    Convalescent plasma in patients admitted to hospital with COVID-19 (RECOVERY): a randomised controlled, open-label, platform trial

    SummaryBackground Azithromycin has been proposed as a treatment for COVID-19 on the basis of its immunomodulatoryactions. We aimed to evaluate the safety and efficacy of azithromycin in patients admitted to hospital with COVID-19.Methods In this randomised, controlled, open-label, adaptive platform trial (Randomised Evaluation of COVID-19Therapy [RECOVERY]), several possible treatments were compared with usual care in patients admitted to hospitalwith COVID-19 in the UK. The trial is underway at 176 hospitals in the UK. Eligible and consenting patients wererandomly allocated to either usual standard of care alone or usual standard of care plus azithromycin 500 mg once perday by mouth or intravenously for 10 days or until discharge (or allocation to one of the other RECOVERY treatmentgroups). Patients were assigned via web-based simple (unstratified) randomisation with allocation concealment andwere twice as likely to be randomly assigned to usual care than to any of the active treatment groups. Participants andlocal study staff were not masked to the allocated treatment, but all others involved in the trial were masked to theoutcome data during the trial. The primary outcome was 28-day all-cause mortality, assessed in the intention-to-treatpopulation. The trial is registered with ISRCTN, 50189673, and ClinicalTrials.gov, NCT04381936.Findings Between April 7 and Nov 27, 2020, of 16 442 patients enrolled in the RECOVERY trial, 9433 (57%) wereeligible and 7763 were included in the assessment of azithromycin. The mean age of these study participants was65·3 years (SD 15·7) and approximately a third were women (2944 [38%] of 7763). 2582 patients were randomlyallocated to receive azithromycin and 5181 patients were randomly allocated to usual care alone. Overall,561 (22%) patients allocated to azithromycin and 1162 (22%) patients allocated to usual care died within 28 days(rate ratio 0·97, 95% CI 0·87–1·07; p=0·50). No significant difference was seen in duration of hospital stay (median10 days [IQR 5 to >28] vs 11 days [5 to >28]) or the proportion of patients discharged from hospital alive within 28 days(rate ratio 1·04, 95% CI 0·98–1·10; p=0·19). Among those not on invasive mechanical ventilation at baseline, nosignificant difference was seen in the proportion meeting the composite endpoint of invasive mechanical ventilationor death (risk ratio 0·95, 95% CI 0·87–1·03; p=0·24).Interpretation In patients admitted to hospital with COVID-19, azithromycin did not improve survival or otherprespecified clinical outcomes. Azithromycin use in patients admitted to hospital with COVID-19 should be restrictedto patients in whom there is a clear antimicrobial indication

    Fungal social influencers: secondary metabolites as a platform for shaping the plant-associated community

    Fungal secondary metabolites (FSMs) are capable of manipulating plant community dynamics by inhibiting or facilitating the establishment of co-habitating organisms. Although production of FSMs is not crucial for survival of the producer, their absence can indirectly impair growth and/or niche competition of these fungi on the plant. The presence of FSMs with no obvious consequence on the fitness of the producer leaves questions regarding ecological impact. This review investigates how fungi employ FSMs as a platform to mediate fungal-fungal, fungal-bacterial and fungal-animal interactions associated with the plant community. We discuss how the biological function of FSMs may indirectly benefit the producer by altering the dynamics of surrounding organisms. We introduce several instances where FSMs influence antagonistic- or alliance-driven interactions. Part of our aim is to decipher the meaning of the FSM 'language' as it is widely noted to impact the surrounding community. Here, we highlight the contribution of FSMs to plant-associated interaction networks that affect the host either broadly or in ways that may have previously been unclear

However, the minority of microbes that are pathogens can severely impact crop quality and yield, thereby endangering food security. By contrast, beneficial microbes provide plants with important services, such as enhanced nutrient uptake and protection against pests and diseases. Like pathogens, beneficial microbes can modulate host immunity to efficiently colonize the nutrient-rich niches within and around the roots and aerial tissues of a plant, a phenomenon mirroring the establishment of commensal microbes in the human gut. Numerous ingenious mechanisms have been described by which pathogenic and beneficial microbes in the plant microbiome communicate with their host, including the delivery of immune-suppressive effector proteins and the production of phytohormones, toxins and other bioactive molecules. Plants signal to their associated microbes via exudation of photosynthetically fixed carbon sources, quorum-sensing mimicry molecules and selective secondary metabolites such as strigolactones and flavonoids. Molecular communication thus forms an integral part of the establishment of both beneficial and pathogenic plant–microbe relations. Here, we review the current knowledge on microbe-derived small molecules that can act as signalling compounds to stimulate plant growth and health by beneficial microbes on the one hand, but also as weapons for plant invasion by pathogens on the other. As an exemplary case, we used comparative genomics to assess the small molecule biosynthetic capabilities of the Pseudomonas genus; a genus rich in both plant pathogenic and beneficial microbes. We highlight the biosynthetic potential of individual microbial genomes and the population at large, providing evidence for the hypothesis that the distinction between detrimental and beneficial microbes is increasingly fading. Knowledge on the biosynthesis and molecular activity of microbial small molecules will aid in the development of successful biological agents boosting crop resiliency in a sustainable manner and could also provide scientific routes to pathogen inhibition or eradication

    Fungi represent an ecologically diverse group of microorganisms that includes plant pathogenic species able to cause considerable yield loses in crop production systems worldwide. In order to establish compatible interactions with their hosts, pathogenic fungi rely on the secretion of molecules of diverse nature during host colonization to modulate host physiology, manipulate other environmental factors or provide self-defence. These molecules, collectively known as effectors, are typically small secreted cysteine-rich proteins, but may also comprise secondary metabolites and sRNAs.     Although jasmonic acid (JA) and JA derivatives are known to activate plant defense mechanisms and provide protection against postharvest fungal diseases for several horticultural crops, JA's ability to protect sugarbeet (Beta vulgaris L.) roots against common causal organisms of storage rot is unknown. To determine the potential of JA to reduce rot due to three common sugarbeet storage pathogens, harvested roots were treated with JA concentrations of 0.01, 0.1, 1, 10, or 100 μM, inoculated with Botrytis cinerea, Penicillium claviforme, or Phoma betae, and evaluated for the severity of rot symptoms after incubation at 20 °C and 90% relative humidity. JA concentrations of 0.01–100 μM significantly reduced rot due to all three pathogens. All concentrations of JA provided statistically equivalent control against B. cinerea and P. betae, and reduced the amount of rotted tissue due to these pathogens by an average of 51 and 71%, respectively. Against P. claviforme, JA concentrations of 0.01–10 μM were equally effective and reduced rot by an average of 44%, while an increase in JA concentration to 100 μM reduced rot by 65%. Against all three pathogens, JA treatment did not affect the incidence of infection, but reduced rot by reducing the progression of disease symptoms in root storage tissue

    De novo genome assembly of Cercospora beticola for microsatellite marker development and validation

    Cercospora leaf spot caused by Cercospora beticola is a significant threat to the production of sugar and table beet worldwide. A de novo genome assembly of C. beticola was used to develop eight polymorphic and reproducible microsatellite markers for population genetic analyses. These markers were used, along with five previously described microsatellite loci to genotype two C. beticola populations from table beet fields in New York, USA. High allelic and genotypic diversity and low population differentiation were found between fields. Linkage disequilibrium of loci after clone-correction of datasets was attributed to the presence of two distinct clonal lineages within the populations. Linkage equilibrium of loci in one of the clusters supported the presence of sexual reproduction. The draft de novo genome assembly will help elucidate the reproductive system of C. beticola through investigating evidence of recombination in the C. beticola genome

    Postharvest salicylic acid treatment reduces storage rots in water-stressed but not unstressed sugarbeet roots

    Exogenous application of salicylic acid (SA) reduces storage rots in a number of postharvest crops. SA's ability to protect sugarbeet (Beta vulgaris L.) taproots from common storage rot pathogens, however, is unknown. To determine the potential of SA to reduce storage losses caused by three common causal organisms of sugarbeet storage rot, freshly harvested roots were treated with 0.01, 0.1, 1.0 or 10 mM SA, inoculated with Botrytis cinerea, Penicillium claviforme, or Phoma betae, and evaluated for the severity of rot symptoms after incubation at 20 °C and 90% relative humidity. Roots were obtained from plants that received sufficient water or were water-stressed prior to harvest. Roots from water-stressed plants were included since water-stress increases sugarbeet root susceptibility to storage rot and SA mitigates drought effects in other plant species. SA at concentrations of 0.01–10 mM had no effect on the severity of storage rot caused by B. cinerea, P. claviforme, or P. betae in roots from plants that received sufficient water prior to harvest. However, SA at these same concentrations reduced the severity of rot symptoms for all three pathogens in roots from plants that were water stressed before harvest. For water-stressed roots, all concentrations of SA produced statistically equivalent reductions in the weight of rotted tissue for each pathogen, and on average, SA reduced rot severity due to B. cinerea, P. claviforme, and P. betae by 54, 45, and 58%, respectively. SA reduced rot from all three pathogens by reducing lesion size, but did not affect the incidence of infection. The ability of SA to reduce rot severity in water-stressed roots, but not in roots that received sufficient water before harvest suggests that SA alleviated the negative impact of water stress but did not directly protect sugarbeet roots against storage rots
