    Serum peptide reactivities may distinguish neuromyelitis optica subgroups and multiple sclerosis

    Objective: To assess in an observational study whether serum peptide antibody reactivities may distinguish aquaporin-4 (AQP4) antibody (Ab)–positive and -negative neuromyelitis optica spectrum disorders (NMOSD) and relapsing-remitting multiple sclerosis (RRMS). Methods: We screened 8,700 peptides that included human and viral antigens of potential relevance for inflammatory demyelinating diseases and random peptides with pooled sera from different patient groups and healthy controls to set up a customized microarray with 700 peptides. With this microarray, we tested sera from 66 patients with AQP4-Ab-positive (n = 16) and AQP4-Ab-negative (n = 19) NMOSD, RRMS (n = 11), and healthy controls (n = 20). Results: Differential peptide reactivities distinguished NMOSD subgroups from RRMS in 80% of patients. However, the 2 NMOSD subgroups were not well-discriminated, although those patients are clearly separated by their antibody reactivities against AQP4 in cell-based assays. Elevated reactivities to myelin and Epstein-Barr virus peptides were present in RRMS and to AQP4 and AQP1 peptides in AQP4-Ab-positive NMOSD. Conclusions: While AQP4-Ab-positive and -negative NMOSD subgroups are not well-discriminated by peptide antibody reactivities, our findings suggest that peptide antibody reactivities may have the potential to distinguish between both NMOSD subgroups and MS. Future studies should thus concentrate on evaluating peptide antibody reactivities for the differentiation of AQP4-Ab-negative NMOSD and MS

    Credit bureaus between risk-management, creditworthiness assessment and prudential supervision

If cited or quoted, reference should be made to the full name of the author, the title, the working paper or other series, the year, and the publisher."This paper discusses the role and operations of consumer Credit Bureaus in the European Union in the context of the economic theories, policies and law within which they work. Across Europe there is no common practice of sharing the credit data of consumers which can be used for several purposes. Mostly, they are used by the lending industry as a practice of creditworthiness assessment or as a risk-management tool to underwrite borrowing decisions or price risk. However, the type, breath, and depth of information differ greatly from country to country. In some Member States, consumer data are part of a broader information centralisation system for the prudential supervision of banks and the financial system as a whole. Despite EU rules on credit to consumers for the creation of the internal market, the underlying consumer data infrastructure remains fragmented at national level, failing to achieve univocal, common, or defined policy objectives under a harmonised legal framework. Likewise, the establishment of the Banking Union and the prudential supervision of the Euro area demand standardisation and convergence of the data used to measure debt levels, arrears, and delinquencies. The many functions and usages of credit data suggest that the policy goals to be achieved should inform the legal and institutional framework of Credit Bureaus, as well as the design and use of the databases. This is also because fundamental rights and consumer protection concerns arise from the sharing of credit data and their expanding use

    We study the impact of wall corrugations in microchannels on the process of capillary filling by means of three broadly used methods - Computational Fluid Dynamics (CFD), Lattice-Boltzmann Equations (LBE) and Molecular Dynamics (MD). The numerical results of these approaches are compared and tested against the Concus-Finn (CF) criterion, which predicts pinning of the contact line at rectangular ridges perpendicular to flow for contact angles theta > 45. While for theta = 30, theta = 40 (no flow) and theta = 60 (flow) all methods are found to produce data consistent with the CF criterion, at theta = 50 the numerical experiments provide different results. Whilst pinning of the liquid front is observed both in the LB and CFD simulations, MD simulations show that molecular fluctuations allow front propagation even above the critical value predicted by the deterministic CF criterion, thereby introducing a sensitivity to the obstacle heigth.Comment: 25 pages, 8 figures, Langmuir in pres

    O controle de plantas daninhas Ă© de suma importĂąncia na cultura do milho, devido aos prejuĂ­zos que podem causar na produtividade de grĂŁos. Uma das alternativas de manejo Ă© a utilização de adubos verdes como a crotalĂĄria, que pode resultar em menor infestação de plantas daninhas devido a ao efeito fĂ­sico de cobertura do solo e/ou liberação de substĂąncias alelopĂĄticas. Assim, objetivou-se com este trabalho identificar e quantificar a incidĂȘncia de plantas daninhas no consĂłrcio de milho segunda safra com Crotalaria spectabilis e Crotalaria ochroleuca. O experimento foi instalado na ĂĄrea experimental da Embrapa Agrossilvipastoril, em Sinop-MT. O delineamento foi em blocos ao acasocasualizados, com quatro repetiçÔes. Os tratamentos foram constituĂ­dos por quatro densidades (10, 20, 30 e 40 kg de sementes m-2) de C. spectabilis e quatro densidades (5, 10, 15, 20 kg de sementes m-2) de C. ochroleuca, cultivares comum, com 80% de germinação, consorciadas com o milho, alĂ©m da testemunha, com milho solteiro. As espĂ©cies de crotalĂĄria foram semeadas a lanço, simulando a utilização de semeadora com caixa adicional para sementes miĂșdas, em semeadura simultĂąnea. As parcelas foram constituĂ­das por 11 linhas de dez metros, espaçadas de 0,45 m entre si, considerando-se como ĂĄrea Ăștil trĂȘs linhas centrais. O milho hib. DKB 175VTPRO2 foi semeado no dia 15 de fevereiro de 2017. Para a avaliação que ocorreu em prĂ©-colheita, utilizou-se o mĂ©todo do quadrado inventĂĄrio (0,25 x 0,25 m), lançado seis vezes ao acaso em cada parcela. Em cada quadro foram identificadas e quantificadas as espĂ©cies de plantas daninhas para o cĂĄlculo de Ă­ndice de importĂąncia relativa (IR), para assim identificar as espĂ©cies mais importantes em cada tratamento. Na anĂĄlise fitossociolĂłgica foram encontradas 16 espĂ©cies de plantas daninhas no experimento. No milho solteiro e nos tratamentos de milho + C. spectabilis, observou-se que Chamaesyce hirta foi a espĂ©cie mais importante (IR de 40,7% a 52,8%). JĂĄ no consĂłrcio de milho + C. ochroleuca, apenas na densidade de 5 kg ha-1 essa espĂ©cie foi a mais importante, enquanto nas demais densidades, Cyperus spp. apresentou a mesma importĂąncia de C. hirta (densidade de 15 kg ha-1) ou foi a espĂ©cie mais importante (IR de 49,2 a 53%). Concluiu-se que densidades de C. ochroleuca a partir de 10 kg ha-1 alteram a estrutura da comunidade de plantas daninhas em relação ao milho solteiro, nĂŁo sendo observado o mesmo efeito em relação ao consĂłrcio de milho + C. spectabilis

    Real and Virtual Compton Scattering off the Nucleon

    A review is given of the very recent developments in the fields of real and virtual Compton scattering off the nucleon. Both real and virtual Compton scattering reactions are discussed at low outgoing photon energy where one accesses polarizabilities of the nucleon. The real Compton scattering at large momentum transfer is discussed which is asymptotically a tool to obtain information on the valence quark wave function of the nucleon. The rapid developments in deeply virtual Compton scattering and associated meson electroproduction reactions at high energy, high photon virtuality and small momentum transfer to the nucleon are discussed. A unified theoretical description of those processes has emerged over the last few years, which gives access to new, generalized parton distributions.     Hydrogen storage systems from waste Mg alloys

    The production cost of materials for hydrogen storage is one of the major issues to be addressed in order to consider them suitable for large scale applications. In the last decades several authors reported on the hydrogen sorption properties of Mg and Mg-based systems. In this work magnesium industrial wastes of AZ91 alloy and Mg-10 wt.% Gd alloy are used for the production of hydrogen storage materials. The hydrogen sorption properties of the alloys were investigated by means of volumetric technique, in situ synchrotron radiation powder X-ray diffraction (SR-PXD) and calorimetric methods. The measured reversible hydrogen storage capacity for the alloys AZ91 and Mg-10 wt.% Gd are 4.2 and 5.8 wt.%, respectively. For the Mg-10 wt.% Gd alloy, the hydrogenated product was also successfully used as starting reactant for the synthesis of Mg(NH2)2 and as MgH2 substitute in the Reactive Hydride Composite (RHC) 2LiBH4 + MgH 2. The results of this work demonstrate the concrete possibility to use Mg alloy wastes for hydrogen storage purposes

    Deleterious variants in TRAK1 disrupt mitochondrial movement and cause fatal encephalopathy

In neurons, mitochondrial trafficking is crucial because of high energy needs and calcium ion buffering along axons to synapses during neurotransmission. The trafficking kinesin proteins (TRAKs) are well characterized for their role in lysosomal and mitochondrial trafficking in cells, especially neurons. Using whole exome sequencing, we identified homozygous truncating variants in TRAK1 (NM_001042646:c.287-2A > C), in six lethal encephalopathic patients from three unrelated families. The pathogenic variant results in aberrant splicing and significantly reduced gene expression at the RNA and protein levels. In comparison with normal cells, TRAK1-deficient fibroblasts showed irregular mitochondrial distribution, altered mitochondrial motility, reduced mitochondrial membrane potential, and diminished mitochondrial respiration. This study confirms the role of TRAK1 in mitochondrial dynamics and constitutes the first report of this gene in association with a severe neurodevelopmental disorder.D.M.E. and J.K. are supported by the Office of Naval Research (ONR) Grant N000141410538.     SLIM Ultrahigh Resolution Ion Mobility Spectrometry Separations of Isotopologues and Isotopomers Reveal Mobility Shifts due to Mass Distribution Changes

    We report on separations of ion isotopologues and isotopomers using ultrahigh-resolution traveling wave-based Structures for Lossless Ion Manipulations with serpentine ultralong path and extended routing ion mobility spectrometry coupled to mass spectrometry (SLIM SUPER IMS-MS). Mobility separations of ions from the naturally occurring ion isotopic envelopes (e.g., [M], [M+1], [M+2], ... ions) showed the first and second isotopic peaks (i.e., [M+1] and [M+2]) for various tetraalkylammonium ions could be resolved from their respective monoisotopic ion peak ([M]) after SLIM SUPER IMS with resolving powers of ∌400–600. Similar separations were obtained for other compounds (e.g., tetrapeptide ions). Greater separation was obtained using argon versus helium drift gas, as expected from the greater reduced mass contribution to ion mobility described by the Mason–Schamp relationship. To more directly explore the role of isotopic substitutions, we studied a mixture of specific isotopically substituted (15N, 13C, and 2H) protonated arginine isotopologues. While the separations in nitrogen were primarily due to their reduced mass differences, similar to the naturally occurring isotopologues, their separations in helium, where higher resolving powers could also be achieved, revealed distinct additional relative mobility shifts. These shifts appeared correlated, after correction for the reduced mass contribution, with changes in the ion center of mass due to the different locations of heavy atom substitutions. The origin of these apparent mass distribution-induced mobility shifts was then further explored using a mixture of Iodoacetyl Tandem Mass Tag (iodoTMT) isotopomers (i.e., each having the same exact mass, but with different isotopic substitution sites). Again, the observed mobility shifts appeared correlated with changes in the ion center of mass leading to multiple monoisotopic mobilities being observed for some isotopomers (up to a ∌0.04% difference in mobility). These mobility shifts thus appear to reflect details of the ion structure, derived from the changes due to ion rotation impacting collision frequency or momentum transfer, and highlight the potential for new approaches for ion structural characterization
