    An increase in N-Ras expression is associated with development of hormone refractory prostate cancer in a subset of patients

    Protein expression of H, K and N-Ras was assessed in hormone sensitive and hormone refractory prostate tumour pairs from 61 patients by immunohistochemistry. Expression of H-Ras and K- Ras was not associated with any known clinical parameters. In contrast an increase in N-Ras membrane expression in the transition from hormone sensitive to hormone refractory prostate cancer was associated with shorter time to relapse (p=0.01) and shorter disease specific survival (p=0.008). In addition, patients with an increase in N-Ras membrane expression had lower levels of PSA at relapse (p=0.02) and expression correlated with phosphorylated MAP kinase (p=0.010) and proliferation index (Ki67, p=0.02). These results suggest that in a subgroup patients N-Ras expression is associated with development of hormone refractory prostate cancer via activation of the MAP kinase cascade

    Running a distributed virtual observatory: US Virtual Astronomical Observatory operations

    Operation of the US Virtual Astronomical Observatory shares some issues with modern physical observatories, e.g., intimidating data volumes and rapid technological change, and must also address unique concerns like the lack of direct control of the underlying and scattered data resources, and the distributed nature of the observatory itself. In this paper we discuss how the VAO has addressed these challenges to provide the astronomical community with a coherent set of science-enabling tools and services. The distributed nature of our virtual observatory-with data and personnel spanning geographic, institutional and regime boundaries-is simultaneously a major operational headache and the primary science motivation for the VAO. Most astronomy today uses data from many resources. Facilitation of matching heterogeneous datasets is a fundamental reason for the virtual observatory. Key aspects of our approach include continuous monitoring and validation of VAO and VO services and the datasets provided by the community, monitoring of user requests to optimize access, caching for large datasets, and providing distributed storage services that allow user to collect results near large data repositories. Some elements are now fully implemented, while others are planned for subsequent years. The distributed nature of the VAO requires careful attention to what can be a straightforward operation at a conventional observatory, e.g., the organization of the web site or the collection and combined analysis of logs.     Facing the Leadership Challenges on Decommissioning United States Navy Ships

    Since the late 1980s, hundreds of U.S. Navy ships have been decommissioned. Although ship decommissionings were postponed immediately after the September 11th attack, today the Navy is again engaged in “rightsizing” its force. Depending on the type of ship and its age, a ship faces different fates after the decommissioning process has been completed. Except on the rare occasion when a ship is turned into a museum, most decommissioned ships are disposed of in one of three ways. Some ships are sold to foreign navies to recoup some costs. Other ships are reassigned to the Military Sealift Command (MSC) to continue serving the fleet, albeit in a quasi-civilian capacity. The last option for a decommissioned ship is to be placed in an inactive status or “mothballs” and eventually sunk. The impact of the decommissioning process on the crew is a major concern. The day the decommissioning ship completes its last operational assignment, the “mission” of the ship changes significantly: The crew must prepare its ship for a kind of death. Those in authority who may have recently mastered the art of leading sailors into hostile environments now must motivate their sailors and themselves to excel in the absence of danger. Consequently, the leaders on the ship are faced with a difficult challenge: to maintain morale and to keep the crew on task even though the mission and tasks have changed dramatically. If a naval leader cannot successfully adapt his/her leadership style to the environment created by the decommissioning, the crew morale may be adversely affected and retention may plummet. The purpose of this research was to begin to explore leadership challenges in naval vessels that were scheduled for decommissioning. The study identified the perceptions of nine naval leaders from three decommissioning ships regarding the decommissioning process. These perceptions were organized into 13 major themes that provide valuable information and insight about naval leadership during the decommissioning process. This information should be useful to prospective naval leaders who encounter decommissioning situations during their tours of duty

    Cellular location and activity of Escherichia coli RecG proteins shed light on the function of its structurally unresolved C-terminus

    RecG is a DNA translocase encoded by most species of bacteria. The Escherichia coli protein targets branched DNA substrates and drives the unwinding and rewinding of DNA strands. Its ability to remodel replication forks and to genetically interact with PriA protein have led to the idea that it plays an important role in securing faithful genome duplication. Here we report that RecG co-localises with sites of DNA replication and identify conserved arginine and tryptophan residues near its C-terminus that are needed for this localisation. We establish that the extreme C-terminus, which is not resolved in the crystal structure, is vital for DNA unwinding but not for DNA binding. Substituting an alanine for a highly conserved tyrosine near the very end results in a substantial reduction in the ability to unwind replication fork and Holliday junction structures but has no effect on substrate affinity. Deleting or substituting the terminal alanine causes an even greater reduction in unwinding activity, which is somewhat surprising as this residue is not uniformly present in closely related RecG proteins. More significantly, the extreme C-terminal mutations have little effect on localisation. Mutations that do prevent localisation result in only a slight reduction in the capacity for DNA repair. © 2014 The Author(s)

    Surface excitonic emission and quenching effects in ZnO nanowire/nanowall systems: limiting effects on device potential.

    We report ZnO nanowire/nanowall growth using a two-step vapour phase transport method on a-plane sapphire. X-ray diffraction and scanning electron microscopy data establish that the nanostructures are vertically well-aligned with c-axis normal to the substrate, and have a very low rocking curve width. Photoluminescence data at low temperatures demonstrate the exceptionally high optical quality of these structures, with intense emission and narrow bound exciton linewidths. We observe a high energy excitonic emission at low temperatures close to the band-edge which we assign to the surface exciton in ZnO at ~ 3.366 eV, the first time this feature has been reported in ZnO nanorod systems. This assignment is consistent with the large surface to volume ratio of the nanowire systems and indicates that this large ratio has a significant effect on the luminescence even at low temperatures. The band-edge intensity decays rapidly with increasing temperature compared to bulk single crystal material, indicating a strong temperature-activated non-radiative mechanism peculiar to the nanostructures. No evidence is seen of the free exciton emission due to exciton delocalisation in the nanostructures with increased temperature, unlike the behaviour in bulk material. The use of such nanostructures in room temperature optoelectronic devices appears to be dependent on the control or elimination of such surface effects

    Ras/Raf-1/MAPK pathway mediates response to tamoxifen but not chemotherapy in breast cancer patients

    <b>Purpose</b>: The expression and activation of the Ras/Raf-1/mitogen-activated protein kinase (MAPK) pathway plays an important role in the development and progression of cancer, and may influence response to treatments such as tamoxifen and chemotherapy. In this study we investigated whether the expression and activation of the key components of this pathway influenced clinical outcome, to test the hypothesis that activation of the MAPK pathway drives resistance to tamoxifen and chemotherapy in women with breast cancer. <b>Experimental Design</b>: Breast tumors from patients at the Glasgow Royal Infirmary and others treated within the BR9601 trial were analyzed for expression of the three Ras isoforms, total Raf-1, active and inactive forms of Raf-1 [pRaf(ser338) and pRaf(ser259), respectively], MAPK, and phospho-MAPK using an immunohistochemical approach. Analyses were done with respect to disease free-survival and overall survival. <b>Results</b>: Expression and activation of the Ras pathway was associated with loss of benefit from treatment with tamoxifen but not chemotherapy. Overexpression of pRaf(ser338) was associated with shortened disease-free and overall survival time in univariate analyses. Multivariate analysis suggested pRaf(ser338) was independent of known prognostic markers in predicting outcome following tamoxifen treatment (<i>P</i>=0.03). <b>Conclusion</b>: This study suggests that activation of the Ras pathway predicts for poor outcome on tamoxifen but not chemotherapy, and identifies pRaf(ser338) as a potential marker of resistance to estrogen receptor–targeted therapy. In addition, it suggests that expression of pRaf(ser338) could identify patients for whom tamoxifen alone is insufficient adjuvant systemic therapy, but for whom the addition of chemotherapy may be of benefit

    OMCat: Catalogue of Serendipitous Sources Detected with the XMM-Newton Optical Monitor

    The Optical Monitor Catalogue of serendipitous sources (OMCat) contains entries for every source detected in the publicly available XMM-Newton Optical Monitor (OM) images taken in either the imaging or ``fast'' modes. Since the OM is coaligned and records data simultaneously with the X-ray telescopes on XMM-Newton, it typically produces images in one or more near-UV/optical bands for every pointing of the observatory. As of the beginning of 2006, the public archive had covered roughly 0.5% of the sky in 2950 fields. The OMCat is not dominated by sources previously undetected at other wavelengths; the bulk of objects have optical counterparts. However, the OMCat can be used to extend optical or X-ray spectral energy distributions for known objects into the ultraviolet, to study at higher angular resolution objects detected with GALEX, or to find high-Galactic-latitude objects of interest for UV spectroscopy.Comment: 25 pages, 22 figures, submitted to PAS

    Assessing latin@ public opinion on foreign affairs and its potential impact on the 2012 presidential election

    As the Latin@ population of the United States grows and the “Sleeping Giant” is expected to play a more significant role in the nation’s elections and governance more attention must be paid to the factors influencing Latin@ public opinion and political behavior on a myriad of issues. While there is a growing literature addressing Latin@ partisanship and opinion on issues such as morality and social welfare programs, there is a dearth of information regarding their opinion on foreign affairs and policy outside of Latin America. We review the limited studies that exist examining Latin@ opinion on policy issues and then explain why this can and will matter in Latin@ voting behavior and finally, using data from the 2006 Pew Hispanic Center survey on Latin@s and Religion in the U.S., we explain how such factors as religion, country of origin and ideology influence Latin@ public opinion on foreign affairs demonstrating a need for much greater study in this area.A medida que la poblaciĂłn latina de los Estados Unidos crece, se espera que el "gigante dormido" desempeñe un papel mĂĄs importante en las elecciones presidenciales y el gobierno debe prestar mĂĄs atenciĂłn a los factores que influyen en la opiniĂłn pĂșblica latina y el comportamiento polĂ­tico de una gran variedad de temas.
