Latrunculin A delays anaphase onset in fission yeast by disrupting an ase1-independent pathway controlling mitotic spindle stability

It has been proposed previously that latrunculin A, an inhibitor of actin polymerization, delays the onset of anaphase by causing spindle misorientation in fission yeast. However, we show that {Delta}mto1 cells, which are defective in nucleation of cytoplasmic microtubules, have profoundly misoriented spindles but are not delayed in the timing of sister chromatid separation, providing compelling evidence that fission yeast does not possess a spindle orientation checkpoint. Instead, we show that latrunculin A delays anaphase onset by disrupting interpolar microtubule stability. This effect is abolished in a latrunculin A-insensitive actin mutant and exacerbated in cells lacking Ase1, which cross-links antiparallel interpolar microtubules at the spindle midzone both before and after anaphase. These data indicate that both Ase1 and an intact actin cytoskeleton are required for preanaphase spindle stability. Finally, we show that loss of Ase1 activates a checkpoint that requires only the Mad3, Bub1, and Mph1, but not Mad1, Mad2, or Bub3 checkpoint proteins

Improving responses to depression and related disorders: evaluation of a innovative, general, mental health care workers training program

<p>Abstract</p> <p>Background</p> <p>Australian General Practitioners have been beneficiaries of extensive training in mental health care delivery over the last few years but less so other workers who support those with mental illness. Training is needed as it is widely recognised that the most effective interventions to prevent and treat mental disorders are often not readily available. The Mental Health Aptitudes into Practice (MAP) training package is a broad, innovative, interdisciplinary, general mental health training aimed at improving responses to individuals with depression and related disorders. The modular structure of this training program meant that such training could be targeted at those with varied backgrounds. Two hundred and seventy one days of free MAP training was delivered across Victoria in 2004/2005. The evaluation reported here assessed whether changes occurred in the trainees' confidence, mental health literacy, attitudes towards effective treatments, mental health knowledge and skills and community mental health ideology following training.</p> <p>Methods</p> <p>These elements were assessed using pen and paper tests prior, immediately following, 6 months after and then 12 months after the training. Trainees' confidence, mental health literacy and social distance were measured using scales that have been used in evaluations of Mental Health First Aid Training. Community mental health ideology was measured using a sub-scale of the Community Attitudes to the Mentally Ill (CAMI) scale. The trainees' knowledge and skills were accessed using instrumentation specifically designed for this evaluation.</p> <p>Results</p> <p>Following training, participants had more confidence in their ability to work with those who have mental health issues and less desire for social distance from them. Participants' knowledge and skills in relation to the treatment of mental disorders increased. These changes were observed immediately after training. The limited existing evidence suggests these changes were sustained six and twelve months later.</p> <p>Conclusions</p> <p>MAP training can be used to develop the capacity and capabilities of mental health workers.</p

THE DECORATED SPINDLE-SHAPED BONE DAGGER FROM ŠARNELĖ: THE EARLIEST EXAMPLE OF HUNTER-GATHERER MOBILE ART IN LITHUANIA

This paper presents the latest data on a spindle-shaped decorated bone dagger, which was found as a stray find in the village of Šarnelė in northwest Lithuania, and which recently been dated to the Final Palaeolithic. It is currently the only one such example of osseous technology in Lithuania dated to this period. In 2016, we started to reinvestigate the Stone Age material from the Samogitian highland by AMS radiocarbon (14C) dating, stable isotopes, use-wear and Raman methods. AMS dating showed that the decorated dagger discovered in the surroundings of the drained Lake Ertenis and the River Varduva at Šarnelė currently is one of the oldest prehistoric art objects found in Lithuania and possibly in the eastern Baltic region as well. The dagger was also analysed by use-wear method, which helped to identify some stages of the production of the article itself and its decoration. Libbie & Grove Urban Design Plan

This plan was created for the City of Richmond Department of Planning and Development Review to serve as a recommendation for urban design improvements and suggested changes to zoning ordinances for the Libbie and Grove commercial area located in the Westhampton neighborhood. To begin, an in-depth demographic analysis was conducted for the Westhampton neighborhood. Special attention was paid to socioeconomic factors and trends in census tracts directly surrounding the Libbie and Grove commercial corridor. Based on these analyses and new development occurring in the Libbie and Grove commercial corridor, we were able to allocate six sites or “study areas” as candidates for redevelopment. All of these sites represent valuable areas within the Libbie and Grove commercial corridor. The sites were selected and designed with different intentions, but aim to create a complete streetscape for the commercial area. Based on this analysis and study, it is our recommendation that a new zoning code be implemented for the Libbie and Grove commercial area in order to codify form based design requirements in order to preserve and enhance a village feel at Grove and Libbie and promote compatible future development

What are the most effective interventions to reduce childhood obesity?

Efforts to increase physical activity or decrease sedentary activities have shown some short-term benefit, and adding dietary changes may be more effective. Aiming interventions at parents, intensive family therapy, comprehensive school-based programs, and selecting motivated children for subspecialty care may improve success. (Grade of recommendation: B, based on poor-quality randomized controlled trials [RCTs] and heterogeneous systematic reviews.) Other potentially effective short-term strategies include screening with body mass index (BMI) for age (grade of recommendation: C, extrapolation from cohort studies and ecological research) or dietary counseling (grade of recommendation: D, conflicting poor-quality RCTs). No drugs are currently approved for pediatric obesity therapy in the United States
