    Cardiac-Specific Expression of the Tetracycline Transactivator Confers Increased Heart Function and Survival Following Ischemia Reperfusion Injury

    Mice expressing the tetracycline transactivator (tTA) transcription factor driven by the rat α-myosin heavy chain promoter (α-MHC-tTA) are widely used to dissect the molecular mechanisms involved in cardiac development and disease. However, these α-MHC-tTA mice exhibit a gain-of-function phenotype consisting of robust protection against ischemia/reperfusion injury in both in vitro and in vivo models in the absence of associated cardiac hypertrophy or remodeling. Cardiac function, as assessed by echocardiography, did not differ between α-MHC-tTA and control animals, and there were no noticeable differences observed between the two groups in HW/TL ratio or LV end-diastolic and end-systolic dimensions. Protection against ischemia/reperfusion injury was assessed using isolated perfused hearts where α-MHC-tTA mice had robust protection against ischemia/reperfusion injury which was not blocked by pharmacological inhibition of PI3Ks with LY294002. Furthermore, α-MHC-tTA mice subjected to coronary artery ligation exhibited significantly reduced infarct size compared to control animals. Our findings reveal that α-MHC-tTA transgenic mice exhibit a gain-of-function phenotype consisting of robust protection against ischemia/reperfusion injury similar to cardiac pre- and post-conditioning effects. However, in contrast to classical pre- and post-conditioning, the α-MHC-tTA phenotype is not inhibited by the classic preconditioning inhibitor LY294002 suggesting involvement of a non-PI3K-AKT signaling pathway in this phenotype. Thus, further study of the α-MHC-tTA model may reveal novel molecular targets for therapeutic intervention during ischemic injury

    SCOR: A secure international informatics infrastructure to investigate COVID-19

    Global pandemics call for large and diverse healthcare data to study various risk factors, treatment options, and disease progression patterns. Despite the enormous efforts of many large data consortium initiatives, scientific community still lacks a secure and privacy-preserving infrastructure to support auditable data sharing and facilitate automated and legally compliant federated analysis on an international scale. Existing health informatics systems do not incorporate the latest progress in modern security and federated machine learning algorithms, which are poised to offer solutions. An international group of passionate researchers came together with a joint mission to solve the problem with our finest models and tools. The SCOR Consortium has developed a ready-to-deploy secure infrastructure using world-class privacy and security technologies to reconcile the privacy/utility conflicts. We hope our effort will make a change and accelerate research in future pandemics with broad and diverse samples on an international scale

    Single-particle probing of edge-state formation in a graphene nanoribbon

    We investigate the effect of a perpendicular magnetic field on the single-particle charging spectrum of a graphene quantum dot embedded inline with a nanoribbon. We observe uniform shifts in the single-particle spectrum which coincide with peaks in the magnetoconductance, implicating Landau level condensation and edge state formation as the mechanism underlying magnetic field-enhanced transmission through graphene nanostructures. The experimentally determined ratio of bulk to edge states is supported by single-particle band-structure simulations, while a fourfold beating of the Coulomb blockade transmission amplitude points to many-body interaction effects during Landau level condensation of the Μ=0 state. © 2012 American Physical Society

    The role of adult education and learning policy in fostering societal sustainability

    The idea of “sustainability” as a core value has slowly permeated policy and practice at governmental and institutional levels, in public and private policy. However, at times when social and economic crises have revealed the fragility of existing institutions and policies, it is important to consider how sustainability is – and could be – integrated into educational policies. In this theoretical contribution to a special issue on “Societal sustainability”, the authors draw on available literature and knowledge. They begin their paper by summarising the conditions under which the concept of “sustainability” entered political discourse in the early 1970s and outline how it has influenced educational research. They then introduce the longstanding debate about the relative role of tradition (in terms of traditional cultural and social order) and change (in terms of efforts to provide learning opportunities for everyone) in adult education. Finally, they argue for a rethinking of the ontology of sustainability: this, they suggest, can shed new light on its relationships with adult education and learning and social justice

    New specimen sheds light on the anatomy and taxonomy of the Early Late Triassic Dinosauriforms from the Chañares Formation, NW Argentina

    The lower Carnian levels of the Chañares Formation (IschigualastoVilla UniĂłn Basin, La Rioja Province) have yielded some of the most informative dinosaur precursor species known so far. However, these species are based on partial skeletons that in several cases hamper the comparison between them because of the absence of overlapping bones. This has generated a contentious debate during the last 20 years about the synonymy between two of these species, Lewisuchus admixtus and Pseudolagosuchus major. Here, we describe a new dinosauriform partial skeleton (CRILARPv 552) recently collected in the Chañares Formation that preserves previously unknown anatomical regions for the dinosaur precursors of this unit (e.g., premaxilla, inner ear, anterior zeugopodium) and allows comparisons with other dinosauriform specimens. CRILAR-Pv 552 is referred to Lewisuchus admixtus because it possesses a proportionally large skull, a laterally projected, shelf-like ridge on the jugal, and recurved, finely serrated middle–posterior maxillary and dentary teeth ankylosed to the bone, and the absence of a coracoid foramen. The new specimen preserves a dorsally bowed dentary with a lateroventral shelf that is identical to a dentary associated with the holotype of Lewisuchus admixtus. Additionally, the morphology of the new specimen is completely congruent with that of specimens of Pseudolagosuchus major, bolstering the hypothesis that the latter species is a subjective junior synonym of Lewisuchus admixtus.
