    Profiling of Glycan Receptors for Minute Virus of Mice in Permissive Cell Lines Towards Understanding the Mechanism of Cell Recognition

    The recognition of sialic acids by two strains of minute virus of mice (MVM), MVMp (prototype) and MVMi (immunosuppressive), is an essential requirement for successful infection. To understand the potential for recognition of different modifications of sialic acid by MVM, three types of capsids, virus-like particles, wild type empty (no DNA) capsids, and DNA packaged virions, were screened on a sialylated glycan microarray (SGM). Both viruses demonstrated a preference for binding to 9-O-methylated sialic acid derivatives, while MVMp showed additional binding to 9-O-acetylated and 9-O-lactoylated sialic acid derivatives, indicating recognition differences. The glycans recognized contained a type-2 Galβ1-4GlcNAc motif (Neu5Acα2-3Galβ1-4GlcNAc or 3′SIA-LN) and were biantennary complex-type N-glycans with the exception of one. To correlate the recognition of the 3′SIA-LN glycan motif as well as the biantennary structures to their natural expression in cell lines permissive for MVMp, MVMi, or both strains, the N- and O-glycans, and polar glycolipids present in three cell lines used for in vitro studies, A9 fibroblasts, EL4 T lymphocytes, and the SV40 transformed NB324K cells, were analyzed by MALDI-TOF/TOF mass spectrometry. The cells showed an abundance of the sialylated glycan motifs recognized by the viruses in the SGM and previous glycan microarrays supporting their role in cellular recognition by MVM. Significantly, the NB324K showed fucosylation at the non-reducing end of their biantennary glycans, suggesting that recognition of these cells is possibly mediated by the Lewis X motif as in 3′SIA-LeX identified in a previous glycan microarray screen

    Chromospheric and photospheric evolution of an extremely active solar region in solar cycle 19

    a comprehensive investigation was made of phenomena attending the disk passage, July 7 to 21, 1959, of active solar center HAO-59Q. At the photospheric level that comprised an aggregate of groups of sunspots of which one group, Mt. Wilson 14284, showed all the attributes deemed typical of solar regions associated with the production of major flares. A special characteristic of 59Q was its capability to eject dark material. Part of this material remained trapped in the strong magnetic fields above group 14284 where it formed a system of interrelated arches, the legs of which passed through components of the bright chromospheric network of the plage and were rooted in various underlying umbrae. Two apparently diffeent kinds of flare were identified in 59Q; namely, prominence flares (which comprised brightenings within part of the suspended dark prominence) and plage flares (which comprised brightenings within part of the chromospheric network). Prominence flares were of three varieties described as 'impact', 'stationary' and 'moving' prominence flares. Plage flares were accompanied in 3 percent of cases by Type III bursts. These latter radio events indicate the associated passage through the corona of energetic electrons in the approximate energy range 10 to 100 keV. At least 87.5 percent, and probably all, impulsive brightenings in 59Q began directly above minor spots, many of which satellites to major umbrae. Stationary and moving prominence flares were individually triggered at sites beneath which magnetic changes occurred within intervals which included each flare's flash phase

    Self-esteem outcomes over a summer camp for obese youth.

    Variation in the existing literature on the psychosocial benefits of weight loss in obese youth results, in part, from methodological limitations and modest weight loss. Accordingly, this research assessed perceived self-competence and low self-esteem during an intensive weight loss programme in a large sample of obese youth and related these to starting weight, gender and weight loss.Over 4 years, 303 obese male and female adolescents (body mass index [BMI] 34.3 kg m(-2) , BMI standard deviation score 2.99; 14.7 years) attended a residential weight loss camp for a mean duration of 31 d. Outcome variables included dimensional self-esteem (Harter) and weight change over the camp.At the start of camp, obese youth scored highest on social acceptance and lowest on physical appearance and athletic competence. Global self-worth and most domains of self-competence improved significantly over the intervention. The proportion with low global self-worth reduced from 35% to 16%, but there was little change in the proportion reporting high self-competence (23%). Mean weight loss was -5.5 kg (BMI standard deviation scores -0.25) with boys and those heaviest at the start losing most. Weight loss was significantly correlated with improved physical appearance (r = 0.13) and athletic competence (r = 0.19), but not global self-worth.This intensive weight loss intervention yielded significant psychological benefit, especially in self-competence and among individuals achieving most weight loss. The weak association with weight loss suggests the influence of other contributing environmental or social features that should be the focus of further research

    Trademark law exists to promote competition. If consumers know which companies make which products, they can more easily find the products they actually want to purchase. Trademark law has long treated “source significance”—the fact that a particular trademark is identified with a particular producer—as both necessary and sufficient for establishing a valid trademark. That is, trademark law has traditionally viewed source significance as the only necessary precondition for a trademark being pro-competitive. In this paper, we argue that this equation of source significance and pro-competitiveness is misguided. Some marks use words that are so closely connected with the product being branded that giving just one firm a monopoly over those words provides that firm with a meaningful competitive advantage—an artificial advantage granted by the state. This problem becomes worse as the number of firms producing (and branding) a type of product increases. The more words cordoned off by trademark law, the more trouble a new entrant will have in describing or attracting attention to its product. Trademark law is thus being hijacked by strategic firms for anti-competitive purposes. Traditional doctrinal tools are inadequate to address this problem because the goal should be to limit the number of such trademarks rather than eliminate them completely. However, costly screens could be used to impose a form of congestion pricing on trademarks, eliminating them in all but the most worthwhile cases. In this paper, we develop a theory of the anti-competitive nature of certain trademark rules. We then propose a series of overlapping doctrinal rules and costly screens to address the problem of rampant anticompetitive trademark

    Physical activity assessment for public health: efficacious use of the single-item measure.

    © 2015 The Royal Society for Public Health. Objectives: The accurate mass assessment of physical activity is essential for effective Public Health policy and practice. Combined with a desire to minimize participant burden, the self-reported single-item physical activity screening measure has become increasingly attractive and widespread. To help reduce any potential misclassification, refining this instrumentation in line with any changes in prescribed activity levels is essential to optimize accuracy. Study design: This study compares the levels of agreement, sensitivity and specificity for the single-item measure versus International Physical Activity Questionnaire (IPAQ) using current physical activity recommendations. Methods: Agreement was assessed in a non-probability sample of 7650 adults. The κ statistic, sensitivity and specificity were used to assess agreement between the tools for classifying participants as sufficiently active for health (≥150 min of physical activity per week) or not, and being classified as inactive (<30 of minutes of physical activity per week) or not. Results: The single-item measure showed weak agreement with the IPAQ for identifying participants who met the current physical activity guidelines (κ = 0.13, 95% CI 0.12 to 0.14), sensitivity was 18.7% and specificity was 97.2%. For the classification of inactive participants it showed a moderate agreement with IPAQ (κ = 0.45, 95% CI 0.43 to 0.47), sensitivity was 74.2% and specificity was 79.7%. Conclusions: The single-item measure had a low diagnostic capacity compared to IPAQ. Further research is needed if it is to be used in large scale surveys and interventions where screening for sufficiently active or inactive individuals is the goal

    Self-diffusion in binary blends of cyclic and linear polymers

    A lattice model is used to estimate the self-diffusivity of entangled cyclic and linear polymers in blends of varying compositions. To interpret simulation results, we suggest a minimal model based on the physical idea that constraints imposed on a cyclic polymer by infiltrating linear chains have to be released, before it can diffuse beyond a radius of gyration. Both, the simulation, and recently reported experimental data on entangled DNA solutions support the simple model over a wide range of blend compositions, concentrations, and molecular weights.Comment: 10 pages, 2 figure

    Equilibrium and out of equilibrium thermodynamics in supercooled liquids and glasses

    We review the inherent structure thermodynamical formalism and the formulation of an equation of state for liquids in equilibrium based on the (volume) derivatives of the statistical properties of the potential energy surface. We also show that, under the hypothesis that during aging the system explores states associated to equilibrium configurations, it is possible to generalize the proposed equation of state to out-of-equilibrium conditions. The proposed formulation is based on the introduction of one additional parameter which, in the chosen thermodynamic formalism, can be chosen as the local minima where the slowly relaxing out-of-equilibrium liquid is trapped.Comment: 7 pages, 4 eps figure

    Initial effects of a free swimming pilot programme on the physical activity levels of young people.

    Concerns remain over the physical activity (PA) levels of young people.1,2 Consequently, identifying interventions that are effective at encouraging young people to adopt and improve PA levels over the life course3 – especially those not meeting PA guidelines – is central to non-communicable disease prevention in later life.1 Efforts to increase PA levels through the promotion of swimming and aquatic activities for children and young people are one such option.4,5 Swimming has been referred to as the UK's ‘major participation sport’ and a mode of exercise that inactive groups contemplate when seeking to increase their PA levels.4 The aspiration to find effective and sustainable models of PA intervention necessitates rigorous monitoring and evaluation within the context where implementation takes place. With those thoughts in mind, this research set out to investigate the effectiveness of a local authority (LA) led pilot programme of free swimming (FS), with this paper reporting the initial key findings emerging from this study
