    Vector Competence of Ixodes scapularis and Ixodes ricinus (Acari: Ixodidae) for Three Genospecies of Borrelia burgdorferi

    The vector competence of 2 tick species, Ixodes ricinus (L.) and Ixodes scapularis Say, was determined and compared for 3 genospecies of Borrelia burgdorferi. The 3 genospecies of B. burgdorferi used in the following experiments were Borrelia burgdorferi sensu stricto (B-31 and B-31.D1 clone), Borrelia afzelii (strain Pgau.C3), and Borrelia garinii (strain VS286 and VSBP). Spirochetes from all 5 strains were inoculated intradermally into outbred mice; larval ticks of both species were subsequently fed on those mice and replete larvae were assayed for infection by culture in BSK-H media every 7 d for 4 wk. Infection frequencies in I. scapularis exposed to the 5 strains were as follows: B-31 (90%), B-31.D1 (83%), Pgau.C3 (87%), VS286 (10%), and VSBP (5%). The comparable infection frequencies for /. ricinus were B-31 (3%), B-31.D1 (3%), Pgau.C3 (90%), VS286 (5%), and VSBP (3%). Resultant nymphal /. scapularis successfully transmitted B-31, B-31.D1, Pgau.C3, and VS286 to outbred mice. /. ricinus nymphs transmitted Pgau.C3 and VS286. Both species failed to transmit strain VSB

    A Case-Control Study of Hantavirus Pulmonary Syndrome during an Outbreak in the Southwestern United States

    In May 1993, an outbreak of hantavirus pulmonary syndrome( HPS) occurred in the south-western United States. A case-control study determined risk factors for HPS. Seventeen case-patients were compared with 3 groups of controls: members of case-patient households( household controls), members of neighboring households( near controls), and members of randomly selected households ≥ 24 km away ( far controls). Investigators trapped more small rodents at case households than at near ( P = .03) or far control households( P = .02). After the number of small rodents was controlled for,case-patients were more likely than household controls to hand plow (odds ratio [OR], 12.3; 95% confidence interval [ CI], 1.1-143.0) or to clean feed storage areas (OR, 33.4; 95% CI, 1.7-666.0). Case-patients were more likely than near controls to plant( OR, 6.2; 95% CI, 1.1-34.0) and more likely than far controls to clean animal sheds( OR, 11.9;95% CI, 1.4-103.0). Peridomestic cleaning, agricultural activities, and an increased number of small rodents at the household were associated with HPS

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∼99% of the euchromatic genome and is accurate to an error rate of ∼1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    Plague is widespread among rodents and their fleas in the western United States, but most human cases occur in several definable, ecologically unique, and geographically limited high risk areas in the Southwest and Pacific Coast states. Control strategies to prevent human cases in high risk areas must vary from one epizootic focus to another, depending on such basic ecological factors as rodent/flea species involved; their distribution, abundance, seasonality, and relationship with the plague organism; climatic factors that affect transmission; the lifestyles of human residents; and others. This paper briefly summarizes preliminary results of a long-term program to define human risk and develop effective surveillance and control measures against plague in a north-central New Mexico plague focus

    Prevalence of Antibodies to \u3ci\u3eToxoplasma gondii\u3c/i\u3e in Wild and Domestic Animals of New Mexico, Arizona and Colorado

    Using the Sabin-Feldman dye test, sera from wild and domestic animals in New Mexico, Arizona and Colorado were tested for the prevalence of antibodies to Toxoplasma gondii. The prevalence of positive titers (≥ 1:8) in animals from these areas was: New Mexico (178 of 569, 31%), Arizona (11 of 56, 20%), and Colorado (2 of 7, 29%). The overall prevalence of antibodies to Toxoplasma was 30% (191 of 632). Nine of 17 fecal samples from wild zoo felines contained Toxoplasma-like oocysts which were inoculated per os and intraperitoneally into mice. Mice from six of these nine inoculations later showed positive dye test titers and tissues from five of these six groups had tissue cysts when examined histologically

    Serologic and Genetic Identification of Peromyscus maniculatus as the Primary Rodent Reservoir for a New Hantavirus in the Southwestern United States

    An outbreak of hantavirus pulmonary syndrome(H PS) in the southwestern United States was etiologically linked to a newly recognized hantavirus. Knowledge that hantaviruses are maintained in rodent reservoirs stimulated a field and laboratory investigation of 1696 small mammals of 31 species. The most commonly captured rodent, the deer mouse( Peromyscus maniculatus), had the highest antibody prevalence(3 %)to four hantavirus antigens. Antibody also was detected in 10 other species of rodent and in 1 species of rabbit. Reverse transcriptase-polymerase chain reaction( RT-PCR)products of hantavirus from rodent tissues were indistinguishable from those from human H PS patients. More than 96% of the seropositive P . maniculatus were positive by RT-PCR, suggesting chronic infection. Antibody prevalences were similar among P . maniculatus trapped from Arizona ( 33%), New Mexico (29%),and Colorado( 29%). The numeric dominance of P. maniculatus,the high prevalence of antibody, and the RT-PCR findings implicate this species as the primary rodent reservoir for a new hantavirus in the southwestern United State
