High fludarabine exposure and relationship with treatment-related mortality after nonmyeloablative hematopoietic cell transplantation.
Despite its common use in nonmyeloablative preparative regimens, the pharmacokinetics of fludarabine are poorly characterized in hematopoietic cell transplantation (HCT) recipients and exposure-response relationships remain undefined. The objective of this study was to evaluate the association between plasma F-ara-A exposure, the systemically circulating moiety of fludarabine, and engraftment, acute GVHD, TRM and OS after HCT. The preparative regimen consisted of CY 50 mg/kg/day i.v. day -6; plus fludarabine 30-40 mg/m²/day i.v. on days -6 to -2 and TBI 200 cGy on day -1. F-ara-A pharmacokinetics were carried out with the first dose of fludarabine in 87 adult patients. Median (range) F-ara-A area-under-the-curve (AUC((0-∞))) was 5.0 μg h/mL (2.0-11.0), clearance 15.3 L/h (6.2-36.6), C(min) 55 ng/mL (17-166) and concentration on day(zero) 16.0 ng/mL (0.1-144.1). Despite dose reductions, patients with renal insufficiency had higher F-ara-A exposures. There was strong association between high plasma concentrations of F-ara-A and increased risk of TRM and reduced OS. Patients with an AUC((0-∞)) greater than 6.5 μg h/mL had 4.56 greater risk of TRM and significantly lower OS. These data suggest that clinical strategies are needed to optimize dosing of fludarabine to prevent overexposure and toxicity in HCT
"The California critical thinking instruments for benchmarking, program assessment, and directing curricular change"

They can be contacted at: [email protected], [email protected], and [email protected]. To assess pharmacy students’ critical thinking (CT) measures and identify areas for curricular reform.
Methods. Pharmacy students were given the California Critical Thinking Skills Test and Disposition Index at various points in the PharmD program. Scores were compared with a national referent group and evaluated for changes across the curriculum and between classes.
Results. Students were comparable to national norms. Pretest and posttest scores for total disposition showed improvement. Scores in all subcategories except for truth-seeking were consistently above 40. The CT skills of the pharmacy students varied compared with those of referent students, but the pharmacy students’ overall score of 18 was in the 73rd percentile. Pre- and post-skills scores showed improvement. Students scoring low on the pretest improved more than those scoring high.
Conclusions. Students had a consistent disposition towards CT and compared favorably to national
norms. Both disposition and skills improved across the curriculum. Dimensions of critical thinking on
which students score low should be areas for curricular and other program changes
Climatic Disequilibrium Threatens Conservation Priority Forests

Available from Wiley Open Access via the DOI in this record.© 2017 Wiley Periodicals, Inc.We test the hypothesis that climatic changes since 1800 have resulted in unrealized potential vegetation changes that represent a "climatic debt" for many ecosystems. Caledonian pinewoods, an EU priority forest type, are used as a model system to explore potential impacts of two centuries of climatic change upon sites of conservation importance and surrounding landscapes. Using methods that estimate topographic microclimate, current and preindustrial climates were estimated for 50 m grid cells and simulations made using a dynamic vegetation model. Core Caledonian pinewood areas are now less suitable for growth of pine and more favorable for oak than in 1800, whereas landscapes as a whole are on average more favorable for both. The most favorable areas for pine are now mainly outside areas designated to conserve historical pinewoods. A paradigm shift is needed in formulating conservation strategies to avoid catastrophic losses of this habitat, and of many others globally with trees or other long-lived perennials as keystone species.Natural Environment Research Council. VEGF(164)-mediated inflammation is required for pathological, but not physiological, ischemia-induced retinal neovascularization
Hypoxia-induced VEGF governs both physiological retinal vascular development and pathological retinal neovascularization. In the current paper, the mechanisms of physiological and pathological neovascularization are compared and contrasted. During pathological neovascularization, both the absolute and relative expression levels for VEGF(164) increased to a greater degree than during physiological neovascularization. Furthermore, extensive leukocyte adhesion was observed at the leading edge of pathological, but not physiological, neovascularization. When a VEGF(164)-specific neutralizing aptamer was administered, it potently suppressed the leukocyte adhesion and pathological neovascularization, whereas it had little or no effect on physiological neovascularization. In parallel experiments, genetically altered VEGF(164)-deficient (VEGF(120/188)) mice exhibited no difference in physiological neovascularization when compared with wild-type (VEGF(+/+)) controls. In contrast, administration of a VEGFk-1/Fc fusion protein, which blocks all VEGF isoforms, led to significant suppression of both pathological and physiological neovascularization. In addition, the targeted inactivation of monocyte lineage cells with clodronate-liposomes led to the suppression of pathological neovascularization. Conversely, the blockade of T lymphocyte-mediated immune responses with an anti-CD2 antibody exacerbated pathological neovascularization. These data highlight important molecular and cellular differences between physiological and pathological retinal neovascularization. During pathological neovascularization, VEGF(164) selectively induces inflammation and cellular immunity. These processes provide positive and negative angiogenic regulation, respectively. Together, new therapeutic approaches for selectively targeting pathological, but not physiological, retinal neovascularization are outlined

Reliable microsatellite genotyping of the Eurasian badger (Meles meles) using faecal DNA
The potential link between badgers and bovine tuberculosis has made it vital to develop
accurate techniques to census badgers. Here we investigate the potential of using genetic
profiles obtained from faecal DNA as a basis for population size estimation. After trialling
several methods we obtained a high amplification success rate (89%) by storing faeces in
70% ethanol and using the guanidine thiocyanate/silica method for extraction. Using 70%
ethanol as a storage agent had the advantage of it being an antiseptic. In order to obtain reliable
genotypes with fewer amplification reactions than the standard multiple-tubes
approach, we devised a comparative approach in which genetic profiles were compared
and replication directed at similar, but not identical, genotypes. This modified method
achieved a reduction in polymerase chain reactions comparable with the maximumlikelihood
model when just using reliability criteria, and was slightly better when using
reliability criteria with the additional proviso that alleles must be observed twice to be considered
reliable. Our comparative approach would be best suited for studies that include
multiple faeces from each individual. We utilized our approach in a well-studied population
of badgers from which individuals had been sampled and reliable genotypes obtained.
In a study of 53 faeces sampled from three social groups over 10 days, we found that direct
enumeration could not be used to estimate population size, but that the application of
mark–recapture models has the potential to provide more accurate results
The Neuroscience Information Framework: A Data and Knowledge Environment for Neuroscience
With support from the Institutes and Centers forming the NIH Blueprint for Neuroscience Research, we have designed and implemented a new initiative for integrating access to and use of Web-based neuroscience resources: the Neuroscience Information Framework. The Framework arises from the expressed need of the neuroscience community for neuroinformatic tools and resources to aid scientific inquiry, builds upon prior development of neuroinformatics by the Human Brain Project and others, and directly derives from the Society for Neuroscience’s Neuroscience Database Gateway. Partnered with the Society, its Neuroinformatics Committee, and volunteer consultant-collaborators, our multi-site consortium has developed: (1) a comprehensive, dynamic, inventory of Web-accessible neuroscience resources, (2) an extended and integrated terminology describing resources and contents, and (3) a framework accepting and aiding concept-based queries.
