    Contextualised Browsing in a Digital Library's Living Lab

    Contextualisation has proven to be effective in tailoring \linebreak search results towards the users' information need. While this is true for a basic query search, the usage of contextual session information during exploratory search especially on the level of browsing has so far been underexposed in research. In this paper, we present two approaches that contextualise browsing on the level of structured metadata in a Digital Library (DL), (1) one variant bases on document similarity and (2) one variant utilises implicit session information, such as queries and different document metadata encountered during the session of a users. We evaluate our approaches in a living lab environment using a DL in the social sciences and compare our contextualisation approaches against a non-contextualised approach. For a period of more than three months we analysed 47,444 unique retrieval sessions that contain search activities on the level of browsing. Our results show that a contextualisation of browsing significantly outperforms our baseline in terms of the position of the first clicked item in the result set. The mean rank of the first clicked document (measured as mean first relevant - MFR) was 4.52 using a non-contextualised ranking compared to 3.04 when re-ranking the result lists based on similarity to the previously viewed document. Furthermore, we observed that both contextual approaches show a noticeably higher click-through rate.     The association between dietary patterns and the novel inflammatory markers platelet-activating factor and lipoprotein-associated phospholipase A2: a systematic review

    CONTEXT: Atherosclerosis is a disease of chronic inflammation. Recent research has identified 2 novel inflammatory biomarkers: platelet-activating factor (PAF) and lipoprotein-associated phospholipase A(2) (Lp-PLA(2)). Diet has been proposed as a mediator of inflammation, but to date, the focus for these novel biomarkers has been on individual foods and nutrients rather than overall dietary patterns. OBJECTIVE: To systematically review the literature on the association between dietary patterns and PAF and Lp-PLA(2). DATA SOURCES: The PubMed, Embase, CINAHL, and Cochrane CENTRAL literature databases were searched. DATA ANALYSIS: Study quality was evaluated using the Quality Criteria Checklist. Sixteen studies (n = 4 observational and n = 12 interventional) were included and assessed for associations between dietary patterns and PAF and Lp-PLA(2). CONCLUSION: Study quality varied from neutral (n = 10) to positive (n = 6). Mediterranean, heart healthy, and vegetarian dietary patterns were associated with improved levels of PAF and Lp-PLA(2). Conversely, Western dietary patterns were less favorable. A range of well-established, healthier dietary patterns may lower inflammation and the risk of atherosclerosis. More well-designed studies are needed to confirm these findings and identify other dietary patterns that improve inflammation

    Propagating ice front induces gas bursts and ultrasonic acoustic emissions from freezing xylem

    Ice formation and propagation in the xylem of plants is a complex process. During freezing of xylem sap, gases dissolved in liquid sap are forced out of the ice lattice due to their low solubility in ice, and supersaturation of xylem sap as well as low water potential (Psi) are induced at the ice-liquid interface. Supersaturation of gases near the ice front may lead to bubble formation and potentially to cavitation and/or to burst of gases driven out from the branch. In this study, we investigated the origin and dynamics of freezing-related gas bursts and ultrasonic acoustic emissions (AEs), which are suggested to indicate cavitation. Picea abies (L.) H. Karst. and Salix caprea L. branch segments were exposed to frost cycles in a temperature test chamber, and CO2 efflux (indicating gas bursts) and AEs were recorded. On freezing, two-thirds of the observed gas bursts originated from the xylem and only one-third from the bark. Simultaneously with gas bursts, AEs were detected. Branch Psi affected both gas bursts and AEs, with high gas burst in saturated and dry samples but relevant AEs only in the latter. Repeated frost cycles led to decreasing gas burst volumes and AE activity. Experiments revealed that the expanding ice front in freezing xylem was responsible for observed gas bursts and AEs, and that branch Psi influenced both processes. Results also indicated that gas bursts and cavitation are independently induced by ice formation, though both may be relevant for bubble dynamics during freezing.Peer reviewe

    Quality of life, health and well-being, as well as the creative potential originating from positive experiences and interaction with our environment and interaction are pivotal human factors underlying a rich and productive social context. Respect and careful treatment of users, i.e., the citizen who will populate, co-create and co-operate in the scenarios, mandate overarching topics for developing user centered designschematas. Virtual Reality (VR) technologies have recently offered tools to anticipate the affordances of future scenarios. In the serious game VR-SenseCity (Paletta et al., 2017) we created tools for the imagination of affective and sensory experiences, in particular, referring to visual affordances that enable induction of imaginated sensory experiences in real urban environments. We investigated the potential for the motivation of elderly people, in particular, persons with dementia, to encounter their daily environment in positive attitude, with pleasant and aware sensory experiences, the purpose to improve their cognitive reserve (Stern, 2009). The functionalities were developed in cooperation with a Co-creation group of four elderly persons providing substantial critical and constructive feedback for the development of the toolbox

    H3K4me3 Is a Potential Mediator for Antiproliferative Effects of Calcitriol (1α,25(OH)2D3) in Ovarian Cancer Biology

    Abstract Posttranslational histone modification plays an important role in tumorigenesis. Histone modification is a dynamic response of chromatin to various signals, such as the exposure to calcitriol (1α,25(OH)2D3). Recent studies suggested that histone modification levels could be used to predict patient outcomes in various cancers. Our study evaluated the expression level of histone 3 lysine 4 trimethylation (H3K4me3) in a cohort of 156 epithelial ovarian cancer (EOC) cases by immunohistochemical staining and analyzed its correlation to patient prognosis. The influence of 1α,25(OH)2D3 on the proliferation of ovarian cancer cells was measured by BrdU proliferation assay in vitro. We could show that higher levels of H3K4me3 were correlated with improved overall survival (median overall survival (OS) not reached vs. 37.0 months, p = 0.047) and identified H3K4me3 as a potential prognostic factor for the present cohort. Ovarian cancer cell 1α,25(OH)2D3 treatment induced H3K4me3 protein expression and exhibited antiproliferative effects. By this, the study suggests a possible impact of H3K4me3 expression on EOC progression as well as its relation to calcitriol (1α,25(OH)2D3) treatment. These results may serve as an explanation on how 1α,25(OH)2D3 mediates its known antiproliferative effects. In addition, they further underline the potential benefit of 1α,25(OH)2D3 supplementation in context of ovarian cancer care

    The G-Protein-Coupled Estrogen Receptor (GPER) Regulates Trimethylation of Histone H3 at Lysine 4 and Represses Migration and Proliferation of Ovarian Cancer Cells In Vitro

    Histone H3 lysine 4 trimethylation (H3K4me3) is one of the most recognized epigenetic regulators of transcriptional activity representing, an epigenetic modification of Histone H3. Previous reports have suggested that the broad H3K4me3 domain can be considered as an epigenetic signature for tumor-suppressor genes in human cells. G-protein-coupled estrogen receptor (GPER), a new membrane-bound estrogen receptor, acts as an inhibitor on cell growth via epigenetic regulation in breast and ovarian cancer cells. This study was conducted to evaluate the relationship of GPER and H3K4me3 in ovarian cancer tissue samples as well as in two different cell lines (Caov3 and Caov4). Silencing of GPER by a specific siRNA and two selective regulators with agonistic (G1) and antagonistic (G15) activity were applied for consecutive in vitro studies to investigate their impacts on tumor cell growth and the changes in phosphorylated ERK1/2 (p-ERK1/2) and H3K4me3. We found a positive correlation between GPER and H3K4me3 expression in ovarian cancer patients. Patients overexpressing GPER as well as H3K4me3 had significantly improved overall survival. Increased H3K4me3 and p-ERK1/2 levels and attenuated cell proliferation and migration were observed in Caov3 and Caov4 cells via activation of GPER by G1. Conversely, antagonizing GPER activity by G15 resulted in opposite effects in the Caov4 cell line. In conclusion, interaction of GPER and H3K4me3 appears to be of prognostic significance for ovarian cancer patients. The results of the in vitro analyses confirm the biological rationale for their interplay and identify GPER agonists, such as G1, as a potential therapeutic approach for future investigations

    Xylem embolism and bubble formation during freezing suggest complex dynamics of pressure-tension in Betula pendula stems

    Freeze-thaw-induced embolism is a key limiting factor for perennial plants in frost-exposed environments. Gas bubbles are formed during freezing, when the low chemical potential of the ice reaches a critical cavitation threshold and expand during thawing. However, when water freezes, its volume increases by 9%, generating local pressures, which can limit the formation of bubbles. To characterize local dynamic of pressure-tension and physical state of the sap during freeze-thaw cycles, we simultaneously used ultrasonic acoustic emissions analysis and synchrotron-based High Resolution Computed Tomography on the diffuse-porous species Betula pendula. Visualization of individual air-filled vessels was performed to measure freeze-thaw induced embolism after successive freeze-thaw cycles down to -10C or -20C during the leafy and the leafless periods. We also measured the distribution of gas bubbles and made additional continuous monitoring of embolism spreading using a dedicated cooling system that allowed X-ray scanning during freezing and thawing. Experiments confirmed that ultrasonic emissions occurred after the onset of ice formation, together with bubble formation, whereas the development of embolism took place after thawing in all cases. The pictures of frozen tissues indicated that upon freezing the balance between negative pressure generated by the low water potential of the ice and the positive pressure induced by the volumetric increase of ice can provoke inward flow from the cell wall toward the lumen of the vessels. We found no evidence that wider vessels within a tissue were more prone to embolism although the occurrence of gas bubbles in larger conduits would make them prone to earlier embolism. These results highlight the need to monitor local pressure as well as ice and air distribution during xylem freezing to understand the mechanism leading to frost-induced embolism.Comment: 41 pages, 7 figures, 4 supplementary figure

    Subcellular distribution of thyroid hormone receptor beta in ovarian cancer

    Background: Since the most well-known function of thyroid hormone receptors (TRs) relies on their ability to act as ligand-activated transcription factors, their subcellular localization has been recognized to be relevant for their biological meaning. The current study aimed to determine the prevalence and subcellular distribution of TR beta and TR beta-1 in ovarian cancer (OC). Methods: Tissue was collected from 153 patients that had undergone surgery due to OC at the Department of Obstetrics and Gynaecology of the Ludwig-Maximilians-University Munich. Immunohistochemistry detecting TR beta and TR beta-1 was performed. Staining signals were quantified and tested for association with clinico-pathological parameters including overall survival (OS). Results: The subcellular distribution of TR beta and TR beta-1 differed among histologic subtypes, grade and FIGO stage. TR beta positivity was strongly linked to shortened overall survival (p < 0.001). Strikingly, this shortened OS was mainly attributed to those cases showing complete (p = 0.005) or incomplete shift of TR beta to the cytoplasm (p < 0.001). Significance was lost in multivariate testing. Conclusions: Cytoplasmatic localization of TR beta was associated with reduced OS, at least in univariate analysis. Since TRs have long been supposed to mainly function via the regulation of gene transcription in the nucleus, cytoplasmatic shifting might be interpreted as a regulator of their activity

    Cytoplasmic versus nuclear THR alpha expression determines survival of ovarian cancer patients

    PURPOSE Thyroid hormone receptors (THR) have manifold functions and are involved in the carcinogenesis of several tumor types. Within this study, we aimed to investigate the expression pattern (nuclear versus cytoplasmic) of the THR alpha and its impact on patients survival in ovarian cancer (OvCa). METHODS The presence of the thyroid hormone receptors THR\textgreeka, THR\textgreeka1 and -~2 was investigated in 156 ovarian cancer samples using immunohistochemistry (IHC) using semi-quantitative immunoreactivity (IR) scores and correlated with clinical, pathological data, subtype of ovarian cancer, clinical data, staining of 20 already described OvCa marker proteins and overall survival (OS). RESULTS Among all subtypes of OvCa, clear cell carcinomas showed the highest THR\textgreeka expression. Furthermore, nuclear THR\textgreeka was associated with a reduced survival in this subtype. However, nuclear expressed THR\textgreeka1 turned out to be a positive prognosticator for all subtypes of OvCa patients. Nuclear THR\textgreeka2 is a positive prognosticator for OvCa patients of the serous subtype. In contrast, cytoplasmic expression THR\textgreeka2 was associated with a reduced OS in all subtypes of OvCa patients; while, cytoplasmic expression of THR\textgreeka1 is associated with reduced OS in mucinous OvCa patients only. In addition, THR\textgreeka expression correlates with gonadotropin receptors, steroid hormone receptors, TA-MUC1 and glycodelin. CONCLUSION Depending on nuclear or cytoplasmic expression, our study shows that THR\textgreeka and its isoforms 1 and 2 provide different prognostic information for ovarian cancer patients. Further investigations should analyze if THRs may represent new endocrine targets for the treatment of ovarian cancer
