection of athletes’ RT in the sprint start. Fifteen sprinters performed six sprint starts while forces from the front block, rear block and hands were recorded. Two experimenters performed visual RT detection on two consecutive occasions using two sizes for the analysis window. Intra- and inter-reliability analysis indicated that the mean level of uncertainty of the visual detection was approximately 20 ms for the starting block data. More research is needed to formally assess the precision of the visual detection, which may result in changes in the current false start regulation

    MAPPING OUT THE RESPONSE SEQUENCE OF THE SPRINT START

    Establishing the limits of sprint start response time (SSRT) requires the mapping of the muscular sequence of activation and mechanical response delays and was the aim of the current study. Sprint start performance of 15 sprinters was examined with kinematic, EMG, and block force data collected. A general muscle activation sequence was identified, with both deltoid muscles, the rear leg rectus femoris, and the rear leg tibialis anterior the first muscles to increase activation from the set position. With ankle dorsiflexion the initial motion during the block push, examining the period between tibialis anterior muscle onset and block force onset is critical for quantifying mechanical response delays . Estimates of this delay period were as low as 7 ms which has implications for our understanding of the minimum SSRT a sprinter can legally produce

    A genome-wide library of CB4856/N2 introgression lines of Caenorhabditis elegans

    Recombinant inbred lines (RILs) derived from Caenorhabditis elegans wild-type N2 and CB4856 are increasingly being used for mapping genes underlying complex traits. To speed up mapping and gene discovery, introgression lines (ILs) offer a powerful tool for more efficient QTL identification. We constructed a library of 90 ILs, each carrying a single homozygous CB4856 genomic segment introgressed into the genetic background of N2. The ILs were genotyped by 123 single-nucleotide polymorphism (SNP) markers. The proportion of the CB4856 segments in most lines does not exceed 3%, and together the introgressions cover 96% of the CB4856 genome. The value of the IL library was demonstrated by identifying novel loci underlying natural variation in two ageing-related traits, i.e. lifespan and pharyngeal pumping rate. Bin mapping of lifespan resulted in six QTLs, which all have a lifespan-shortening effect on the CB4856 allele. We found five QTLs for the decrease in pumping rate, of which four colocated with QTLs found for average lifespan. This suggests pleiotropic or closely linked QTL associated with lifespan and pumping rate. Overall, the presented IL library provides a versatile resource toward easier and efficient fine mapping and functional analyses of loci and genes underlying complex traits in C. elegans

    Whole Genome Sequencing Highlights Genetic Changes Associated with Laboratory Domestication of C. elegans

    Defining the mutational landscape when individuals of a species grow separately and diverge over many generations can provide insights into trait evolution. A specific example of this involves studying changes associated with domestication where different lines of the same wild stock have been cultivated independently in different standard environments. Whole genome sequence comparison of such lines permits estimation of mutation rates, inference of genes' ancestral states and ancestry of existing strains, and correction of sequencing errors in genome databases. Here we study domestication of the C. elegans Bristol strain as a model, and report the genome sequence of LSJ1 (Bristol), a sibling of the standard C. elegans reference wild type N2 (Bristol). The LSJ1 and N2 lines were cultivated separately from shortly after the Bristol strain was isolated until methods to freeze C. elegans were developed. We find that during this time the two strains have accumulated 1208 genetic differences. We describe phenotypic variation between N2 and LSJ1 in the rate at which embryos develop, the rate of production of eggs, the maturity of eggs at laying, and feeding behavior, all the result of post-isolation changes. We infer the ancestral alleles in the original Bristol isolate and highlight 2038 likely sequencing errors in the original N2 reference genome sequence. Many of these changes modify genome annotation. Our study provides a starting point to further investigate genotype-phenotype association and offers insights into the process of selection as a result of laboratory domestication

    À partir de l’été 1914, les sociétés européennes paraissent brutalement saisies par la guerre et, ce faisant, saisies par l’État. C’est en son nom que des millions d’hommes vont s’affronter, sous l’uniforme, et que s’opère une gigantesque « mobilisation » des corps, des esprits et des ressources, pour reprendre le terme de l’époque toujours employé par les historiens et les historiennes. Cent ans plus tard, alors que tous les États ayant fait la guerre ont engagé de vastes programmes de commémoration, le moment semblait particulièrement opportun pour comprendre comment l’État parvient à faire la guerre et ce que la guerre fait à l’État. L’emprise de l’État est-elle immédiate, progressive, continue ou discontinue ? Connaît-elle des phases d’essoufflement, des ratés ? S’accompagne-t-elle de phénomènes parallèles de « déprise » ? Loin de toute généralité ou de toute extrapolation hasardeuse, est-il possible de repérer des formes de résistance ou d’évitement ? Interroger le processus de nationalisation des sociétés européennes, tel est l’un des enjeux de cet ouvrage pluridisciplinaire, largement ouvert dans l’espace et dans le temps autour du point de référence de 1914. Composé d’enquêtes bien circonscrites, l’ouvrage s’inscrit dans une histoire sociale de la guerre, et permet de questionner ce qui semble une évidence, au moins en France : la spectaculaire capacité de l’État à mobiliser, presque du jour au lendemain, une société tout entière.From the summer of 1914, European societies seem brutally seized by war and, as a consequence, seized by the State. In the name of the State, millions of men enrolled in the armed forces are to fight one another. Bodies, minds and resources are subjected to a gigantic "mobilization", a contemporary word still used by historians. A hundred years later, when all the warring States, as well as the States that were born from the conflict, are launching ambitious commemorative programs, the moment seems well chosen to study how the State wages war and, in return, how war transforms the State. As part of this vast topic, this international and multidisciplinary (history, political science, sociology) conference will address the invention of the War State, from the perspective of all the processes through which the event has – or does not have – an impact on the organisation, actions and conduct of the public power. The aim is to identify potential changes or limited adjustments, but always within situations of transition born from the conflict. Is the intensification of the State's hold on society immediate or gradual, continuous or discontinuous? Are there slower phases, failures? Is it paralleled with a loss of influence in other areas? Is it possible to detect forms of resistance or avoidance, while refraining from all generalizations and risky extrapolations? Questioning the process of nationalisation of European societies is one of the challenges of this multidisciplinary work, which is widely open in space and time around the 1914 key date. Composed of well-defined surveys, the book falls within a social perspective of war, and allows us to question what seems obvious, at least in France: the spectacular capacity of the State to mobilize, almost overnight, an entire society

    Bias and Evolution of the Mutationally Accessible Phenotypic Space in a Developmental System

    Genetic and developmental architecture may bias the mutationally available phenotypic spectrum. Although such asymmetries in the introduction of variation may influence possible evolutionary trajectories, we lack quantitative characterization of biases in mutationally inducible phenotypic variation, their genotype-dependence, and their underlying molecular and developmental causes. Here we quantify the mutationally accessible phenotypic spectrum of the vulval developmental system using mutation accumulation (MA) lines derived from four wild isolates of the nematodes Caenorhabditis elegans and C. briggsae. The results confirm that on average, spontaneous mutations degrade developmental precision, with MA lines showing a low, yet consistently increased, proportion of developmental defects and variants. This result indicates strong purifying selection acting to maintain an invariant vulval phenotype. Both developmental system and genotype significantly bias the spectrum of mutationally inducible phenotypic variants. First, irrespective of genotype, there is a developmental bias, such that certain phenotypic variants are commonly induced by MA, while others are very rarely or never induced. Second, we found that both the degree and spectrum of mutationally accessible phenotypic variation are genotype-dependent. Overall, C. briggsae MA lines exhibited a two-fold higher decline in precision than the C. elegans MA lines. Moreover, the propensity to generate specific developmental variants depended on the genetic background. We show that such genotype-specific developmental biases are likely due to cryptic quantitative variation in activities of underlying molecular cascades. This analysis allowed us to identify the mutationally most sensitive elements of the vulval developmental system, which may indicate axes of potential evolutionary variation. Consistent with this scenario, we found that evolutionary trends in the vulval system concern the phenotypic characters that are most easily affected by mutation. This study provides an empirical assessment of developmental bias and the evolution of mutationally accessible phenotypes and supports the notion that such bias may influence the directions of evolutionary change
