    Complex responses to movement-based disease control: when livestock trading helps

    Livestock disease controls are often linked to movements between farms, for example, via quarantine and pre- or post-movement testing. Designing effective controls, therefore, benefits from accurate assessment of herd-to-herd transmission. Household models of human infections make use of R*, the number of groups infected by an initial infected group, which is a metapopulation level analogue of the basic reproduction number R0 that provides a better characterization of disease spread in a metapopulation. However, existing approaches to calculate R* do not account for individual movements between locations which means we lack suitable tools for livestock systems. We address this gap using next-generation matrix approaches to capture movements explicitly and introduce novel tools to calculate R* in any populations coupled by individual movements. We show that depletion of infectives in the source group, which hastens its recovery, is a phenomenon with important implications for design and efficacy of movement-based controls. Underpinning our results is the observation that R* peaks at intermediate livestock movement rates. Consequently, under movement-based controls, infection could be controlled at high movement rates but persist at intermediate rates. Thus, once control schemes are present in a livestock system, a reduction in movements can counterintuitively lead to increased disease prevalence. We illustrate our results using four important livestock diseases (bovine viral diarrhoea, bovine herpes virus, Johne's disease and Escherichia coli O157) that each persist across different movement rate ranges with the consequence that a change in livestock movements could help control one disease, but exacerbate another

    The environments produced by entry into Jupiter and Saturn atmospheres are summarized. Worst case design environments are identified and the effect of entry angle, type of atmosphere and ballistic coefficient variations are presented. The range of environments experienced during earth entry is parametrically described as a function of initial entry conditions. The sensitivity of these environments to vehicle ballistic coefficient and nose radius is also shown. An elliptical deorbit maneuver strategy is defined in terms of the velocity increment required versus initial entry conditions and apoapsis altitude. Mission time, ground track, and out of plane velocity penalties are also presented. Performance capabilities of typical shuttle launched boosters are described including the initial entry conditions attainable as a function of paylaod mass and apoapsis altitude

    The feasibility of using the space shuttle to launch planetary and DoD entry flight experiments was examined. The results of the program are presented in two parts: (1) simulating outer planet environments during an earth entry test, the prediction of Jovian and earth radiative heating dominated environments, mission strategy, booster performance and entry vehicle design, and (2) the DoD entry test needs for the 1980's, the use of the space shuttle to meet these DoD test needs, modifications of test procedures as pertaining to the space shuttle, modifications to the space shuttle to accommodate DoD test missions and the unique capabilities of the space shuttle.     Activin B is produced early in antral follicular development and suppresses thecal androgen production

    Little is known about the role of activin B during folliculogenesis. This study investigated the expression levels of activin/inhibin subunits (βA, βB, and α), steroid enzyme, and gonadotrophin receptors in theca (TC) and granulosa cells (GC) by QPCR and activin A and B and inhibin A protein levels in follicular fluid (FF) of developing sheep follicles during estrus and anestrus. The effect of activin B on androgen production from primary TC cultures in vitro was also assessed. During folliculogenesis, in anestrus and estrus, FF activin B concentrations and thecal and GC activin βB mRNA levels decreased as follicle diameter increased from 1–3 to >6 mm regardless of estrogenic status. Estrogenic preovulatory follicles had reduced concentrations of FF activins B and A, and TC and GCs expressed higher levels of activin βA mRNA at 3–4 mm, and TCs more inhibin α mRNA at >4 mm stages of development compared with nonestrogenic follicles. Activin B decreased androstenedione production from primary TCs in vitro, an effect blocked by inhibin A. Thus, sheep follicles 1–3 mm in diameter contained high FF levels of activin B, which decreased as the follicle size increased, and, like activin A, suppressed thecal androgen production in vitro, an effect blocked by inhibin. Furthermore, the theca of large estrogenic follicles expressed high levels of inhibin α and activin βA mRNA suggesting local thecal derived inhibin A production. This would inhibit the negative effects of thecal activins B and A ensuring maximum androgen production for enhanced estradiol production by the preovulatory follicle(s)

    Bone morphogenetic protein-4 interacts with activin and GnRH to modulate gonadotrophin secretion in LβT2 gonadotrophs

    We have shown previously that, in sheep primary pituitary cells, bone morphogenetic proteins (BMP)-4 inhibits FSHβ mRNA expression and FSH release. In contrast, in mouse LβT2 gonadotrophs, others have shown a stimulatory effect of BMPs on basal or activin-stimulated FSHβ promoter-driven transcription. As a species comparison with our previous results, we used LβT2 cells to investigate the effects of BMP-4 on gonadotrophin mRNA and secretion modulated by activin and GnRH. BMP-4 alone had no effect on FSH production, but enhanced the activin+GnRH-induced stimulation of FSHβ mRNA and FSH secretion, without any effect on follistatin mRNA. BMP-4 reduced LHβ mRNA up-regulation in response to GnRH (±activin) and decreased GnRH receptor expression, which would favour FSH, rather than LH, synthesis and secretion. In contrast to sheep pituitary gonadotrophs, which express only BMP receptor types IA (BMPRIA) and II (BMPRII), LβT2 cells also express BMPRIB. Smad1/5 phosphorylation induced by BMP-4, indicating activation of BMP signalling, was the same whether BMP-4 was used alone or combined with activin±GnRH. We hypothesized that activin and/or GnRH pathways may be modulated by BMP-4, but neither the activin-stimulated phosphorylation of Smad2/3 nor the GnRH-induced ERK1/2 or cAMP response element-binding phosphorylation were modified. However, the GnRH-induced activation of p38 MAPK was decreased by BMP-4. This was associated with increased FSHβ mRNA levels and FSH secretion, but decreased LHβ mRNA levels. These results confirm 1. BMPs as important modulators of activin and/or GnRH-stimulated gonadotrophin synthesis and release and 2. important species differences in these effects, which could relate to differences in BMP receptor expression in gonadotrophs

    Novel expression of Haemonchus contortus vaccine candidate aminopeptidase H11 using the free-living nematode Caenorhabditis elegans

    With the problem of parasitic nematode drug resistance increasing, vaccine development offers an alternative sustainable control approach. For some parasitic nematodes, native extracts enriched for specific proteins are highly protective. However, recombinant forms of these proteins have failed to replicate this protection. This is thought to be due to differences in glycosylation and/or conformation between native and recombinant proteins. We have exploited the free-living nematode Caenorhabditis elegans to examine its suitability as an alternative system for recombinant expression of parasitic nematode vaccine candidates. We focussed on Haemonchus contortus aminopeptidase H11 glycoprotein, which is enriched in a gut membrane fraction capable of inducing significant protection against this important ovine gastrointestinal nematode. We show that H. contortus H11 expressed in C. elegans is enzymatically active and MALDI mass spectrometry identifies similar di- and tri-fucosylated structures to those on native H11, with fucose at the 3- and/or 6-positions of the proximal GlcNAc. Some glycan structural differences were observed, such as lack of LDNF. Serum antibody to native H11 binds to C. elegans recombinant H11 and most of the antibody to rH11 or native H11 is directed to glycan moieties. Despite these similarities, no reduction in worm burden or faecal egg count was observed following immunisation of sheep with C. elegans-expressed recombinant H11 protein. The findings suggest that the di- and tri-fucosylated N-glycans expressed on rH11 do not contribute to the protective effect of H11 and that additional components present in native H11-enriched extract are likely required for enhancing the antibody response necessary for protection

    Overt and relational aggression in Russian nursery-school-age children: parenting style and marital linkages.

    Maternal and paternal parenting styles and marital interactions linked to childhood aggressive behavior as described in Western psychological literature were measured in an ethnic Russian sample of 207 families of nursery-school-age children. Results corroborated and extended findings from Western samples. Maternal and paternal coercion, lack of responsiveness, and psychological control (for mothers only) were significantly correlated with children\u27s overt aggression with peers. Less responsiveness (for mothers and fathers) and maternal coercion positively correlated with relational aggression. Some of these associations differed for boys versus girls. Marital conflict was also linked to more overt and relational aggression for boys. When entered into the same statistical model, more marital conflict (for boys only), more maternal coercion, and less paternal responsiveness were found to be the most important contributors to overt and relational aggression in younger Russian children
