    Space-based geoengineering: challenges and requirements

    The prospect of engineering the Earth's climate (geoengineering) raises a multitude of issues associated with climatology, engineering on macroscopic scales, and indeed the ethics of such ventures. Depending on personal views, such large-scale engineering is either an obvious necessity for the deep future, or yet another example of human conceit. In this article a simple climate model will be used to estimate requirements for engineering the Earth's climate, principally using space-based geoengineering. Active cooling of the climate to mitigate anthropogenic climate change due to a doubling of the carbon dioxide concentration in the Earth's atmosphere is considered. This representative scenario will allow the scale of the engineering challenge to be determined. It will be argued that simple occulting discs at the interior Lagrange point may represent a less complex solution than concepts for highly engineered refracting discs proposed recently. While engineering on macroscopic scales can appear formidable, emerging capabilities may allow such ventures to be seriously considered in the long term. This article is not an exhaustive review of geoengineering, but aims to provide a foretaste of the future opportunities, challenges, and requirements for space-based geoengineering ventures

    Quantum dot labelling of adenovirus allows for highly sensitive single cell flow and imaging cytometry

    A quantum dot method for highly efficient labelling of single adenoviral particles is developed. The technique has no impact on viral fitness and allows the imaging and tracking of virus binding and internalisation events using a variety of techniques including imaging cytometry and confocal microscopy. The method is applied to characterise the tropism of different adenoviral vectors

    ‘It’s just taking our souls back’: discourses of apartheid and race

    Although apartheid officially ended in 1994, the issue of race as a primary identity marker has continued to permeate many aspects of private and public life in post-apartheid South Africa. This paper seeks to understand how youth at two South African tertiary institutions position themselves in relation to race and the apartheid past. Our data include four focus group interviews from two universities, one which can be described as historically ‘black’ and the other as historically ‘white’. Given the complex nature of the data, we elected to use a combination of corpus linguistics and discourse analysis as our methodological approach. We explore how words such as black, white, coloured, they, we, us and them feature in the interviews. Our analysis shows that the positioning by the interviewees reflects a complexity and ambivalence that is at times contradictory although several broader discourse patterns can be distilled. In particular, we argue, that all groups employ a range of discursive strategies so as to resist being positioned in the historical positions of ‘victim’ and ‘perpetrator’.     Copper(II) Complexes with Tetradentate Piperazine-Based Ligands: DNA Cleavage and Cytotoxicity

    Five-coordinate Cu(II) complexes, [Cu(Ln)X]ClO4/PF6, where Ln = piperazine ligands bearing two pyridyl arms and X = ClO4− for Ln = L1 (1-ClO4), L2 (2-ClO4), L3 (3-ClO4), and L6 (6-ClO4) as well as [Cu(Ln)Cl]PF6 for Ln = L1 (1-Cl), L4 (4-Cl), and L5 (5-Cl) have been synthesized and characterized by spectroscopic techniques. The molecular structures of the last two complexes were determined by X-ray crystallography. In aqueous acetonitrile solutions, molar conductivity measurements and UV-VIS spectrophotometric titrations of the complexes revealed the hydrolysis of the complexes to [Cu(Ln)(H2O)]2+ species. The biological activity of the Cu(II) complexes with respect to DNA cleavage and cytotoxicity was investigated. At micromolar concentration within 2 h and pH 7.4, DNA cleavage rate decreased in the order: 1-Cl ≈ 1-ClO4 > 3-ClO4 ≥ 2-ClO4 with cleavage enhancements of up to 23 million. Complexes 4-Cl, 5-Cl, and 6-ClO4 were inactive. In order to elucidate the cleavage mechanism, the cleavage of bis(4-nitrophenyl)phosphate (BNPP) and reactive oxygen species (ROS) quenching studies were conducted. The mechanistic pathway of DNA cleavage depends on the ligand’s skeleton: while an oxidative pathway was preferable for 1-Cl/1-ClO4, DNA cleavage by 2-ClO4 and 3-ClO4 predominantly proceeds via a hydrolytic mechanism. Complexes 1-ClO4, 3-ClO4, and 5-Cl were found to be cytotoxic against A2780 cells (IC50 30–40 µM). In fibroblasts, the IC50 value was much higher for 3-ClO4 with no toxic effect

    Immunization with recombinant FliD confers protection against Helicobacter pylori infection in mice

    Nearly half of the world's population is infected with Helicobacter pylori. Clinical manifestations of this infection range from gastritis and peptic ulcers to gastric adenocarcinoma and lymphoma. Due to the emerging of antibiotic resistant strains and poor patient compliance of the antibiotic therapy, there is increasing interest in the development of a protective vaccine against H. pylori infection. The bacterial protein FliD forms a capping structure on the end of each flagellum which is critical to prevent depolymerization and structural degradation. In this study, the potential of FliD as a prospective H. pylori subunit vaccine was assessed. For this purpose, immunogenicity and protective efficacy of recombinant FliD (rFliD) from H. pylori was evaluated in C57BL/6 mice. Purified rFliD was formulated with different adjuvants and administered via subcutaneous or oral route. Subcutaneous immunization with rFliD elicited predominantly mixed Th1 and Th17 immune responses, with high titers of specific IgG 1 and IgG 2a . Splenocytes of immunized mice exhibited strong antigen-specific memory responses, resulting in the secretion of high amounts of IFN-γ and IL-17, and low levels of IL-4. Immunization with rFliD caused a significant reduction in H. pylori bacterial load relative to naïve control mice (p < 0.001), demonstrating a robust protective effect. Taken together, these results suggest that subcutaneous vaccination with rFliD formulated with CpG or Addavax could be considered as a potential candidate for the development of a subunit vaccine against H. pylori infection. © 2018 Elsevier Lt

    Macdonald Polynomials from Sklyanin Algebras: A Conceptual Basis for the pp-Adics-Quantum Group Connection

    We establish a previously conjectured connection between pp-adics and quantum groups. We find in Sklyanin's two parameter elliptic quantum algebra and its generalizations, the conceptual basis for the Macdonald polynomials, which ``interpolate'' between the zonal spherical functions of related real and pp\--adic symmetric spaces. The elliptic quantum algebras underlie the ZnZ_n\--Baxter models. We show that in the n \air \infty limit, the Jost function for the scattering of {\em first} level excitations in the ZnZ_n\--Baxter model coincides with the Harish\--Chandra\--like cc\--function constructed from the Macdonald polynomials associated to the root system A1A_1. The partition function of the Z2Z_2\--Baxter model itself is also expressed in terms of this Macdonald\--Harish\--Chandra\ cc\--function, albeit in a less simple way. We relate the two parameters qq and tt of the Macdonald polynomials to the anisotropy and modular parameters of the Baxter model. In particular the pp\--adic ``regimes'' in the Macdonald polynomials correspond to a discrete sequence of XXZ models.     Current whole-body MRI applications in the neurofibromatoses

    ObjectivesThe Response Evaluation in Neurofibromatosis and Schwannomatosis (REiNS) International Collaboration Whole-Body MRI (WB-MRI) Working Group reviewed the existing literature on WB-MRI, an emerging technology for assessing disease in patients with neurofibromatosis type 1 (NF1), neurofibromatosis type 2 (NF2), and schwannomatosis (SWN), to recommend optimal image acquisition and analysis methods to enable WB-MRI as an endpoint in NF clinical trials.MethodsA systematic process was used to review all published data about WB-MRI in NF syndromes to assess diagnostic accuracy, feasibility and reproducibility, and data about specific techniques for assessment of tumor burden, characterization of neoplasms, and response to therapy.ResultsWB-MRI at 1.5T or 3.0T is feasible for image acquisition. Short tau inversion recovery (STIR) sequence is used in all investigations to date, suggesting consensus about the utility of this sequence for detection of WB tumor burden in people with NF. There are insufficient data to support a consensus statement about the optimal imaging planes (axial vs coronal) or 2D vs 3D approaches. Functional imaging, although used in some NF studies, has not been systematically applied or evaluated. There are no comparative studies between regional vs WB-MRI or evaluations of WB-MRI reproducibility.ConclusionsWB-MRI is feasible for identifying tumors using both 1.5T and 3.0T systems. The STIR sequence is a core sequence. Additional investigation is needed to define the optimal approach for volumetric analysis, the reproducibility of WB-MRI in NF, and the diagnostic performance of WB-MRI vs regional MRI

    On unitarizability in the case of classical p-adic groups

    In the introduction of this paper we discuss a possible approach to the unitarizability problem for classical p-adic groups. In this paper we give some very limited support that such approach is not without chance.
