    Word segmentation of Vietnamese texts: a comparison of approaches

    International audienceWe present in this paper a comparison between three segmentation systems for the Vietnamese language. Indeed, the majority of Vietnamese words is built by semantic composition from about 7,000 syllables, that also have a meaning as isolated words. So the identification of word boundaries in a text is not a simple task, and ambiguities often appear. Beyond the presentation of the tested systems, we also propose a standard definition for word segmentation in Vietnamese, and introduce a reference corpus developed for the purpose of evaluating such a task. The results observed confirm that it can be relatively well treated by automatic means, although a solution needs to be found to take into account out-of-vocabulary words

    Experimental investigation of mechanical and fracture properties of offshore wind monopile weldments: SLIC interlaboratory test results

    S355 structural steel is commonly used in fabrication of offshore structures including offshore wind turbine monopiles. Knowledge of mechanical and fracture properties in S355 weldments and the level of scatter in these properties is extremely important for ensuring the integrity of such structures through engineering critical assessment. An interlaboratory test programme was created to characterise the mechanical and fracture properties of S355 weldments, including the base metal, heat‐affected zone, and the weld metal, extensively. Charpy impact tests, chemical composition analysis, hardness tests, tensile tests, and fracture toughness tests have been performed on specimens extracted from each of the 3 material microstructures. The experimental test results from this project are presented in this paper, and their importance in structural integrity assessment of offshore wind turbine monopiles has been discussed. The results have shown a decreasing trend in the Charpy impact energy and Jmax values with an increase in yield stress from base metal to heat‐affected zone to weld metal. Moreover, the JIC fracture toughness value in the heat‐affected zone and weld metal is on average around 60% above and 40% below the base metal value, respectively. In addition, the average Charpy impact energy value in the heat‐affected zone and weld metal is around 5% and 30% below the base metal value, respectively. The effects of mechanical and fracture properties on the critical crack size estimates have been investigated, and the results are discussed concerning the impact of material properties on structural design and integrity assessment of monopiles

    A secretagogin locus of the mammalian hypothalamus controls stress hormone release

Although corticotropin-releasing hormone (CRH), produced by parvocellular neurons of the hypothalamic paraventricular nucleus (PVN) and released into the portal circulation at the median eminence, is known to prime downstream hormone release, the molecular mechanism regulating phasic CRH release remains poorly understood. Here, we find a cohort of parvocellular cells interspersed with magnocellular PVN neurons expressing secretagogin. Single-cell transcriptome analysis combined with protein interactome profiling identifies secretagogin neurons as a distinct CRH-releasing neuron population reliant on secretagogin's Ca2+ sensor properties and protein interactions with the vesicular traffic and exocytosis release machineries to liberate this key hypothalamic releasing hormone. Pharmacological tools combined with RNA interference demonstrate that secretagogin's loss of function occludes adrenocorticotropic hormone release from the pituitary and lowers peripheral corticosterone levels in response to acute stress. Cumulatively, these data define a novel secretagogin neuronal locus and molecular axis underpinning stress responsiveness.PostprintPeer reviewe

    Diggin' on u(biquitin):a novel method for the identification of physiological E3 ubiquitin ligase substrates

    The ubiquitin-proteasome system (UPS) plays a central role in maintaining protein homeostasis, emphasized by a myriad of diseases that are associated with altered UPS function such as cancer, muscle-wasting, and neurodegeneration. Protein ubiquitination plays a central role in both the promotion of proteasomal degradation as well as cellular signaling through regulation of the stability of transcription factors and other signaling molecules. Substrate specificity is a critical regulatory step of ubiquitination and is mediated by ubiquitin ligases. Recent studies implicate ubiquitin ligases in multiple models of cardiac diseases such as cardiac hypertrophy, atrophy, and ischemia/reperfusion injury, both in a cardioprotective and maladaptive role. Therefore, identifying physiological substrates of cardiac ubiquitin ligases provides both mechanistic insights into heart disease as well as possible therapeutic targets. Current methods identifying substrates for ubiquitin ligases rely heavily upon non-physiologic in vitro methods, impeding the unbiased discovery of physiological substrates in relevant model systems. Here we describe a novel method for identifying ubiquitin ligase substrates utilizing Tandem Ubiquitin Binding Entities (TUBE) technology, two-dimensional differential in gel electrophoresis (2-D DIGE), and mass spectrometry, validated by the identification of both known and novel physiological substrates of the ubiquitin ligase MuRF1 in primary cardiomyocytes. This method can be applied to any ubiquitin ligase, both in normal and disease model systems, in order to identify relevant physiological substrates under various biological conditions, opening the door to a clearer mechanistic understanding of ubiquitin ligase function and broadening their potential as therapeutic targets

    Dor crônica em pacientes esquizofrênicos: prevalência e características Chronic pain in schizophrenic patients: prevalence and characteristics

    A prevalência de dor crônica entre pacientes com transtornos psiquiátricos é, possivelmente, no mínimo igual à encontrada entre a população geral. Para estimar a prevalência de dor crônica em pacientes com esquizofrenia, comparar os grupos com e sem dor crônica e caracterizar a dor foi realizado um estudo transversal, com uma amostra probabilística de 205 pacientes adultos, com diagnóstico de esquizofrenia (idade média 37 anos; 65% homens; média de escolaridade de nove anos; 87% sem companheiro(a); 65% residentes com os pais), atendidos em hospital público do Município de São Paulo, Brasil. A prevalência de dor foi de 36,6%; a dor foi mais presente no abdômen (30,7%), seguida da dor de cabeça, face e boca (24%) e região lombar, sacra e cóccix (14,7%). Com relação à freqüência, 24% dos entrevistados referiram ter dor todos os dias. O tempo médio de dor foi de 41 meses, com intensidade moderada. A prevalência de dor crônica em pacientes esquizofrênicos foi semelhante à encontrada para a população geral, e o quadro álgico foi significativo em termos de duração, intensidade e freqüência dos episódios dolorosos.<br>Chronic pain may be at least as prevalent in psychiatric patients as in the general population. To estimate the prevalence of chronic pain in schizophrenic patients, compare the groups with and without chronic pain, and characterize the pain, a cross-sectional study was performed on a probabilistic sample of 205 adult patients with a diagnosis of schizophrenia (mean age 37 years; 65% men; mean schooling nine years; 87% single; 65% living with parents), treated at a public hospital in the city of São Paulo, Brazil. Prevalence of pain was 36.6%, and the most frequent sites were abdomen (30.7%), head, face, and mouth (24%), and lower back (14.7%). Twenty-four percent of patients reported feeling pain every day. Mean duration of pain was 41 months, with moderate intensity. Prevalence of chronic pain in schizophrenic patients was similar to that in the general population, and the clinical course was significant in terms of duration, intensity, and frequency
