    The planning of biological isolation measures for the Mars Surface Sample Return Mission is discussed in terms of personnel and organizational management. Deficiencies in past operation of the Lunar Receiving Laborator are analyzed. It was found that the failure to clearly define relationship among the government agencies involved and to effectively integrate their objectives and responsibilities was a major cause of Laboratory deficiencies. Possible solutions to these problems are presented for application to future missions

    Collective response of DNA to THz electric fields is studied in a simple pair bond model. We confirm, with some caveats, a previous observation of destabilising DNA breather modes and explore the parameter-dependence of these modes.     All too often lawyers and students of law are inclined to assume that some principle or formula· has become so deeply rooted in the law that it has acquired a sanctity which gives it an all-embracive effect, a cure-all, as it were, for legal ailments. This has been the usual approach when the question has arisen of the propriety of spending corporate funds to solicit proxies. The ever-faithful panacea has been to say, quite automatically, that where the intra-corporate contest is concerned with matters of policy as distinguished from personnel of management, then corporate funds may be used to inform the stockholders of the policy issues and to secure their support of the views advocated by the management. While there has been an occasional case which would seem to reject this approach, the policy formula as a justification for corporate expenditures has enjoyed some popularity. A statement of the Delaware court indicates that the formula may have even grown beyond the point of being a judicial cure accepted primarily because of its supposed virtues. The Delaware court has said, Moreover, for good or evil, the incumbent board of directors of a Delaware corporation may look to the corporation for payment of expenses incurred by them in soliciting proxies where a question of policy is involved

    This study had two principal goals. The first was to take a broad look at policing in Indian Country in order to better understand the many arrangements for administering reservation police departments, develop an initial assessment of the challenges facing Indian policing, and identify policing strategies and approaches that might be successful in responding to the growing crime problem in Indian Country. The second was to evaluate the prospects for community policing in Indian Country. Could this strategy, which grew out of the experience of police departments in urban settings, be usefully applied to the strikingly different cultural, geographic, and demographic features typical of Indian reservations? This study is a first effort to characterize the variety of arrangements for reservation policing combined with a more comprehensive effort to better understand the operations of a limited set of representative departments and their tribal contexts

    Mitigating sampling error when measuring internet client IPv6 capabilities

    Despite the predicted exhaustion of unallocated IPv4 addresses be- tween 2012 and 2014, it remains unclear how many current clients can use its successor, IPv6, to access the Internet. We propose a refinement of previous measurement studies that mitigates intrin- sic measurement biases, and demonstrate a novel web-based tech- nique using Google ads to perform IPv6 capability testing on a wider range of clients. After applying our sampling error reduction, we find that 6% of world-wide connections are from IPv6-capable clients, but only 1–2% of connections preferred IPv6 in dual-stack (dual-stack failure rates less than 1%). Except for an uptick around IPv6-day 2011 these proportions were relatively constant, while the percentage of connections with IPv6-capable DNS resolvers has in- creased to nearly 60%.     Further Analyses of the Safety of Verubecestat in the Phase 3 EPOCH Trial of Mild-To-Moderate Alzheimer’s Disease

    Background: Verubecestat, a BACE1 inhibitor that reduces Aβ levels in the cerebrospinal fluid of humans, was not effective in a phase 3 trial (EPOCH) of mild-to-moderate AD and was associated with adverse events. To assist in the development of BACE1 inhibitors, we report detailed safety findings from EPOCH. Methods: EPOCH was a randomized, double-blind, placebo-controlled 78-week trial evaluating verubecestat 12 mg and 40 mg in participants with mild-to-moderate AD diagnosed clinically. The trial was terminated due to futility close to its scheduled completion. Of 1957 participants who were randomized and took treatment, 652 were assigned to verubecestat 12 mg, 652 to verubecestat 40 mg, and 653 to placebo. Adverse events and relevant laboratory, vital sign, and ECG findings were assessed. Results: Verubecestat 12 mg and 40 mg were associated with an increase in the percentage of participants reporting adverse events versus placebo (89 and 92% vs. 82%), although relatively few participants discontinued treatment due to adverse events (8 and 9% vs. 6%). Adverse events that were increased versus placebo included falls and injuries, suicidal ideation, weight loss, sleep disturbance, rash, and hair color change. Most were mild to moderate in severity. Treatment differences in suicidal ideation emerged within the first 3 months but did not appear to increase after 6 months. In contrast, treatment differences in falls and injuries continued to increase over time. Conclusions: Verubecestat was associated with increased risk for several types of adverse events. Falls and injuries were notable for progressive increases over time. While the mechanisms underlying the increased adverse events are unclear, they may be due to BACE inhibition and should be considered in future clinical development programs of BACE1 inhibitors

    Clinical Efficacy of TroVax in the Treatment of Progressive Castration-resistant Prostate Cancer

    With approximately 240,890 new cases expected in 2011, prostate cancer remains the leading cause of non-melanoma cancer deaths in men. Immunotherapies using viral vector-based delivery systems targeting tumor-specific antigens are being studied. Viral vector-based delivery systems present tumor-targeted antigens (TAAs) to the immune system while breaking self-tolerance. Modified vaccinia ankara has been combined with the oncofetal antigen 5T4 to create TroVax for the treatment of castration-resistant prostate cancer (CRPC). The 5T4 antigen is highly expressed in a large number of carcinomas, including prostate cancer, but is rarely expressed in healthy tissue. TroVax has been demonstrated to be safe and highly immunogenic, both as monotherapy and in combination with other standard of care therapies in colorectal, renal cell, and prostate cancer. With minimal side effects and the ability to produce a strong immunogenic response, TroVax (MVA-5T4) is a viable addition to the treatment of prostate cancer
