Spatial GIS Database for Adaptive Grassland Management in Dudhwa National Park, India
Ecologically important tall grasslands of Dudhwa National Park (DNP) in Northern India are being threatened by various natural and biotic factors including varying grassland burning regimes viz. cut and burn, harrowed and burn, or only burn adopted by management for their maintenance (De 2001). Uncertainty regarding resultant grassland composition and disturbance to grassland obligate species prompted the management and scientific communities concerned to develop an adaptive management strategy for long term conservation goals (Chadden et al. 2004). In view of this, the present study attempted to develop comprehensive GIS (Geographic Information System) spatial database and associated hard copy maps of park at the scale of 1:25,000, to support field practitioners for successful implementation of proposed Adaptive Grassland Management
Traumatic Endophthalmitis due to Cellulosimicrobium cellulans
Purpose. To report a case of traumatic endophthalmitis due to Cellulosimicrobium cellulans. Design. Case report. Methods. Retrospective chart review. Results. To our knowledge, this is the first report of traumatic endophthalmitis due to C. cellulans, which did not respond to intravitreal antibiotics or repeat vitrectomy and ultimately led to the discovery of an occult intraocular foreign body. Conclusions. C. cellulans is a rare cause of endophthalmitis. Greater awareness of this bacterium in the ophthalmic literature as a cause of endophthalmitis and its association with foreign bodies may allow earlier and more purposeful intervention in future cases
Thyroid profile in newly diagnosed male HIV patients: a study from North Western part of India
Background: The aim of this study was to determine proportion of newly diagnosed male HIV cases with thyroid dysfunction at different levels of CD4 counts.Methods: 195 newly diagnosed male HIV patients attending medical OPD, ART centre and medical wards of SMS Medical College and Hospital, during a period of May 2012 to April 2013 were enrolled in the study. These patients were divided in three groups on the basis of CD4 cell counts. Group A: CD4 counts 500/mm3.Results: We concluded a negative correlation between the CD4 counts and serum TSH level (r = -0.382) which was significant (p-value <0.05). Overall 32 (16.41%) patients had increased TSH, 4 (2.05%) patients had decreased and 159 (81.53%) patients had normal TSH level. Plasma TSH values in group A were higher than group B and C and they were highly significant (p<.001). Mean plasma TSH values in patients of group A, B and C was 4.56±3.60 µIU/mL (range: 1.10-17.74), 2.20±1.02 µIU/mL (range:0.24-4.22) and 2.23±1.06 µIU/mL (range:0.28-4.25) respectively. (Reference normal value = 0.4-4.0 µIU/mL). There was significantly positive correlation (p-value < 0.01) found between the CD4 counts and serum free T4 levels (r = +0.378).Conclusions: This study has demonstrated a high prevalence of thyroid dysfunction in HIV infected patients of this part of country. High prevalence of thyroid dysfunction may contribute to the morbidity of the patients and have a bearing on quality of life of the HIV infected patients. T Oligo-Primed Polymerase Chain Reaction (TOP-PCR): A Robust Method for the Amplification of Minute DNA Fragments in Body Fluids.
Body fluid DNA sequencing is a powerful noninvasive approach for the diagnosis of genetic defects, infectious agents and diseases. The success relies on the quantity and quality of the DNA samples. However, numerous clinical samples are either at low quantity or of poor quality due to various reasons. To overcome these problems, we have developed T oligo-primed polymerase chain reaction (TOP-PCR) for full-length nonselective amplification of minute quantity of DNA fragments. TOP-PCR adopts homogeneous "half adaptor" (HA), generated by annealing P oligo (carrying a phosphate group at the 5' end) and T oligo (carrying a T-tail at the 3' end), for efficient ligation to target DNA and subsequent PCR amplification primed by the T oligo alone. Using DNA samples from body fluids, we demonstrate that TOP-PCR recovers minute DNA fragments and maintains the DNA size profile, while enhancing the major molecular populations. Our results also showed that TOP-PCR is a superior method for detecting apoptosis and outperforms the method adopted by Illumina for DNA amplification
Biowaiver monographs for immediate release solid oral dosage forms: prednisolone.
Literature data relevant to the decision to allow a waiver of in vivo bioequivalence (BE) testing for the approval of immediate release (IR) solid oral dosage forms containing prednisolone are reviewed. Data on its solubility, oral absorption, and permeability are not totally conclusive, but strongly suggest a BCS Class 1 classification. Prednisolone's therapeutic indications and therapeutic index, pharmacokinetics, and the possibility of excipient interactions were also taken into consideration. Available evidence indicates that a biowaiver for IR solid oral dosage forms formulated with the excipients tabulated in this article would be unlikely to expose patients to undue risks
Biowaiver monographs for immediate release solid oral dosage forms: acetaminophen (paracetamol).
Literature data are reviewed on the properties of acetaminophen (paracetamol) related to the biopharmaceutics classification system (BCS). According to the current BCS criteria, acetaminophen is BCS Class III compound. Differences in composition seldom, if ever, have an effect on the extent of absorption. However, some studies show differences in rate of absorption between brands and formulations. In particular, sodium bicarbonate, present in some drug products, was reported to give an increase in the rate of absorption, probably caused by an effect on gastric emptying. In view of Marketing Authorizations (MAs) given in a number of countries to acetaminophen drug products with rapid onset of action, it is concluded that differences in rate of absorption were considered therapeutically not relevant by the Health Authorities. Moreover, in view of its therapeutic use, its wide therapeutic index and its uncomplicated pharmacokinetic properties, in vitro dissolution data collected according to the relevant Guidances can be safely used for declaring bioequivalence (BE) of two acetaminophen formulations. Therefore, accepting a biowaiver for immediate release (IR) acetaminophen solid oral drug products is considered scientifically justified, if the test product contains only those excipients reported in this paper in their usual amounts and the test product is rapidly dissolving, as well as the test product fulfils the criterion of similarity of dissolution profiles to the reference product
Lectin-mediated bacterial modulation by the intestinal nematode Ascaris suum
Ascariasis is a global health problem for humans and animals. Adult Ascaris nematodes are long-lived in the host intestine where they interact with host cells as well as members of the microbiota resulting in chronic infections. Nematode interactions with host cells and the microbial environment are prominently mediated by parasite-secreted proteins and peptides possessing immunomodulatory and antimicrobial activities. Previously, we discovered the C-type lectin protein AsCTL-42 in the secreted products of adult Ascaris worms. Here we tested recombinant AsCTL-42 for its ability to interact with bacterial and host cells. We found that AsCTL-42 lacks bactericidal activity but neutralized bacterial cells without killing them. Treatment of bacterial cells with AsCTL-42 reduced invasion of intestinal epithelial cells by Salmonella. Furthermore, AsCTL-42 interacted with host myeloid C-type lectin receptors. DIALib-QC an assessment tool for spectral libraries in data-independent acquisition proteomics.
Data-independent acquisition (DIA) mass spectrometry, also known as Sequential Window Acquisition of all Theoretical Mass Spectra (SWATH), is a popular label-free proteomics strategy to comprehensively quantify peptides/proteins utilizing mass spectral libraries to decipher inherently multiplexed spectra collected linearly across a mass range. Although there are many spectral libraries produced worldwide, the quality control of these libraries is lacking. We present the DIALib-QC (DIA library quality control) software tool for the systematic evaluation of a library\u27s characteristics, completeness and correctness across 62 parameters of compliance, and further provide the option to improve its quality. We demonstrate its utility in assessing and repairing spectral libraries for correctness, accuracy and sensitivity
