    Introducing best practice for reproducibility in government

    There is an appetite to improve the reproducibility of quantitative analysis undertaken across government. Our team supports conversion of regular publications into Reproducible Analytical Pipelines (RAP). To achieve this, we developed a roadmap to work with analysts in our organisation and help them transform their pipelines and build their skills. An audit of the current RAP status of all regular pipelines assisted in resource allocation and planning. The maturity of a RAP is evaluated on 7 criteria required to reach a minimum viable product (MVP) and 7 additional advanced criteria as outlined by the Analysis Function (AF). We use a combination of hands-on pair-coding with analysis teams, regular and ad hoc code reviews, and training sessions to convert existing pipelines into RAPs, while simultaneously upskilling the analysts. We have also developed guidance and training documentation to share internally and externally. Currently, out of 73 regular publications, 12 have reached the MVP, with an average score of 4.36 out of 7. This scoring is reassessed monthly, allowing us to track the progress in real-time. Self-assessment of technical skills increased by between 43% and 89% and 97% said their understanding of RAP principals improved because of the training and 77% said they are now able to implement best practice into their work. By working with the pipeline owners instead of just refactoring the code directly, we are ensuring business resilience. The in-depth knowledge of the pipeline and skills required to maintain it are present within the analysis team. Publishing our methods, documentation and tools facilitates adoption of RAP for those without a dedicated RAP team. We are on track to convert all our regular publications into RAPs and move to “RAP by default”, in line with the AF RAP Strategy. This will improve the reproducibility, quality, efficiency, transparency, and trustworthiness of analysis within government. We hope other organisations can learn from our methods

    Developing a Choice-Based Digital Fiction for Body Image Bibliotherapy

    Body dissatisfaction is so common in the western world that it has become the norm, especially among women and girls. Writing New Body Worlds is a transdisciplinary research-creation project that aims to address these issues by developing an interactive digital fiction for body image bibliotherapy. It is created with the critical co-design participation of a group of young women and non-binary individuals (aged 18–25) from diverse backgrounds, who are representative of its intended audience. This article discusses how our participant research influenced the creative development of the digital fiction, its characters and its novel ludonarrative or story-game design. It theorizes how the specific affordances of a choice-based interactive narrative, that situates the reader-player in the mind of the fictional protagonist, may lead to enhanced empathic identification and agency and, therefore, a more profoundly immersive and potentially transformative experience. This process of “diegetic enactment” is where we postulate the therapeutic value lies: an ontological oscillation between the reader-player’s mind and the fictional mind, which may induce the reader-player to reflect upon, and perhaps subtly alter, their own body image.publishedVersio

    Stimulation of Chitin Synthesis Rescues Candida albicans from Echinocandins

    Echinocandins are a new generation of novel antifungal agent that inhibit cell wall β(1,3)-glucan synthesis and are normally cidal for the human pathogen Candida albicans. Treatment of C. albicans with low levels of echinocandins stimulated chitin synthase (CHS) gene expression, increased Chs activity, elevated chitin content and reduced efficacy of these drugs. Elevation of chitin synthesis was mediated via the PKC, HOG, and Ca2+-calcineurin signalling pathways. Stimulation of Chs2p and Chs8p by activators of these pathways enabled cells to survive otherwise lethal concentrations of echinocandins, even in the absence of Chs3p and the normally essential Chs1p, which synthesize the chitinous septal ring and primary septum of the fungus. Under such conditions, a novel proximally offset septum was synthesized that restored the capacity for cell division, sustained the viability of the cell, and abrogated morphological and growth defects associated with echinocandin treatment and the chs mutations. These findings anticipate potential resistance mechanisms to echinocandins. However, echinocandins and chitin synthase inhibitors synergized strongly, highlighting the potential for combination therapies with greatly enhanced cidal activity
