    Growth inhibition of bacterial isolates recovered from two types of Portuguese dry smoked sausages (chouriço)

    Potassium sorbate (PS), sodium benzoate (SB) and methyl p-hydroxybenzoate (MHB) were investigated as surface treatments for their ability to inhibit the growth of 18 isolates of spoilage and pathogenic bacteria from two types of Portuguese dry smoked sausages (Chouric o). MHB significantly inhibited the growth rate of 12 of the isolates (p < 0.05) whereas no effect was observed for four isolates of lactic acid bacteria, identified as Enterococcus faecium, Pediococcus acidilactici and Lactobacillus curvatus, and two isolates identified as Clostridium aminovalericum and Staphylococcus epidermidis. PS and SB had less influence on the bacterial growth rates.     Folatos em pseudocereais: efeito da cozedura e maltagem

    Os pseudocereais, amaranto, quinoa e trigo-sarraceno, são plantas com sementes que podem ser utilizadas e processadas como cereais. São sementes isentas de glúten, sendo por isso importante na dieta de indivíduos com hipersensibilidade a essa proteína, como os celíacos. O folato é um termo genérico para tetrahidrofolato (THF) (formas naturais). A deficiência nutricional de folato é comum em indivíduos com baixa ingestão, má absorção, e mulheres grávidas por aumento de exigência neste nutriente. Foram recolhidas amostras, de cada pseudocereal, durantes os anos de 2013 e 2014 (n=7). As amostras foram sujeitas a cozedura (tradicional e a vapor) e a maltagem. Para a determinação simultânea de formas naturais de folatos, ácido fólico (FA), 5-metiltetrahidrofolato (5-MTHF), tetrahidrofolato (THF), 5-formiltetrahidrofolato (5-CHOTHF) e 10-formiltetrahidrofolato (10-CHOTHF), foi utilizado um cromatógrafo líquido de ultra eficiência com deteção por espectrometria de massa num triplo quadrupolo (UPLC-MS/MS). As amostras foram sujeitas a um processo de extração tri-enzimático (α-amilase, protease e soro de rato) precedido de dissolução com tampão bicarbonato 0,005 M (1% ácido ascórbico, 0.5% DTT) a pH 7.2. As amostras antes de serem injetadas foram sujeitas a um processo de ultrafiltração com filtros de 5 KDa de poro. O método inclui a utilização de padrões internos de referência, marcados isotopicamente (13C5 FA e 13C5 5-MTHF). Foi utilizada uma coluna AcQuity HSS T3 1.8 µm 2.1 x 150 mm da Waters e como fase móvel, 0.1 % ácido fórmico (A) e acetonitrilo (B) em gradiente. A temperatura da coluna foi de 45 ˚C com fluxo de 0.35 mL/min. A forma predominante de folato encontrada em todas as amostras foi o 5-MTHF. A quinoa é o pseudocereal que apresenta os valores mais elevados da soma das diferentes formas (294 ± 20,8 mg /100 g), seguido respetivamente pelo amaranto (246 ± 30,9 mg/100 g) e trigo-sarraceno (139 ± 8,51 mg/100 g). O processo de maltagem apresenta para o amaranto e trigo- sarraceno a taxa de retenção mais elevada (100%), o mesmo se observou na quinoa para o processo de cozedura tradicional. Podemos concluir que o processo de maltagem não alteram significativamente o teor de folatos totais

Abstract It is well stated in the literature that medical treatment for peptic ulcer is based on a combination of proton pump inhibitors (PPIs) and antibiotics to eradicate Helicobacter pylori. This treatment is associated with a high rate of immediate success and a low rate of recurrence at 12 months, although it is not effective in all patients. Peptic ulcer (PU) perforation is a serious problem that leads to high complication and mortality rates. Surgical treatment, with its various possibilities, constitutes the ideal treatment. Surgical intervention in these cases, however, can be directed to treating the perforation alone, or it can offer definitive treatment of the ulcer itself. With the hope of establishing why such complications and mortality were seen in the patients in our hospital population, we gathered the facts about PU perforations and the types of surgery performed. We studied 210 consecutive patients (150 men, 60 women) who had undergone surgery at our hospital because of perforation between January 1, 1990 and December 31, 2000. The patients' median age was 53.0 +/- 20.6 years (men 47.7 +/- 17.3 years; women 66.3 +/- 22.0 years). Altogether, 86 patients had significant associated illnesses, 62 were admitted more than 24 hours after the perforation, and 25 were admitted in shock. We performed resections in 10 patients; 88 patients were treated by suturing the perforation with or without a patch of epiploon; and 112 underwent a troncular vagotomy with drainage (VT + Dr). A total of 21 patients died (10%). Significant risk factors that led to complications were identified by statistical studies. They were a perforation that had been present more than 24 hours, the coexistence of significant associated illnesses, and resection surgery. The significant risk factors that led to death were the presence of shock at admission, the coexistence of significant illnesses, and resection surgery. There was no statistically significant difference concerning morbidity and mortality between simple closure of the perforation and definitive surgery (VT + Dr). [email protected] Abstract Gastric cancer is a disease in which the main treatment is surgical extirpation. The modifications introduced in the surgical treatment over the last decades were accompanied by a clear increase of survival, which reaches global values of 61% at 5 years in Japan. One of the reasons that contribute to this improvement is early diagnosis of the lesions. In the period between January 1, 1990 and December 31, 1999 662 patients with gastric adenocarcinoma were treated in the Service of Surgery 1 of our hospital; 110 were refused surgical treatment. Of the resected patients, 91 (21.4%) were classified as early gastric cancer according to the definition of the Japanese Society of Digestive Endoscopy. There were 30 women and 61 men, with a median age of 60.2 +/- 15 years; 3 patients had a preoperative diagnosis of gastric ulcer; 2 others were operated without recent histology; and 1 patient was urgently resected for a bleeding ulcer. In all the remaining patients biopsy confirmed the presence of cancer (89%) or serious dysplasia (4.6%). The lesions had been distributed essentially in the medium 1/3 (48.3%) and distal 1/3 of the stomach. Subtotal gastrectomy was accomplished in 48 patients, total gastrectomy in 40, total desgastrogastrectomy in 3, and in 9 patients the surgery involved the spleen (8 patients) and the spleen and tail of the pancreas in 1 patient. Lymphadenectomy was not performed in 5 patients, lymph nodes by the first lymph node barrier were removed in 25 patients and by the second barrier in 61 patients (67%). Median tumor size was 26 +/- 1.8 mm. The lesion reached the mucosa in 46 patients and the mucosa and submucosa in 45. In 6 patients the removed lymph nodes were microscopically invaded (6.7%). Five patients died (5.7%). The median follow-up of the patients is 41 +/- 26 months; 7 patients died (8.1%) during this period; 4 died unequivocally of disease progression. The median survival of patients was 85% at 5 years and 80% at 10 years. In our series, survival was affected by the presence of invaded lymph nodes, not by the penetration in depth of the lesion or the size of the tumor. PMID: 11865370 [PubMed - indexed for MEDLINE

    Generation of two-mode nonclassical states and a quantum phase gate operation in trapped ion cavity QED

    We propose a scheme to generate nonclassical states of a quantum system, which is composed of the one-dimensional trapped ion motion and a single cavity field mode. We show that two-mode SU(2) Schr\"odinger-cat states, entangled coherent states, two-mode squeezed vacuum states and their superposition can be generated. If the vibration mode and the cavity mode are used to represent separately a qubit, a quantum phase gate can be implemented.Comment: to appear in PR

    Electrochemical sensing of ecstasy with electropolymerized molecularly imprinted poly(o-phenylenediamine) polymer on the surface of disposable screen-printed carbon electrodes

    This study demonstrates the ability of an electrochemical sensor based on molecularly imprinted polymers (MIPs) to selectively quantify 3,4-methylenedioxymethamphetamine (MDMA), also known as ecstasy, in biological samples. The device was constructed using ortho-phenylenediamine (o-PD) as the MIP’s building monomer at the surface of a screen-printed carbon electrode (SPCE). The step-by-step construction of the SPCE-MIP sensor was characterized by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Density functional theory (DFT) calculations and modelling were performed not only to understand template-monomer interaction but also to comprehend which possible polymer structure - linear or ramified poly(o-PD) – indeed interacts with the analyte. The prepared sensor worked by directly measuring the MDMA oxidation signal through square-wave voltammetry (SWV) after an incubation period of 10 min. Several parameters were optimized, such as the monomer/template ratio, the number of electropolymerization scanning cycles, and the incubation period, to obtain the best sensing efficiency. Optimized sensors exhibited suitable selectivity, repeatability (2.6%), reproducibility (7.7%) and up to one month of stable response. A linear range up to 0.2 mmol L−1 was found with an r2 of 0.9990 and a limit of detection (LOD) and quantification (LOQ) of 0.79 and 2.6 μmol L−1 (0.15 and 0.51 μg mL−1), respectively.     Galactic Halos of Fluid Dark Matter

    Dwarf spiral galaxies - and in particular the prototypical DDO 154 - are known to be completely dominated by an unseen component. The putative neutralinos - so far the favored explanation for the astronomical dark matter - fail to reproduce the well measured rotation curves of those systems because these species tend to form a central cusp whose presence is not supported by observation. We have considered here a self-coupled charged scalar field as an alternative to neutralinos and investigated whether a Bose condensate of that field could account for the dark matter inside DDO 154 and more generally inside dwarf spirals. The size of the condensate turns out to be precisely determined by the scalar mass m and self-coupling lambda of the field. We find actually that for m^4 / lambda = 50 - 75 eV^4, the agreement with the measurements of the circular speed of DDO 154 is impressive whereas it lessens for larger systems. The cosmological behavior of the field is also found to be consistent - yet marginally - with the limits set by BBN on the effective number of neutrino families. We conclude that classical configurations of a scalar and self-coupled field provide a possible solution to the astronomical dark matter problem and we suggest further directions of research.Comment: 20 pages, 7 figures; one reference added, version to be published in PR

    Generation of arbitrary two dimensional motional state of a trapped ion

    We present a scheme to generate an arbitrary two-dimensional quantum state of motion of a trapped ion. This proposal is based on a sequence of laser pulses, which are tuned appropriately to control transitions on the sidebands of two modes of vibration. Not more than (M+1)(N+1)(M+1)(N+1) laser pulses are needed to generate a pure state with upper phonon number MM and NN in the xx and yy direction respectively.Comment: to appear in PR
