    Zoonotic Risks of Rodents in Livestock Production

    Besides economic losses through spoilage of feed or structural damage, rodent presence on livestock farms can have serious consequences for public and animal health. Direct contact between humans and rodents can be a way of pathogen transmission, but rodents can also transfer (zoonotic) pathogens via livestock to consumers of livestock products. As prevention of food safety hazards is a priority throughout the food chain, the main aim of the work presented in this thesis was to obtain a better understanding of the risk of on-farm rodent presence in the Netherlands

    Role of rodents in transmission of Salmonella and Campylobacter

    Salmonella and Campylobacter are generally regarded as the most important food-borne pathogens in the world. Reduction or elimination of these pathogens in the first part of the food chain (on the farm) is important to prevent disease among consumers of animal products. In organic farming, elimination becomes more difficult, as food animals are allowed outdoors and have easy access to potential sources of hazardous pathogens. Whilst rodents are often associated by organic farmers with infrastructural damage and eating or spoiling of stored feed and products, their zoonotic risks are frequently underestimated. They can amplify the number of pathogens in the environment and transfer them to food animals. Thus organic farmers should be aware of the need for rodent control from a food safety perspective. Preferably, rodent control should form an integral part of a total package of hygiene measures to prevent transfer of food-borne pathogens. These should also include e.g. control of wild birds and flies and obligatory disinfection of boots/clothes and equipment for farm workers and visitors

    Animal-friendly production systems may cause re-emergence of Toxoplasma gondii

    Toxoplasmosis is still one of the most common parasitic infections in the world, although in Europe improvements in hygiene and the introduction of ‘total’ indoor farming in livestock production have rapidly diminished the problem during the past decades. As a result of public dislike, however, introduction of alternative and more acceptable animal-friendly livestock production systems including outdoor access are gaining ground. Potentially these systems can lead to increased prevalence of certain zoonotic diseases, including Toxoplasmosis. To retain prevalence of this disease in humans at current levels, emphasis should be on disease control at farm-level. This article provides an analysis of various risk factors for farm animals to get infected with Toxoplasma gondii. Access of cats to the farm premises, the use of compost and goat whey, and rodent control were identified as possible risk factors that should be addressed. Consumers should be aware of the fact that Toxoplasma infection, besides through meat, can also be caused by the uptake of contaminated water, soil, fruit and vegetables

    Controlling risks of pathogen transmission by flies on organic pig farms - A review

    Fly prevention and control on animal production units is necessary to prevent the transmission of pathogens that could affect animal and human health and the maintenance of good hygiene. Organic farmers are often hesitant to apply insecticides for this purpose because of their farming philosophy. Organic production systems are relatively open as pigs generally have access to the outdoors. Here, we investigate the need for fly control and analyse various possibilities that organic farmers have to reduce the number of flies on their farms. We conclude that although biological control looks promising, more research should be done concerning its side effects. Currently, optimal monitoring and prevention seem to offer the best solution

    Changing climate—changing pathogens: Toxoplasma gondii in North-Western Europe

    In this review, we describe the effects of global climate change for one specific pathogen: the parasite Toxoplasma gondii. It is postulated that an increase of T. gondii prevalence in humans can occur in some regions of North-Western Europe as a result of changing environmental conditions. Such a change can be predicted by using Global Climate Change models. We have elaborated such a prediction for one scenario (SRES A1) by using one specific model (CCSR/NRIES) as an example. Next to environmental factors, also anthropogenic factors may contribute to increased prevalence of T. gondii in this region. In order to counter the potential severe consequences of a potential increase resulting from the combination of climatic and anthropogenic factors, there is an urgent need for the development of a human vaccine. Until a vaccine that offers complete protection is developed, the emphasis should be on treatment optimization and prevention

    Animal-related factors associated with moderate-to-severe diarrhea in children younger than five years in western Kenya: A matched case-control study

    Background Diarrheal disease remains among the leading causes of global mortality in children younger than 5 years. Exposure to domestic animals may be a risk factor for diarrheal disease. The objectives of this study were to identify animal-related exposures associated with cases of moderate-to-severe diarrhea (MSD) in children in rural western Kenya, and to identify the major zoonotic enteric pathogens present in domestic animals residing in the homesteads of case and control children. Methodology/Principal findings We characterized animal-related exposures in a subset of case and control children (n = 73 pairs matched on age, sex and location) with reported animal presence at home enrolled in the Global Enteric Multicenter Study in western Kenya, and analysed these for an association with MSD. We identified potentially zoonotic enteric pathogens in pooled fecal specimens collected from domestic animals resident at children’s homesteads. Variables that were associated with decreased risk of MSD were washing hands after animal contact (matched odds ratio [MOR] = 0.2; 95% CI 0.08–0.7), and presence of adult sheep that were not confined in a pen overnight (MOR = 0.1; 0.02–0.5). Variables that were associated with increased risk of MSD were increasing number of sheep owned (MOR = 1.2; 1.0–1.5), frequent observation of fresh rodent excreta (feces/urine) outside the house (MOR = 7.5; 1.5–37.2), and participation of the child in providing water to chickens (MOR = 3.8; 1.2–12.2). Of 691 pooled specimens collected from 2,174 domestic animals, 159 pools (23%) tested positive for one or more potentially zoonotic enteric pathogens (Campylobacter jejuni, C. coli, non-typhoidal Salmonella, diarrheagenic E. coli, Giardia, Cryptosporidium, or rotavirus). We did not find any association between the presence of particular pathogens in household animals, and MSD in children. Conclusions and significance Public health agencies should continue to promote frequent hand washing, including after animal contact, to reduce the risk of MSD. Future studies should address specific causal relations of MSD with sheep and chicken husbandry practices, and with the presence of rodents

    Biosecurity and Yield Improvement Technologies Are Strategic Complements in the Fight against Food Insecurity

    The delivery of food security via continued crop yield improvement alone is not an effective food security strategy, and must be supported by pre- and post-border biosecurity policies to guard against perverse outcomes. In the wake of the green revolution, yield gains have been in steady decline, while post-harvest crop losses have increased as a result of insufficiently resourced and uncoordinated efforts to control spoilage throughout global transport and storage networks. This paper focuses on the role that biosecurity is set to play in future food security by preventing both pre- and post-harvest losses, thereby protecting crop yield. We model biosecurity as a food security technology that may complement conventional yield improvement policies if the gains in global farm profits are sufficient to offset the costs of implementation and maintenance. Using phytosanitary measures that slow global spread of the Ug99 strain of wheat stem rust as an example of pre-border biosecurity risk mitigation and combining it with post-border surveillance and invasive alien species control efforts, we estimate global farm profitability may be improved by over US$4.5 billion per annum

    Effect of synthetic hormones on reproduction in Mastomys natalensis

    Rodent pest management traditionally relies on some form of lethal control. Developing effective fertility control for pest rodent species could be a major breakthrough particularly in the context of managing rodent population outbreaks. This laboratory-based study is the first to report on the effects of using fertility compounds on an outbreaking rodent pest species found throughout sub-Saharan Africa. Mastomys natalensis were fed bait containing the synthetic steroid hormones quinestrol and levonorgestrel, both singly and in combination, at three concentrations (10, 50, 100 ppm) for seven days. Consumption of the bait and animal body mass was mostly the same between treatments when analysed by sex, day and treatment. However, a repeated measures ANOVA indicated that quinestrol and quinestrol+levonorgestrel treatments reduced consumption by up to 45%, particularly at the higher concentrations of 50 and 100 ppm. Although there was no clear concentration effect on animal body mass, quinestrol and quinestrol+levonorgestrel lowered body mass by up to 20% compared to the untreated and levonorgestrel treatments. Quinestrol and quinestrol+levonorgestrel reduced the weight of male rat testes, epididymis and seminal vesicles by 60-80%, and sperm concentration and motility were reduced by more than 95%. No weight changes were observed to uterine and ovarian tissue; however, high uterine oedema was observed among all female rats consuming treated bait at 8 days and 40 days from trial start. Trials with mate pairing showed there were significant differences in the pregnancy rate with all treatments when compared to the untreated control group of rodents

    Multiple Geographic Origins of Commensalism and Complex Dispersal History of Black Rats

    The Black Rat (Rattus rattus) spread out of Asia to become one of the world's worst agricultural and urban pests, and a reservoir or vector of numerous zoonotic diseases, including the devastating plague. Despite the global scale and inestimable cost of their impacts on both human livelihoods and natural ecosystems, little is known of the global genetic diversity of Black Rats, the timing and directions of their historical dispersals, and the risks associated with contemporary movements. We surveyed mitochondrial DNA of Black Rats collected across their global range as a first step towards obtaining an historical genetic perspective on this socioeconomically important group of rodents. We found a strong phylogeographic pattern with well-differentiated lineages of Black Rats native to South Asia, the Himalayan region, southern Indochina, and northern Indochina to East Asia, and a diversification that probably commenced in the early Middle Pleistocene. We also identified two other currently recognised species of Rattus as potential derivatives of a paraphyletic R. rattus. Three of the four phylogenetic lineage units within R. rattus show clear genetic signatures of major population expansion in prehistoric times, and the distribution of particular haplogroups mirrors archaeologically and historically documented patterns of human dispersal and trade. Commensalism clearly arose multiple times in R. rattus and in widely separated geographic regions, and this may account for apparent regionalism in their associated pathogens. Our findings represent an important step towards deeper understanding the complex and influential relationship that has developed between Black Rats and humans, and invite a thorough re-examination of host-pathogen associations among Black Rats
