    Production of Human Type Glycosylated Tissue Plasminogen Activator and the Role of Its Carbohydrate Moiety

    To begin the examination of the role of human type carbohydrate moiety of tissue plasminogen activator (t-PA) on the binding of the enzyme to fibrin,the naturally glycosylated enzyme was produced by microcarrier culture of human cells established from normal uterine muscle. The cells grown on microcarrers in Haunks' MEM supplemented with 10 % FBS (1.6 x 10 6 cells/ml) rapidly detached themselves from microcarrier in a serum-free medium (t-PA production medium) within 5 days, and it was difficult to produce t-PA for long time (t-PA production: an average of 3 IU/ml/day over 5 days). Addition of 0.5% beef extract to the serum-free medium suppressed their detaching from microcarriers. By regulating the pH (7.4) and dissolved oxygen(4 ppm) of the serum free medium,the cell density of microcarrier culture increased to 1.2 x10 7 cells/ml and t-PA was produced over 38 days (t-PA production: an average of 836 IU/ml/day over 38 days). Native t-PAs purified to homogeneity from the culture broth had the molecular weight of 63,000 and 65,000 containing 5.6 and 8.5 % carbohydrate,respectively (molecular mass of protein moiety was calculated to be 59,500). By enzymatic digestion of carbohydrate moiety in native t-PA,we obtained partially deglycosylated t-PA with molecular weights of 60,000 and 62,000. Completely deglycosylated t-PA was obtained by t-PA production in the presence of 10μg/ml tunicamycin (N-glycosylation inhibitor). This suggests that N-glycosylationof t-PA occurs while O-glycosylation is absent. The binding strength of these enzymes to fibrin increased with decrease of the carbohydrate content. The carbohydrate moiety of human type glycosylated t-PA probably modulates the binding strength of the enzyme to fibrin

    女性の年齢と食育の認識が食生活に及ぼす影響

    本研究では、成人女性の年齢別に食育の認識が食生活にどのような影響を及ぼしたのかを明らかにすることを目的とし分析を行った。その結果、年齢の差が食育の認識や多くの食生活に影響していた。また、年齢別に食育の認識と食生活を比較すると、全年齢において食育の認識が「食事バランスガイド・食生活指針の認識」に影響していた。一部の年齢では「朝食摂取頻度」「食前・食後の挨拶」「食品表示の確認」「よく噛むことへの意識」に食育の認識の有無が影響していたが、「家族との共食頻度」「夕食開始時刻」等は全年齢において食育の認識の影響が確認されなかった。今後の成人女性に対する食育活動では、食育の認識がない者に対してはこれまでと同様に食育の普及に努め、食育を認識できている者に対しては家族との共食頻度の増加や夕食開始時刻の健康への影響等の食育における意味を周知させることが必要と考える。In this study, in order to clarify how awareness of dietary education had an effect on dietary habit according to adult\u27s female-age, we analyzed the data. As a result, it was found that age-gap had an effect on awareness of dietary education and many dietary habits. When comparing awareness of dietary education with dietary habit according to age, it was revealed that awareness of dietary education had an effect on“guidance of well-balanced meal and awareness of guideline of dietary habit”in all age groups. Although, in some generation, presence or absence of dietary awareness had an effect on“frequency of having breakfast”,“greeting before/after meal”,“confirmation of food-labeling”and“awareness of chewing-well”, but it was not confirmed that awareness of diet had an effect on“frequency of having meals with family”nor on“starting time of dinner”in all generations. The results indicate the positive benefits of popularizing dietary education among adult females who are not knowledgeable about this topic. At the same time, it is important to inform health-conscious people of the health effects of having meals with family and having dinner at a certain time

    Correlation between HIV disease and lipid metabolism in antiretroviral-naïve HIV-infected patients in Japan

    Antiretroviral therapy alters lipid metabolism in HIV-infected patients. However, interpreting the impact of HIV infection on lipid metabolism is difficult because of various associated factors, including antiretroviral drugs and demographic characteristics. A few studies have associated HIV infection with lipid metabolism in antiretroviral-naïve HIV-infected patients. Because there were no data in this regard from Japan, the present study examined the impact of HIV infection, as well as demographic and clinical features, on lipid metabolism in antiretroviral-naïve HIV-infected patients in Japan. We performed a cross-sectional study to examine the impact of HIV disease, demographic and clinical characteristics on lipid metabolism among 168 HIV-infected Japanese men who were antiretroviral naïve and who did not have hemophilia, including patients who took medication for dyslipidemia. The mean age of the patients was 45.7 years; 0.6% of the patients took medication to dyslipidemia. The mean CD4 lymphocyte count was 289/μL, the mean baseline log10 HIV viral load was 4.2 HIV-1 RNA copies/mL, and 22% of the patients had a history of AIDS-defining events. A higher HDL-C concentration was associated with a higher CD4 lymphocyte count (p = 0.043). Also, a higher LDL-C concentration was associated with a higher CD4 lymphocyte count (p = 0.003). Infection with HIV was associated with dyslipidemia in antiretroviral-naïve patients. More advanced HIV disease was associated with less favorable lipid homeostatic profiles. These results are similar to findings from other countries

    健康への配慮とメタボリックシンドローム対策状況について -佐世保市民の健康に関する実態調査報告-

    本研究では、健康への意識の有無とメタボリックシンドローム(以下、MetS)予防および改善行動の実践の有無から分析を行い、意識や行動の違いが健康に関する項目にどのような影響を与えるのか検討を行った。その結果、健康への意識を持つことにより日常的に健康の維持増進に向けた生活習慣を送り、健康診断やがん検診の受診など健康に向けた望ましい行動や、共食日数の増加や余暇活動の増加など QOL の向上にもつながることが示唆された。また、健康を意識することに加えて MetS 予防を行っているものでは、さらに望ましい生活習慣状況となっており、MetS 予防といった明確な目的およびその対策を実践しているという自覚によってより一層健康的な日常生活の実践に結びついていることが考えられた。In the present study, we analyzed the data from the presence or absence of the awareness of health and from the presence or absence of the practice for prevention of metabolic syndrome(hereinafter called“MetS”)and the behavior for improvement, and examined what effect the differences in the awareness and behavior might influence on the items on health. As a result, it was suggested that, by having an awareness of health, it would be possible to establish a life-style enabling the maintenance and promotion of health, and to be linked to the desirable behavior for health such as having regularly a health check-up and a cancer examination, and to go far towards the advancement of QOL such as the increased number of days of‘eating in company\u27and the increased level of activity for leisure. In addition to having an awareness of health, those who were conducting the preventive measure against MetS were found to lead a more, desirable life-style, and to be related to the practice of a healthier daily life by becoming self-conscious of having a definite objective of preventing MetS and practicing its measure

    長崎がんばらんば国体での昼食弁当献立開発の取り組み

    平成26年10月に長崎県で開催された国民体育大会秋季大会(長崎がんばらんば国体)では、各開催地で来県者をもてなすための様々な取り組みが行われた。その取り組みの一つとして、本学所在地である佐世保市でも、大会中、希望者に提供する昼食弁当を佐世保らしい内容でもてなしたい、と本学に献立開発の依頼があった。本学健康管理学部で献立開発に従事することは、教員のみならず学生の実践教育にも効果的であることから、健康管理学部の卒業研究として取り組むこととした。また、作成した献立が提供する弁当として実現可能かどうか具体的に検証するために、大会当日に弁当を作成する指定弁当調製業者も加わり、本学と佐世保市実行委員会の三者協働でワークショップや打合せ、試作を重ねながら開発に取り組んだ。その取り組んだ過程と完成した弁当の内容について報告する。During the Fall National Sports Festival that took place in Nagasaki Prefecture in October 2014, diverse approaches were implemented to appropriately accommodate visitors at each venue. As one of such approaches, Sasebo City, where our university is located, requested us to develop original recipes for lunch boxes to be offered during the event. Considering that such an activity might have positive effects on not only our faculty staff members, but also on practical education for our students at the Faculty of Health Management, we accepted their request, as the graduate study within the faculty. In addition to our faculty and Sasebo City, a designated lunch box catering service provider in charge of the delivery of lunch boxes on the day of the event also participated in workshops, discussions, and trial creation during the development process to confirm the applicability of the developed recipes. This paper reports the details of the process and describes the finished lunch boxes

    佐世保市民における食育の認知及び実践

    佐世保市では平成18年に「佐世保市食育推進計画」を策定し、食育に取り組んできた。本報告では平成18年からの5か年で佐世保市民の食育に関する知識や食生活がどのように変化したのかを平成22年9月に行った「食育に関するアンケート調査」の結果を基に検討し、今後の食育推進における資料を得ることを目的とした。食育に関する知識は平成18年と比較すると広く普及していたが、食生活の改善に結びついていない項目も多くみられた。また、食生活の改善には食育の取り組みを行うという意識を持つことが最も重要であることが示されたが、食育に関する知識がない者も正しい知識を得ることで、改善する可能性が示された。以上のことより、佐世保市においては今後も継続した食育の推進を図ることが必要であり、食生活の改善のためには、食育に取り組むことを目指すだけでなく、食育に関する正しい知識の普及が重要であることが示された。“Sasebo Food Education Promotion Plan”was designed in 2006 in Sasebo city and then incorporated into food education. Subsequently, after 5 years, a“questionnaire survey related to food education”was conducted in September 2010 in order to examine whether there was a change in the dietary habits and dietary knowledge of the residents of Sasebo city. The objective of the present report was to obtain a future data regarding the promotion of food education on the basis of the results of the questionnaire administered in 2010.The information about food education is spreading widely now, as compared to 2006. However, many instances not related to the improvement of dietary habits were found. Moreover, it was indicated that in order to improve dietary habits, one has to be aware of incorporating the food education plan in his or her life, but the right dietary knowledge should be provided to people who do not have any knowledge regarding this.Therefore, in future, a plan for continuous promotion of food education should be designed in Sasebo city, and it should not only be incorporated in food education, but the right information should be imparted in order to improve the dietary habits

    佐世保市の食育に関するアンケート調査報告(学齢期)

    近年の子どもたちを取り巻く社会環境においては、朝食欠食や偏食等にみられるような不規則な食生活など、様々な問題が提起されている1-4)。平成17年に食育基本法が公布され、佐世保市においても、平成18年度に「佐世保市食育推進計画」(平成18年度~平成22年度)6) を作成し、食育の推進に努めてきた。この食育推進計画における目標値の達成状況を把握するために「食育に関するアンケート調査」7) を行った。本報告においては、特に学齢期の子どもを対象とした調査結果および数値目標との比較を行った。その結果、計画策定時の状態よりも改善されている事項が多く、食育推進の取組の効果があった。ただし、目標値にまだ達していないことから、今後も継続した食育推進の取組が必要であり、それには、家庭や教育機関、社会など環境が関与する必要があることが示唆された。Children in recent years are irregular eating habits in breakfast skipping a meal or an unbalanced diet, and have become a social problem. The basic law on nutritional education was proclaimed in 2005, the Sasebo food education promotion plan was drawn up also in Sasebo in 2006, and it has striven for promotion of food education. In this food education promotion plan, the questionnaire about food education was performed to the original investigation, and the progress which is a plan was checked. Especially this report compared results of an investigation and a desired value for the school age child. As a result, there were many improving points as an effect of a food education promotion plan. However, since a desired value has not been reached yet, the continued food education promotion is required. And it was suggested to goal achievement that the influence of a home, educational facilities, and society is a theme

    長崎国際大学生における食生活の現状と骨密度の関係

    日本人はカルシウムの摂取量が慢性的に不足していることから、骨粗鬆症、骨軟化症、さらにくる病などの骨関連疾患が大きな問題となっている。一方で、その予防には食生活が重要性な要因であると認識されている。そこで今回、大学生の生活状況を調査するため、骨形成に関与する栄養素に注目し、長崎国際大学健康管理学部健康栄養学科で実施されている「健康栄養調査プロジェクト」の結果を解析した。男子学生のカルシウム(Ca)摂取量は、目標量(650mg/日)をほぼ満たす量(647±79mg/日)であったが、女子学生の Ca 摂取量(480±11mg/日)は目標量(600mg/日)の80%と不足していた。食品群別にみると、女子学生においてミネラル分が豊富な海藻類、小魚の摂取頻度が少なく、このことが女子学生でみられた Ca 摂取不足の主たる原因と考えられた。また、牛乳の摂取頻度が週7回以上の群は、7回未満の群に比べ骨密度の高い傾向がみられ、このことから若年期においても牛乳の摂取頻度が骨密度に影響している可能性が示唆された。Japanese people is chronically low level of calcium ingestion, therefore it is increased disorders related bone, osteoporosis, osteomalacia, rickets and other. On the other hand their prevention is closely related to dietary habit. In this study, to see about dietary habit on young people, we analyzed the result of "The Health and Nutritional Survey Project" conducted in Department of Health and Nutrition, Faculty of Health Management, Nagasaki International University about nutrient factor relative to bone formation. The calcium ingestion on male students (647±79mg/day) was close on tentative dietary goal for preventing life-style related diseases (DG) (650mg/day) by dietary reference intakes for Japanese [2005]. But calcium ingestion on female students (480±11mg/day) was lower, about 80% compared with DG (600mg). From the results of food group frequency, this low level of calcium ingestion on female students is likely to be caused by that frequency of sea vegetable and fingerling was few. And bone density evaluation was significantly high in group over 7 times/week compared with group below 7 times/week about frequency of milk. This result suggests that frequency of milk has an affect on bone density in early life

    学生の健康及び食生活意識と血液検査値の関係に対する一考察 - 健康栄養調査プロジェクト報告 第2報 -

    長崎国際大学健康管理学部健康栄養学科に所属する学生を対象に平成17年度に実施した「健康栄養プロジェクト」の結果のうち、学生(男子43名、女子285名)の食生活意識、身体状況および血液生化学検査(総コレステロール、トリアシルグリセロール、ヘモグロビン A1c、血清鉄)の関係について検討を加えた。その結果、欠食頻度が高い者ほど疲労愁訴を有する割合が高くなる傾向があった。また、欠食回数及び食生活意識と血液生化学検査値との関係に男女差が見られた。男子学生では欠食がある場合は検査値が低くなる傾向がみられたが、女子学生では欠食が無いにもかかわらず低値を示す者が多かった。このことから、今後食生活指導を行う際には、食事内容を具体的に把握し、適正な食事量を認識させる重要性が示唆された。This study, "The Health and Nutrition Survey Project" for the undergraduates (43 males and 285 females) who belong to the department of health and nutrition in Nagasaki International University, was examined to grasp their life style, physical condition, eating-habits consciousness and blood biochemical levels. We focused on their physical condition, eating-habits, especially skipping-a-meal and some blood biochemical levels (total cholesterol, triacylglycerol, hemoglobin A1c and serum iron). First of all, the complaint of physical fatigue is more on much skipping-a-meal undergraduates. Moreover, the relation the times of skipping a meal and some blood biochemical levels was different between male and female. In male undergraduates, triacylglycerol and hemoglobin A1c levels were lower on skipping a meal. In female undergraduates, although no skipping a meal, much low total cholesterol, triacylglycerol, hemoglobin A1c and serum iron levels had shown on. It was suggested we have to instruct the proper amount of meals in them, grasping not only the habit of skipping-a-meal, but also concretely their meal contents

    休養状況から見た健康に向けた実践状況 -佐世保市民の健康に関する実態調査報告-

    健康づくりの要素として「栄養・食生活」「身体活動・運動」「休養・こころの健康づくり」の3本柱があげられるが、本報告では佐世保市において平成24年1月~2月に行われた「佐世保市民の健康に関する実態調査」の調査結果のうち、休養状況を示す「休養・こころの健康づくり」に着目をし、休養と健康についての分析を行った。その結果、日常的に健康に配慮しているもの及び配慮している生活習慣として「栄養・食生活」を挙げた者が最も多く、「休養」をあげた者の割合は、「栄養・食生活」「運動」に対して少なかった。「運動」を挙げた者は休養がとれている者ととれていない者との差が大きく、また、休養がとれている者は、積極的に身体を動かしていたことを示した。休養を適切にとることは、日常生活が健康的な生活につながることを示唆した。しかし、休養が健康の3本柱でありながら、健康のために配慮することとして休養は考えられにくい状況であったことから、休養と健康とのつながりについて周知していくことが必要となると考える。It is thought that“nutrition and dietary life”,“physical activity and exercise”and“rest and mental health promotion”are the most important factors for health promotion. Previously, we made a survey on actual conditions of health for people living in Sasebo in a period from January to February of 2012. Based on results of the survey, an analysis was made on a rest and health paying attention to the results of“rest and mental health promotion”, which indicates the situation of people\u27s rest. The number of people usually paying attention to own health and those who chose the item“nutrition and dietary life”as their important life habit was the most. Whereas the number of subjects who chose the item“rest”was less than the number of subjects who chose the item“nutrition and dietary life”or“exercise”. Regarding the subjects who chose the item of exercise, there was a large difference between the numbers of subjects taking a rest enough and those who were not. Moreover, the present findings suggested that physical activity of people taking a rest enough is mostly high. These results suggest that to take an appropriate rest would lead to healthy life. “Rest”is one of three indispensable factors to keep health and so it is important to be concerned about“rest”for health maintenance. However, such idea was not so widely received until recently and“rest”was apt to be disregarded. Therefore, it is thought necessary to widely spread the idea about close correlation between rest and health
