Cryptococcus neoformans Thermotolerance to Avian Body Temperature Is Sufficient For Extracellular Growth But Not Intracellular Survival In Macrophages
Cryptococcus neoformans is a fatal fungal pathogen of humans that efficiently parasitises macrophages. Birds can be colonised by cryptococci and can transmit cryptococcosis to humans via inhalation of inoculated bird excreta. However, colonisation of birds appears to occur in the absence of symptomatic infection. Here, using a pure population of primary bird macrophages, we demonstrate a mechanism for this relationship. We find that bird macrophages are able to suppress the growth of cryptococci seen in mammalian cells despite C. neoformans being able to grow at bird body temperature, and are able to escape from bird macrophages by vomocytosis. A small subset of cryptococci are able to adapt to the inhibitory intracellular environment of bird macrophages, exhibiting a large cell phenotype that rescues growth suppression. Thus, restriction of intracellular growth combined with survival at bird body temperature explains the ability of birds to efficiently spread C. neoformans in the environment whilst avoiding systemic disease
Experimental investigation on residual stresses in heavy wide flange QST steel sections
This paper presents the experimental results of residual stress measurements conducted on heavy wide flange quenched and self-tempered (QST) steel sections which have been developed by ArcelorMittal under the proprietary name HISTAR (HIgh-STrength ARcelorMittal). These sections are often applied in high-rise buildings, trusses or offshore structures and combine high strength with good toughness and weldability. The experiments are part of a larger study to arrive at buckling curves for these members as they are currently not provided by the European code. Two different sections with flange thicknesses greater than 100 mm are investigated and two types are examined: the stocky HD and more slender HL type. The sectioning method is adopted for measuring the residual stresses. It is found that both types display compres-sive residual stresses at the flange tips and the web and tensile residual stresses at the web-to-flange junc-tions. In absolute sense the residual stresses are greater in the HL type. From the experimental results a residual stress model is derived which can serve as the initial stress state of a heavy HISTAR section in non-linear finite element analyses
Eroding market stability by proliferation of financial instruments
We contrast Arbitrage Pricing Theory (APT), the theoretical basis for the
development of financial instruments, with a dynamical picture of an
interacting market, in a simple setting. The proliferation of financial
instruments apparently provides more means for risk diversification, making the
market more efficient and complete. In the simple market of interacting traders
discussed here, the proliferation of financial instruments erodes systemic
stability and it drives the market to a critical state characterized by large
susceptibility, strong fluctuations and enhanced correlations among risks. This
suggests that the hypothesis of APT may not be compatible with a stable market
dynamics. In this perspective, market stability acquires the properties of a
common good, which suggests that appropriate measures should be introduced in
derivative markets, to preserve stability.Comment: 26 pages, 8 figure
Women, anger, and aggression an interpretative phenomenological analysis
This study reports a qualitative phenomenological investigation of anger and anger-related aggression in the context of the lives of individual women. Semistructured interviews with five women are analyzed using interpretative phenomenological analysis. This inductive approach aims to capture the richness and complexity of the lived experience of emotional life. In particular, it draws attention to the context-dependent and relational dimension of angry feelings and aggressive behavior. Three analytic themes are presented here: the subjective experience of anger, which includes the perceptual confusion and bodily change felt by the women when angry, crying, and the presence of multiple emotions; the forms and contexts of aggression, paying particular attention to the range of aggressive strategies used; and anger as moral judgment, in particular perceptions of injustice and unfairness. The authors conclude by examining the analytic observations in light of phenomenological thinking
Lambda^0 polarization as a probe for production of deconfined matter in ultra-relativistic heavy-ion collisions
We study the polarization change of Lambda^0's produced in ultra-relativistic
heavy-ion collisions with respect to the polarization observed in proton-proton
collisions as a signal for the formation of a Quark-Gluon Plasma (QGP).
Assuming that, when the density of participants in the collision is larger than
the critical density for QGP formation, the Lambda^0 production mechanism
changes from recombination type processes to the coalescence of free valence
quarks, we find that the Lambda^0 polarization depends on the relative
contribution of each process to the total number of Lambda^0's produced in the
collision. To describe the polarization of Lambda^0's in nuclear collisions for
densities below the critical density for the QGP formation, we use the
DeGrand-Miettinen model corrected for the effects introduced by multiple

A glucuronoxylomannan epitope exhibits serotype-specific accessibility and redistributes towards the capsule surface during Titanisation of the fungal pathogen Cryptococcus neoformans.
Disseminated infections with the fungal species Cryptococcus neoformans or, less frequently, C. gattii, are an important cause of mortality in immunocompromised individuals. Central to the virulence of both species is an elaborate polysaccharide capsule that consists predominantly of glucuronoxylomannan (GXM). Due to its abundance, GXM is an ideal target for host antibodies, and several monoclonal antibodies (mAbs) have previously been derived using purified GXM or whole capsular preparations as antigen. In addition to their application in the diagnosis of cryptococcosis, anti-GXM mAbs are invaluable tools for studying capsule structure. In this study, we report the production and characterisation of a novel anti-GXM mAb, Crp127, that unexpectedly reveals a role for GXM remodelling during the process of fungal Titanisation. We show that Crp127 recognises a GXM epitope in an O-acetylation dependent, but xylosylation-independent, manner. The epitope is differentially expressed by the four main serotypes of Cryptococcus neoformans and gattii, is heterogeneously expressed within clonal populations of C. gattii serotype B strains and is typically confined to the central region of the enlarged capsule. Uniquely, however, this epitope redistributes to the capsular surface in Titan cells, a recently characterised morphotype where haploid 5 μm cells convert to highly polyploid cells >10 μm with distinct but poorly understood capsular characteristics. Titans are produced in the host lung and critical for successful infection. Crp127 therefore advances our understanding of cryptococcal morphological change and may hold significant potential as a tool to differentially identify cryptococcal strains and subtypes
Epidemic Enhancement in Partially Immune Populations
We observe that a pathogen introduce/pmcdata/journal/plosone/2-2007/1/ingest/pmcmod/sgml/pone.0000165.xmld into a population containing individuals with acquired immunity can result in an epidemic longer in duration and/or larger in size than if the pathogen were introduced into a naive population. We call this phenomenon “epidemic enhancement,” and use simple dynamical models to show that it is a realistic scenario within the parameter ranges of many common infectious diseases. This finding implies that repeated pathogen introduction or intermediate levels of vaccine coverage can lead to pathogen persistence in populations where extinction would otherwise be expected
Agricultural practices drive elevated rates of topsoil decline across Kenya, but terracing and reduced tillage can reverse this
As agricultural land area increases to feed an expanding global population, soil erosion will likely accelerate, generating unsustainable losses of soil and nutrients. This is critical for Kenya where cropland expansion and nutrient loading from runoff and erosion is contributing to eutrophication of freshwater ecosystems and desertification. We used the Revised Universal Soil Loss Equation (RUSLE) to predict soil erosion rates under present land cover and potential natural vegetation nationally across Kenya. Simulating natural vegetation conditions allows the degree to which erosion rates are elevated under current land use practices to be determined. This methodology exploits new digital soil maps and two vegetation cover maps to model topsoil (top 20 cm) erosion rates, lifespans (the mass of topsoil divided by erosion rate), and lateral nutrient fluxes (nutrient concentration times erosion rate) under both scenarios. We estimated the mean soil erosion rate under current land cover at ~5.5 t ha−1 yr−1, ~3 times the rate estimated for natural vegetation cover (~1.8 t ha−1 yr−1), and equivalent to ~320 Mt yr−1 of topsoil lost nationwide. Under present erosion rates, ~8.8 Mt, ~315 Kt, and ~ 110 Kt of soil organic carbon, nitrogen and phosphorous are lost from soil every year, respectively. Further, 5.3 % of topsoils (~3.1 Mha), including at >25 % of croplands, have short lifespans (<100 years). Additional scenarios were tested that assume combinations of terracing and reduced tillage practices were adopted on croplands to mitigate erosion. Establishing bench terraces with zoned tillage could reduce soil losses by ≥75 %; up to 87.1 t ha−1 yr−1. These reductions are comparable to converting croplands to natural vegetation, demonstrating most agricultural soils can be conserved successfully. Extensive long-term monitoring of croplands with terraces and reduced tillage established is required to verify the efficacy of these agricultural support practices as indicated by our modelling
Electronic and structural properties of superconducting MgB, CaSi and related compounds
We report a detailed study of the electronic and structural properties of the
39K superconductor \mgbtwo and of several related systems of the same family,
namely \mgalbtwo, \bebtwo, \casitwo and \cabesi. Our calculations, which
include zone-center phonon frequencies and transport properties, are performed
within the local density approximation to the density functional theory, using
the full-potential linearized augmented plane wave (FLAPW) and the
norm-conserving pseudopotential methods. Our results indicate essentially
three-dimensional properties for these compounds; however, strongly
two-dimensional -bonding bands contribute significantly at the Fermi
level. Similarities and differences between \mgbtwo and \bebtwo (whose
superconducting properties have not been yet investigated) are analyzed in
detail. Our calculations for \mgalbtwo show that metal substitution cannot be
fully described in a rigid band model. \casitwo is studied as a function of
pressure, and Be substitution in the Si planes leads to a stable compound
