    Time's Up: Homeless New Yorkers Demand Alternatives to Bloomberg's Failed Five-Year Plan

    More people are living in homeless shelters now than when Mayor Bloomberg took office in 2002. On June 24th, 2004, Mayor Michael Bloomberg unveiled his Five Year Plan to reduce homelessness in New York City by two-thirds. The failure of Bloomberg's plan is evidence that what is needed are fundamental changes to housing policy in NYC, which is at the root of what is falsely portrayed as a homeless crisis.New York Magazine has said that his homeless policies are "the single biggest failure of the Bloomberg administration." The 2009 Mayor's Management Report found an across-the-board increase in the shelter census. As the five years of Bloomberg's plan comes to a close, this report focuses on the failures of one of its cornerstones, the Rental Subsidies Programs. Family and child homelessness have increased under these programs, even with thousands of households receiving vouchers, the rental subsidies have built-in obstacles to employment and self-sufficiency so crucial to making the transition out of the shelter system possible

    Exchangeable zinc pool size at birth in Pakistani small for gestational age and appropriate for gestational age infants do not differ but are lower than in US infants

    Objectives: Small for gestational age (SGA) infants are more susceptible to infectious morbidity and growth faltering compared to their appropriate for gestational age (AGA) counterparts. Zinc supplementation of SGA infants may be beneficial but the underlying susceptibility to zinc deficiency of SGA infants has not been examined.Methods: In a community-based, observational, longitudinal study in a peri-urban settlement of Karachi, Pakistan, we compared the size of the exchangeable zinc pools (EZPs) in term SGA and AGA infants at birth and at 6 months of age, hypothesizing that the EZP would be lower in the SGA group. To measure EZP size, a zinc stable isotope was intravenously administered within 48 hours of birth (n = 17 and 22) at 6 months (n = 11 and 14) in SGA and AGA infants, respectively. Isotopic enrichment in urine was used to determine EZP.Results: No significant difference was detected in the mean (±standard deviation) EZP between SGA and AGA infants at birth, with values of 9.8 ± 3.5 and 10.1 ± 4.1 mg/kg, respectively (P = 0.86), or at 6 months. Longitudinal EZP measurements demonstrated a significant decline in EZP relative to body weight in both groups at 6 months (P \u3c 0.001). Mean EZP (adjusted for body weight) size at birth for the combined Pakistani groups was significantly lower than AGA infants at birth in the United States (P = 0.017).Conclusions: These results did not support a difference in zinc endowment between SGA and AGA Pakistani infants. They, however, do suggest lower in utero zinc transfer to the fetus in a setting where poor maternal nutritional status may confer a high susceptibility to postnatal zinc deficiency

    Functional Oligomeric State of Avian Sarcoma Virus Integrase

    Retroviral integrase, one of only three enzymes encoded by the virus, catalyzes the essential step of inserting a DNA copy of the viral genome into the host during infection. Using the avian sarcoma virus integrase, we demonstrate that the enzyme functions as a tetramer. In presteady-state active site titrations, four integrase protomers were required for a single catalytic turnover. Volumetric determination of integrase-DNA complexes imaged by atomic force microscopy during the initial turnover additionally revealed substrate-induced assembly of a tetramer. These results suggest that tetramer formation may be a requisite step during catalysis with ramifications for antiviral design strategies targeting the structurally homologous human immunodeficiency virus, type 1 (HIV-1) integrase

    We report on a 10 ks simultaneous Chandra/HETG-NuSTAR observation of the Bursting Pulsar, GRO J1744-28, during its third detected outburst since discovery and after nearly 18 years of quiescence. The source is detected up to 60 keV with an Eddington persistent flux level. Seven bursts, followed by dips, are seen with Chandra, three of which are also detected with NuSTAR. Timing analysis reveals a slight increase in the persistent emission pulsed fraction with energy (from 10% to 15%) up to 10 keV, above which it remains constant. The 0.5-70 keV spectra of the persistent and dip emission are the same within errors, and well described by a blackbody (BB), a power-law with an exponential rolloff, a 10 keV feature, and a 6.7 keV emission feature, all modified by neutral absorption. Assuming that the BB emission originates in an accretion disc, we estimate its inner (magnetospheric) radius to be about 4x10^7 cm, which translates to a surface dipole field B~9x10^10 G. The Chandra/HETG spectrum resolves the 6.7 keV feature into (quasi-)neutral and highly ionized Fe XXV and Fe XXVI emission lines. XSTAR modeling shows these lines to also emanate from a truncated accretion disk. The burst spectra, with a peak flux more than an order of magnitude higher than Eddington, are well fit with a power-law with an exponential rolloff and a 10~keV feature, with similar fit values compared to the persistent and dip spectra. The burst spectra lack a thermal component and any Fe features.     Common Raven Impacts on Nesting Western Snowy Plovers: Integrating Management to Facilitate Species Recovery

    The U.S. Pacific coast population of the western snowy plover (Charadrius nivosus nivosus; plover) has declined due to loss and degradation of coastal habitats, predation, and anthropogenic disturbance. The U.S. Fish and Wildlife Service listed the subspecies in 1993 as threatened under the Endangered Species Act due to the population declines and habitat loss. Predation of nests and chicks has been identified as an important cause of historic population declines, and thus, most predator management actions for this subspecies are focused on reducing this pressure. In recent years, common ravens (Corvus corax; ravens) have become the most common and pervasive predators of plover nests and chicks, especially in areas with subsidized food sources for ravens and sites without predator management. We compiled data from a variety of sources to document the impact of raven predation on plover nesting success. We discuss current raven management and suggest several tools and strategies to increase plover nesting success, including multi-state approval for the use of the avicide DRC-1339, the use of lures and new trap types, and an increase in funding for predator management. The lack of coordinated and integrated management continues to impede the recovery of the Pacific coast plover population

    The membrane mucin MUC4 is elevated in breast tumor lymph node metastases relative to matched primary tumors and confers aggressive properties to breast cancer cells

    Abstract Introduction Previous studies indicate that overexpression of the membrane-associated mucin MUC4 is potently anti-adhesive to cultured tumor cells, and suppresses cellular apoptotic response to a variety of insults. Such observations raise the possibility that MUC4 expression could contribute to tumor progression or metastasis, but the potential involvement of MUC4 in breast cancer has not been rigorously assessed. The present study aimed to investigate the expression of the membrane mucin MUC4 in normal breast tissue, primary breast tumors and lymph node metastases, and to evaluate the role of MUC4 in promoting the malignant properties of breast tumor cells. Methods MUC4 expression levels in patient-matched normal and tumor breast tissue was initially examined by immunoblotting lysates of fresh frozen tissue samples with a highly specific preparation of anti-MUC4 monoclonal antibody 1G8. Immunohistochemical analysis was then carried out using tissue microarrays encompassing patient-matched normal breast tissue and primary tumors, and patient-matched lymph node metastases and primary tumors. Finally, shRNA-mediated knockdown was employed to assess the contribution of MUC4 to the cellular growth and malignancy properties of JIMT-1 breast cancer cells. Results Immunoblotting and immunohistochemistry revealed that MUC4 levels are suppressed in the majority (58%, p < 0.001) of primary tumors relative to patient-matched normal tissue. On the other hand, lymph node metastatic lesions from 37% (p < 0.05) of patients expressed higher MUC4 protein levels than patient-matched primary tumors. MUC4-positive tumor emboli were often found in lymphovascular spaces of lymph node metastatic lesions. shRNA-mediated MUC4 knockdown compromised the migration, proliferation and anoikis resistance of JIMT-1 cells, strongly suggesting that MUC4 expression actively contributes to cellular properties associated with breast tumor metastasis. Conclusions Our observations suggest that after an initial loss of MUC4 levels during the transition of normal breast tissue to primary tumor, the re-establishment of elevated MUC4 levels confers an advantage to metastasizing breast tumor cells by promoting the acquisition of cellular properties associated with malignancy

    Perspective on Coarse-Graining, Cognitive Load, and Materials Simulation

    The predictive capabilities of computational materials science today derive from overlapping advances in simulation tools, modeling techniques, and best practices. We outline this ecosystem of molecular simulations by explaining how important contributions in each of these areas have fed into each other. The combined output of these tools, techniques, and practices is the ability for researchers to advance understanding by efficiently combining simple models with powerful software. As specific examples, we show how the prediction of organic photovoltaic morphologies have improved by orders of magnitude over the last decade, and how the processing of reacting epoxy thermosets can now be investigated with million-particle models. We discuss these two materials systems and the training of materials simulators through the lens of cognitive load theory. For students, the broad view of ecosystem components should facilitate understanding how the key parts relate to each other first, followed by targeted exploration. In this way, the paper is organized in loose analogy to a coarse-grained model: The main components provide basic framing and accelerated sampling from which deeper research is better contextualized. For mentors, this paper is organized to provide a snapshot in time of the current simulation ecosystem and an on-ramp for simulation experts into the literature on pedagogical practice

    The quality of surgical pathology care for men undergoing radical prostatectomy in the U.S.

    BACKGROUND. The authors assessed adherence with the College of American Pathologists (CAP) radical prostatectomy (RP) practice protocol in a national sample of men who underwent RP for early-stage prostate cancer. METHODS. Using the National Cancer Data Base, the authors identified a nationally representative sample of 1240 men (unweighted) who underwent RP. For each patient, local cancer registrars performed an explicit medical record review to assess patient-level compliance with surgical pathology report documentation of 7 morphologic criteria (ie, quality indicators). Applying the CAP prognostic factor classification framework, composite measures and all-or-none measures of quality indicator compliance were calculated for the following analytic categories: 1) a strict subset of CAP category I prognostic factors (3 indicators), 2) a broad subset of CAP category I factors (6 indicators), and 3) the full set of 7 indicators. RESULTS. Among a weighted sample of 24,420 patients who underwent RP, compliance with documentation of the CAP category I factors varied from 54% (95% confidence interval [95% CI], 50–58%) for pathologic tumor, lymph node, metastases classification (according to the American Joint Committee on Cancer staging system) to 97% (95% CI, 96–99%) for Gleason score. In composite, RP pathology reports contained 83% (95% CI, 81–84%), 85% (95% CI, 84–87%), and 79% (95% CI, 78–80%) of the recommended data elements measured by the strict CAP category I subset, the broad CAP category I subset, and the full set of 7 indicators, respectively. In contrast to the generally higher composite scores, only 52% (95% CI, 48–56%) and 41% (95% CI, 37–45%) of men who underwent RP had complete documentation in their pathology reports for the strict and broad CAP category I subsets, respectively. CONCLUSIONS. RP surgical pathology reports contained most of the recommended data elements; however, the frequent absence of pathologic stage provides an opportunity for quality improvement.     Testing both affordability-availability and psychological-coping mechanisms underlying changes in alcohol use during the COVID-19 pandemic

    Two theoretical perspectives have been proffered to explain changes in alcohol use during the pandemic: the ‘affordability-availability’ mechanism (i.e., drinking decreases due to changes in physical availability and/or reduced disposable income) and the ‘psychological-coping’ mechanism (i.e., drinking increases as adults attempt to cope with pandemic-related distress). We tested these alternative perspectives via longitudinal analyses of the COVID-19 Psychological Consortium (C19PRC) Study data (spanning three timepoints during March to July 2020). Respondents provided data on psychological measures (e.g., anxiety, depression, posttraumatic stress, paranoia, extraversion, neuroticism, death anxiety, COVID-19 anxiety, intolerance of uncertainty, resilience), changes in socio-economic circumstances (e.g., income loss, reduced working hours), drinking motives, solitary drinking, and ‘at-risk’ drinking (assessed using a modified version of the AUDIT-C). Structural equation modelling was used to determine (i) whether ‘at-risk’ drinking during the pandemic differed from that recalled before the pandemic, (ii) dimensions of drinking motives and the psychosocial correlates of these dimensions, (iii) if increased alcohol consumption was predicted by drinking motives, solitary drinking, and socio-economic changes. The proportion of adults who recalled engaging in ‘at-risk’ drinking decreased significantly from 35.9% pre-pandemic to 32.0% during the pandemic. Drinking to cope was uniquely predicted by experiences of anxiety and/or depression and low resilience levels. Income loss or reduced working hours were not associated with coping, social enhancement, or conformity drinking motives, nor changes in drinking during lockdown. In the earliest stage of the pandemic, psychological-coping mechanisms may have been a stronger driver to changes in adults’ alcohol use than ‘affordability-availability’ alone
