    Gold nanoparticle interactions with endothelial cells cultured under physiological conditions

    PEGylated gold nanoparticles (AuNPs) have an extended circulation time after intravenous injection in vivo and exhibit favorable properties for biosensing, diagnostic imaging, and cancer treatment. No impact of PEGylated AuNPs on the barrier forming properties of endothelial cells (ECs) has been reported, but recent studies demonstrated that unexpected effects on erythrocytes are observed. Almost all studies to date have been with static-cultured ECs. Herein, ECs maintained under physiological cyclic stretch and flow conditions and used to generate a blood–brain barrier model were exposed to 20 nm PEGylated AuNPs. An evaluation of toxic effects, cell stress, the release profile of pro-inflammatory cytokines, and blood–brain barrier properties showed that even under physiological conditions no obvious effects of PEGylated AuNPs on ECs were observed. These findings suggest that 20 nm-sized, PEGylated AuNPs may be a useful tool for biomedical applications, as they do not affect the normal function of healthy ECs after entering the blood stream

    An Exceptional Radio Flare in Markarian 421

    In September 2012, the high-synchrotron-peaked (HSP) blazar Markarian 421 underwent a rapid wideband radio flare, reaching nearly twice the brightest level observed in the centimeter band in over three decades of monitoring. In response to this event we carried out a five epoch centimeter- to millimeter-band multifrequency Very Long Baseline Array (VLBA) campaign to investigate the aftermath of this emission event. Rapid radio variations are unprecedented in this object and are surprising in an HSP BL Lac object. In this flare, the 15 GHz flux density increased with an exponential doubling time of about 9 days, then faded to its prior level at a similar rate. This is comparable with the fastest large-amplitude centimeter-band radio variability observed in any blazar. Similar flux density increases were detected up to millimeter bands. This radio flare followed about two months after a similarly unprecedented GeV gamma-ray flare (reaching a daily E>100 MeV flux of (1.2 +/- 0.7)x10^(-6) ph cm^(-2) s^(-1)) reported by the Fermi Large Area Telescope (LAT) collaboration, with a simultaneous tentative TeV detection by ARGO-YBJ. A cross-correlation analysis of long-term 15 GHz and LAT gamma-ray light curves finds a statistically significant correlation with the radio lagging ~40 days behind, suggesting that the gamma-ray emission originates upstream of the radio emission. Preliminary results from our VLBA observations show brightening in the unresolved core region and no evidence for apparent superluminal motions or substantial flux variations downstream.Comment: 5 pages, 8 figures.     O.5.1-6 Bucking the mid-life inactivity trend: a study of 40–64-year-olds' participation in Zwift, a real-time online exercise community, through a social identity lens

    Purpose Despite evidence suggesting a decline in the physical activity of middle-aged adults, 40–64-year-olds participation in group exercise is actually increasing. One potential reason for this increase in activity is the increase in a range of different online group exercise formats and contexts. Recent research suggests that people join, participate, and stay involved in, exercise groups that reflect who they are - a process of social identification. The purpose of this research is to qualitatively explore participants’ experiences and facilitators of social identification related to the online group exercise platform known as Zwift. Methods Zwift participants (n = 17) aged between 40-64 (midlife) (13 Men, 4 Women) were recruited into the study for three stages of data collection: (stage 1) an initial semi-structured interview exploring participant exercise history and use of Zwift, (stage 2) completing a two-week post exercise diary to capture their experiences of social identification after participation, (stage 3) a follow up final interview to explore topics raised over the first two data collection steps. This generated 23 hours of interviews, that were then transcribed and combined with the participant diaries resulting in 468 pages of data. Results Reflexive thematic analysis was used to identify themes from the data. The results from this analysis indicate the following themes from the data: (1) Online exercise as an enabling facilitator of exercise identity; (2). Online exercise provides exercise identity continuity or exercise identity creation depending on the exercisers, exercise history; (3) Online exercise identification via communities goes through phases of identification; (4) Two-way communication contributes to perceptions of identification phases of identification; and (5) Social identity leadership is demonstrated by online exercisers norms, behaviours and beliefs Conclusions Overall, this research highlights three key suggestions for mid-life physical activity promotion: (1) That facilitators and processes of social identification may be context specific; (2) That social identification processes should be considered in the design of online exercise; and (3) That online exercise that enables social identification should be considered as a vehicle of maintaining/ increasing exercise participation Funding The study was funded by the researcher

    Intrinsic brightness temperatures of blazar jets at 15 GHz

    We have developed a new Bayesian Markov Chain Monte Carlo method to deconvolve light curves of blazars into individual flares, including proper estimation of the fit errors. We use the method to fit 15GHz light curves obtained within the OVRO 40-m blazar monitoring program where a large number of AGN have been monitored since 2008 in support of the Fermi Gamma-Ray Space Telescope mission. The time scales obtained from the fitted models are used to calculate the variability brightness temperature of the sources. Additionally, we have calculated brightness temperatures of a sample of these objects using Very Long Baseline Array data from the MOJAVE survey. Combining these two data sets enables us to study the intrinsic brightness temperature distribution in these blazars at 15 GHz. Our preliminary results indicate that the mean intrinsic brightness temperature in a sample of 14 sources is near the equipartition brightness temperature of ~ 10^(11)K

    Evaluation of Rapid Syphilis Testing Using the Syphilis Health Check in Florida, 2015–2016

    The Syphilis Health Check (SHC) had low estimated specificity (91.5%) in one Florida county. We investigated use of SHC by a range of Florida publicly-funded programs between 2015 and 2016 to estimate specificity, positive predictive value (PPV), field staff acceptance, and impacts on programmatic outcomes. All reported SHC results were extracted from routinely collected program data. Field staff were surveyed about SHC’s utility. Analyses investigated differences between SHC and traditional syphilis testing outcomes. Of 3,630 SHC results reported, 442 were reactive; 92 (20.8%) had prior diagnoses of syphilis; 7 (1.6%) had no further testing. Of the remaining 343; 158 (46.0%) were confirmed cases, 168 (49.0%) were considered false-positive, and 17 (5.0%) were not cases but not clearly false-positive. Estimated specificity of SHC was 95.0%. Overall, 48.5% of positives became confirmed cases (PPV). PPV varied according to prevalence of syphilis in populations tested. Staff (90%) thought SHC helped identify new cases but expressed concern regarding discordance between reactive SHC and lab-based testing. Programmatic outcomes assessment showed shorter time to treatment and increased numbers of partners tested for the SHC group; these enhanced outcomes may better mitigate the spread of syphilis compared to traditional syphilis testing alone, but more research is needed

    The SDSS-III Baryon Oscillation Spectroscopic Survey: Quasar Target Selection for Data Release Nine

    The SDSS-III Baryon Oscillation Spectroscopic Survey (BOSS), a five-year spectroscopic survey of 10,000 deg^2, achieved first light in late 2009. One of the key goals of BOSS is to measure the signature of baryon acoustic oscillations in the distribution of Ly-alpha absorption from the spectra of a sample of ~150,000 z>2.2 quasars. Along with measuring the angular diameter distance at z\approx2.5, BOSS will provide the first direct measurement of the expansion rate of the Universe at z > 2. One of the biggest challenges in achieving this goal is an efficient target selection algorithm for quasars over 2.2 < z < 3.5, where their colors overlap those of stars. During the first year of the BOSS survey, quasar target selection methods were developed and tested to meet the requirement of delivering at least 15 quasars deg^-2 in this redshift range, out of 40 targets deg^-2. To achieve these surface densities, the magnitude limit of the quasar targets was set at g <= 22.0 or r<=21.85. While detection of the BAO signature in the Ly-alpha absorption in quasar spectra does not require a uniform target selection, many other astrophysical studies do. We therefore defined a uniformly-selected subsample of 20 targets deg^-2, for which the selection efficiency is just over 50%. This "CORE" subsample will be fixed for Years Two through Five of the survey. In this paper we describe the evolution and implementation of the BOSS quasar target selection algorithms during the first two years of BOSS operations. We analyze the spectra obtained during the first year. 11,263 new z>2.2 quasars were spectroscopically confirmed by BOSS. Our current algorithms select an average of 15 z > 2.2 quasars deg^-2 from 40 targets deg^-2 using single-epoch SDSS imaging. Multi-epoch optical data and data at other wavelengths can further improve the efficiency and completeness of BOSS quasar target selection.     High-excitation OH and H_2O lines in Markarian 231: the molecular signatures of compact far-infrared continuum sources

    The ISO/LWS far-infrared spectrum of the ultraluminous galaxy Mkn 231 shows OH and H_2O lines in absorption from energy levels up to 300 K above the ground state, and emission in the [O I] 63 micron and [C II] 158 micron lines. Our analysis shows that OH and H_2O are radiatively pumped by the far-infrared continuum emission of the galaxy. The absorptions in the high-excitation lines require high far-infrared radiation densities, allowing us to constrain the properties of the underlying continuum source. The bulk of the far-infrared continuum arises from a warm (T_dust=70-100 K), optically thick (tau_100micron=1-2) medium of effective diameter 200-400 pc. In our best-fit model of total luminosity L_IR, the observed OH and H2O high-lying lines arise from a luminous (L/L_IR~0.56) region with radius ~100 pc. The high surface brightness of this component suggests that its infrared emission is dominated by the AGN. The derived column densities N(OH)>~10^{17} cm^{-2} and N(H_2O)>~6x10^{16} cm^{-2} may indicate XDR chemistry, although significant starburst chemistry cannot be ruled out. The lower-lying OH, [C II] 158 micron, and [O I] 63 micron lines arise from a more extended (~350 pc) starburst region. We show that the [C II] deficit in Mkn 231 is compatible with a high average abundance of C+ because of an extreme overall luminosity to gas mass ratio. Therefore, a [C II] deficit may indicate a significant contribution to the luminosity by an AGN, and/or by extremely efficient star formation.Comment: 16 pages, 6 figures, accepted for publication in The Astrophysical Journa

    Who Meets Whom: Access and Lobbying During the Coalition Years

    In 2010, the incoming Coalition government announced that it would publish details of meetings between ministers and outside interests. We have collated and coded these data and, in this article, describe patterns of access between 2010 and 2015. In some respects, access is notably fragmented. No single organisation attends more than 2.5% of the 6292 meetings held by ministers. On the contrary, business, collectively, attends fully 45% of all meetings: more than twice the share of any other category of organisation. We also find evidence of distinctive policy communities characterised by high levels of access between particular interests and ministers within specific departments
