    The Positive Role of Negative Emotions: Fear, Anxiety, Conflict and Resistance as Productive Experiences in Academic Study and in the Emergence of Learner Autonomy

    Although affect is widely recognized as a powerful force in determining students’ academic success,researchers and practitioners have paid little attention to emotional barriers that often impede college success or how instructors may respond constructively when such barriers arise. The purpose of this paper is to initiate discussion of this important problem by offering a model of how an initially resistant, fearful, and/or anxious student can use emotionally unpleasant experiences to transform himself or herself into a more autonomous and successful learner. We offer prima facie support for this model by presenting the results of two cases of first year students. Although this model may not apply to all anxious first year students, it nevertheless has value (a) as a resource for instructors working with students who fit this pattern and (b) as an example of how the role of emotions in learning can profitably be studied

    Spatial intensity distribution analysis quantifies the extent and regulation of homodimerization of the secretin receptor

    Previous studies have indicated that the G protein-coupled secretin receptor is present as a homo-dimer, organized through symmetrical contacts in transmembrane domain IV, and that receptor dimerization is critical for high potency signalling by secretin. However, whether all of the receptor exists in the dimeric form or if this is regulated, is unclear. We used measures of quantal brightness of the secretin receptor tagged with monomeric enhanced green fluorescent protein (mEGFP) and Spatial Intensity Distribution Analysis to assess this. Calibration using cells expressing plasma membrane-anchored forms of mEGFP initially allowed demonstration that the Epidermal Growth Factor receptor is predominantly monomeric in the absence of ligand and whilst wild type receptor was rapidly converted to a dimeric form by ligand, a mutated form of this receptor remained monomeric. Equivalent studies showed that at moderate expression levels the secretin receptor exists as a mixture of monomeric and dimeric forms, with little evidence of higher-order complexity. However, sodium butyrate induced up-regulation of the receptor resulted in a shift from monomeric towards oligomeric organization. By contrast, a form of the secretin receptor containing a pair of mutations on the lipid-facing side of transmembrane domain IV was almost entirely monomeric. Down-regulation of the secretin receptor-interacting G protein Gαs did not alter receptor organization, indicating that dimerization is defined specifically by direct protein-protein interactions between copies of the receptor polypeptide, whilst short term treatment with secretin had no effect on organization of the wild type receptor but increased the dimeric proportion of the mutated receptor variant

    Hydrological assessment of urban nature-based solutions for urban planning using ecosystem service toolkit applications

    Ecosystem Service (ES) toolkits are increasingly used to quantify and visualize the benefits gained from Nature-based-solutions (NBS) but modules for hydrology are often absent, or if present they lack meaningful hydrological functionality or validation. This leads to gaps in the evidence base required by decision makers. To bridge the gap between such limitations and more complex hydrological models this paper presents a hydrologically based NBS model compatible with spatial ES toolkits. The approach ‘Adapted Nature-based-solutions Rational Method’ (ANaRM) is based on the Rational Method, widely used in hydrology. We apply this model to the city of Birmingham, England, to validate its performance and to analyse the effects of different NBS scenarios. The validated ANaRM model provided robust estimates of peak flow using design storm rainfall. It proved capable of simulating the hydrological effects of NBS such as land use change from urban to green, or installation of SuDS and ponds. Results suggest ponds are found most effective for achieving peak flow reduction in channels and are the best option for mitigating fluvial flooding downstream. Reduction in localised runoff and pluvial flooding is best achieved by converting impervious surfaces such as buildings, hardstanding and roads to green solutions such as green roofs, permeable pavements and greenspace. This paper highlights the importance of considering the spatial effects of urban NBS on hydrology, and that these can be captured with relatively simple modelling approaches such as ANaRM. Its ease of use means it suits any level of user looking to represent the flood mitigation aspect of NBS spatially and has high potential as part of any ES toolkit focused on representing the spatial effects of NBS on ecosystem services

    The use of routine outcome measures in two child and adolescent mental health services: a completed audit cycle

    Background: Routine outcome measurement (ROM) is important for assessing the clinical effectiveness of health services and for monitoring patient outcomes. Within Child and Adolescent Mental Health Services (CAMHS) in the UK the adoption of ROM in CAMHS has been supported by both national and local initiatives (such as government strategies, local commissioning policy, and research). Methods: With the aim of assessing how these policies and initiatives may have influenced the uptake of ROM within two different CAMHS we report the findings of two case-note audits: a baseline audit conducted in January 2011 and a re-audit conducted two years later in December 2012-February 2013. Results: The findings show an increase in both the single and repeated use of outcome measures from the time of the original audit, with repeated use (baseline and follow-up) of the Health of the Nation Outcome Scale for Children and Adolescents (HoNOSCA) scale increasing from 10% to 50% of cases. Re-audited case-notes contained more combined use of different outcome measures, with greater consensus on which measures to use. Outcome measures that were applicable across a wide range of clinical conditions were more likely to be used than symptom-specific measures, and measures that were completed by the clinician were found more often than measures completed by the service user. Conclusions: The findings show a substantial improvement in the use of outcome measures within CAMHS. These increases in use were found across different service organisations which were subject to different types of local service priorities and drivers

    The extracellular surface of the GLP-1 receptor is a molecular trigger for biased agonism

    Ligand-directed signal bias offers opportunities for sculpting molecular events, with the promise of better, safer therapeutics. Critical to the exploitation of signal bias is an understanding of the molecular events coupling ligand binding to intracellular signaling. Activation of class B G protein-coupled receptors is driven by interaction of the peptide N terminus with the receptor core. To understand how this drives signaling, we have used advanced analytical methods that enable separation of effects on pathway-specific signaling from those that modify agonist affinity and mapped the functional consequence of receptor modification onto three-dimensional models of a receptor-ligand complex. This yields molecular insights into the initiation of receptor activation and the mechanistic basis for biased agonism. Our data reveal that peptide agonists can engage different elements of the receptor extracellular face to achieve effector coupling and biased signaling providing a foundation for rational design of biased agonists

    Bridging Alone: Religious Conservatism, Marital Homogamy, and Voluntary Association Membership

    This study characterizes social insularity of religiously conservative American married couples by examining patterns of voluntary associationmembership. Constructing a dataset of 3938 marital dyads from the second wave of the National Survey of Families and Households, the author investigates whether conservative religious homogamy encourages membership in religious voluntary groups and discourages membership in secular voluntary groups. Results indicate that couples’ shared affiliation with conservative denominations, paired with beliefs in biblical authority and inerrancy, increases the likelihood of religious group membership for husbands and wives and reduces the likelihood of secular group membership for wives, but not for husbands. The social insularity of conservative religious groups appears to be reinforced by homogamy—particularly by wives who share faith with husbands

    Neptunism and transformism:Robert Jameson and other evolutionary theorists in early nineteenth-century Scotland

    This paper sheds new light on the prevalence of evolutionary ideas in Scotland in the early nineteenth century and establish what connections existed between the espousal of evolutionary theories and adherence to the directional history of the earth proposed by Abraham Gottlob Werner and his Scottish disciples. A possible connection between Wernerian geology and theories of the transmutation of species in Edinburgh in the period when Charles Darwin was a medical student in the city was suggested in an important 1991 paper by James Secord. This study aims to deepen our knowledge of this important episode in the history of evolutionary ideas and explore the relationship between these geological and evolutionary discourses. To do this it focuses on the circle of natural historians around Robert Jameson, Wernerian geologist and professor of natural history at the University of Edinburgh from 1804 to 1854. From the evidence gathered here there emerges a clear confirmation that the Wernerian model of geohistory facilitated the acceptance of evolutionary explanations of the history of life in early nineteenth-century Scotland. As Edinburgh was at this time the most important center of medical education in the English-speaking world, this almost certainly influenced the reception and development of evolutionary ideas in the decades that followed.</p

    Status of Muon Collider Research and Development and Future Plans

    The status of the research on muon colliders is discussed and plans are outlined for future theoretical and experimental studies. Besides continued work on the parameters of a 3-4 and 0.5 TeV center-of-mass (CoM) energy collider, many studies are now concentrating on a machine near 0.1 TeV (CoM) that could be a factory for the s-channel production of Higgs particles. We discuss the research on the various components in such muon colliders, starting from the proton accelerator needed to generate pions from a heavy-Z target and proceeding through the phase rotation and decay (π→μνμ\pi \to \mu \nu_{\mu}) channel, muon cooling, acceleration, storage in a collider ring and the collider detector. We also present theoretical and experimental R & D plans for the next several years that should lead to a better understanding of the design and feasibility issues for all of the components.
