    Survival of lichens and bacteria exposed to outer space conditions - Results of the Lithopanspermia experiments

    n the space experiments Lithopanspermia, experimental support was provided to the likelihood of the lithopanspermia concept that considers a viable transport of microorganisms between the terrestrial planets by means of meteorites. The rock colonising lichens Rhizocarpon geographicum and Xanthoria elegans, the vagrant lichen Aspicilia fruticulosa, and endolithic and endoevaporitic communities of cyanobacteria and bacteria with their natural rock substrate were exposed to space for 10 days onboard the Biopan facility of the European Space Agency (ESA). Biopan was closed during launch and re-entry. In addition, in the Stone facility, one sample of R. geographicum on its natural granitic substrate was attached at the outer surface of the re-entry capsule close to the stagnation point, only protected by a thin cover of glass textolite. Post-flight analysis, which included determination of the photosynthetic activity, LIVE/DEAD staining, and germination capacity of the ascospores, demonstrated that all three lichen were quite resistant to outer space conditions, which include the full spectrum of solar extraterrestrial electromagnetic radiation or selected wavelength ranges. This high resistance of the lichens to space appears to be due to their symbiotic nature and protection by their upper pigmented layer, the cortex. In contrast, the rock- or halite-inhabiting bacteria were severely damaged by the same exposure. After atmospheric re-entry, the granite of the Stone sample was transformed into a glassy, nearly homogenous material, with several friction striae. None of the lichen cells survived this re-entry process. The data suggest that lichens are suitable candidates for testing the concept of lithopanspermia, because they are extremely resistant to the harsh environment of outer space. The more critical event is the atmospheric re-entry after being captured by a planet. Experiments simulating the re-entry process of a microbe-carrying meteoroid did not show any survivors

    Fostering English-taught higher education programs in a Spanish university: the "TechEnglish" innovative project

    In recent years, coinciding with adjustments to the Bologna process, many European universities have attempted to improve their international profile by increasing course offerings in English. According to the Institute of International Education (IIE), Spain has notably increased its English-taught higher education programs, ranking fifth in the list of European countries by number of English-taught Master's programs in 2013. This article presents the goals and preliminary results of an on-going innovative education project (TechEnglish) that aims to promote course offerings in English at the Technical University of Madrid (Universidad Politécnica de Madrid, UPM). The UPM is the oldest and largest of all Technical Universities in Spain. It offers graduate and postgraduate programs that cover all the engineering disciplines as well as architecture. Currently, the UPM has no specific bilingual/multilingual program to promote teaching in English, although there is an Educational Model Whitepaper (with a focus on undergraduate degrees) that promotes the development of activities like an International Semester or a unique shared curriculum. The TechEnglish project is an attempt to foster courses taught in English at 7 UPM Technical Schools, including students and 80 faculty members. Four tasks were identified: (1) to design a university wide framework to increase course offerings, (2) to identify administrative difficulties, (3) to increase visibility of courses offered, and (4) to disseminate the results of the project. First, to design a program we analyzed existing programs at other Spanish universities, and other projects and efforts already under way at the UPM. A total of 13 plans were analyzed and classified according to their relation with students (learning), professors (teaching), administration, course offerings, other actors/institutions within the university (e.g., language departments), funds and projects, dissemination activities, mobility plans and quality control. Second, to begin to identify administrative and organizational difficulties in the implementation of teaching in English, we first estimated the current and potential course offerings at the undergraduate level at the UPM using a survey (student, teacher and administrative demand, level of English and willingness to work in English). Third, to make the course offerings more attractive for both Spanish and international students we examined the way the most prestigious universities in Spain and in Europe try to improve the visibility of their academic offerings in English. Finally, to disseminate the results of the project we created a web page and a workspace on the Moodle education platform and prepared conferences and workshops within the UPM. Preliminary results show that increasing course offerings in English is an important step to promote the internationalization of the University. The main difficulties identified at the UPM were related to how to acknowledge/certify the departments, teachers or students involved in English courses, how students should register for the courses, how departments should split and schedule the courses (Spanish and English), and the lack of qualified personnel. A concerted effort could be made to increase the visibility of English-taught programs offered on-line

    Modelado y evaluación de prestaciones de un juego multimedia basado en el paradigma de Televisión Social

    La implantación de los juegos interactivos basados en el paradigma de la televisión social (Social TV) provocará en los próximos años un notable incremento del consumo de recursos en las redes de comunicaciones. En particular generará un aumento en el ancho de banda de los canales de subida de las redes de acceso, lo que dará lugar a nuevos patrones de utilización. Por ello, en este artículo se pretenden evaluar las pautas de consumo de recursos de estos juegos ante un posible despliegue masivo. Como caso de estudio se ha diseñado un juego de preguntas-respuestas por turnos, con contenidos multimedia de alta calidad, en el que cada jugador tiene comunicación bidireccional de audio y vídeo con el resto de jugadores. Para llevar a cabo el análisis, se ha monitorizado el tráfico de la aplicación y se ha caracterizado estadísticamente. Basándose en esta información se ha diseñado de un modelo de la aplicación, que se ha validado y desplegado sobre un escenario de red de comunicaciones diseñado para estas pruebas. Los resultados muestran el alto consumo de recursos que produce este tipo de aplicaciones interactivas

    Immunoelectron Microscopy of RNA Combined with Nucleic Acid Cytochemistry in Plant Nucleoli

    The immunoelectron microscopy detection of RNA using anti-RNA monoclonal antibodies has been performed for the first time over different plant cells. The use of the methylation-acetylation (MA) method permits clear distinction among the nuclear and nucleolar compartments and can be combined with the immunogold approach. Cytochemical methods for nucleic acids were performed together with the immunoassays, providing additional data about the different composition of the various nucleolar components. Anti-RNA antibodies highly labeled the ribosome-rich areas of the cytoplasm and the nucleolus. The interchromatin region also is labeled. The labeling was intense in the granular component, lower in the dense fibrillar component, and very scarce in the fibrillar centers. The MA method made possible the statistical evaluation of the labeling density in the various nuclear compartments by permitting the clear assignment of the particles to precise nuclear structures.This work was supported by project DIGICYT 87/92 PB033201 and PB92-0079-C03-01.Peer reviewe

    Dynamics of ribosome biogenesis during the pollen developmental program by in situ localization of nucleolar targets

    The nucleolus, is a highly dynamic organelle whose structural components and funtional organization vary in relation to the ribosomal synthesis rate. During microsporogenesis, the nucleolus progressively reactivates and the molecular machinery of ribosomal transcription and processing is reorganized. The in situ techniques based on the use of molecular probes to localize specific targes makes it possible to localize molecules and functions in specific subcellular structures, and in cells at precise developmental stages. In this paper, the immunolocalization of various nucleolar antigens and the in situ hibridization for localizing rRNA have been used at light (LM) and transmission electron (TEM) microscopy levels to study the ribosomal transcription and processing in relation to nucleolar architecture during the in vivo pollen developmental process, in Capsicum annuum L., pepper.Peer reviewe

    Characterisation of a flavonoid ligand of the fungal protein Alt a 1

    Spores of pathogenic fungi are virtually ubiquitous and cause human disease and severe losses in crops. The endophytic fungi Alternaria species produce host-selective phytotoxins. Alt a 1 is a strongly allergenic protein found in A. alternata that causes severe asthma. Despite the well-established pathogenicity of Alt a 1, the molecular mechanisms underlying its action and physiological function remain largely unknown. To gain insight into the role played by this protein in the pathogenicity of the fungus, we studied production of Alt a 1 and its activity in spores. We found that Alt a 1 accumulates inside spores and that its release with a ligand is pH-dependent, with optimum production in the 5.0-6.5 interval. The Alt a 1 ligand was identified as a methylated flavonoid that inhibits plant root growth and detoxifies reactive oxygen species. We also found that Alt a 1 changes its oligomerization state depending on the pH of the surrounding medium and that these changes facilitate the release of the ligand. Based on these results, we propose that release of Alt a 1 should be a pathogenic target in approaches used to block plant defenses and consequently to favor fungal entry into the plant
