    BPS states and their reductions

    We develop a method to identify the BPS states in the Hilbert space of a supersymmetric field theory on a generic curved space which preserves at least two real supercharges. We also propose a one-to-one map between BPS states in d-dimensional field theories and states that contribute to the supersymmetric partition function of a corresponding (d-1)-dimensional field theory. As an application we obtain the superconformal index on rounded and squashed three spheres, and we show a natural reduction of the respective indices to the three-dimensional exact partition functions. We discuss the validity of the correspondence both at the perturbative and at the non-perturbative level and exploit the idea to uplift the computation of the exact supersymmetric partition function on a general manifold to a higher dimensional index.Comment: 25 pages, 1 figure, typos fixe

    Liquiritigenin reduces osteoclast activity in zebrafish model of glucocorticoid-induced osteoporosis

    Drug and therapies currently used to treat human bone diseases have a lot of severe side effects. Liquiritigenin is a flavonoid extracted from Glycyrrhiza glabra roots which has been reported to have positive effects in vitro on osteoblasts activity and bone mineralization as well as inhibitory effect on osteoclasts differentiation and activity in vitro. The present study was aimed to evaluate the in vivo effects of liquiritigenin on bone structure and metabolism in physiological and pathological conditions using Danio rerio as experimental animal model. Treatments with liquiritigenin were performed on embryos to evaluate the osteogenesis during skeletal development. Other treatments were performed on adult fish affected by glucocorticoid-induced osteoporosis to assay the therapeutic potential of liquiritigenin in the reversion of bone-loss phenotype in scale model. Liquiritigenin treatment of zebrafish embryo significantly enhances the osteogenesis during development in a dose-dependent manner. In addition, liquiritigenin inhibits the formation of the osteoporotic phenotype in adult zebrafish model of glucocorticoid-induced osteoporosis preventing osteoclast activation in scales. Interestingly, liquiritigenin does not counteract the loss of osteoblastic activity in scales. The liquiritigenin exhibits in vivo anti-osteoporotic activity on adult fish scale model. It can be considered a good candidate to develop new drugs against osteoporosis

    Iterleukin 1 alpha is a marker of endothelial cellular senescent

    BACKGROUND: The functional changes associated with endothelial senescence may be involved in human aging and age-related vascular disorders. Since the inflammatory cytokine interleukin (IL-)1 inhibits endothelial growth, we evaluated the expression of IL-1α, IL-1β and their antagonist, the IL-1 receptor antagonist (IL-1ra), in endothelial in vitro senescence and quiescence. We also examined the expression of IL-1α in human senescent and progeric fibroblasts. RESULTS: We found that the overexpression of IL-1α specifically characterizes endothelial senescence. No modulation of this cytokine was observed in endothelial quiescence and in senescent or progeric human fibroblasts. The expression of IL-1β and IL-1ra was also assessed and found not to be affected by senescence. CONCLUSION: Our results indicate that a dysfunction of the cytokine network associates with aging and point to a specific role of IL-1α in endothelial senescence

    Abstract Endothelial cell differentiation is a crucial step in angiogenesis. Here we report the identification of EDF-1, a novel gene product that is down-regulated when endothelial cells are induced to differentiate in vitro. The cDNA encodingEDF-1 was isolated by RNA fingerprinting from human endothelial cells exposed to human immunodeficiency virus type 1 Tat, a viral protein known to be angiogenic. The deduced amino acid sequence of EDF-1 encodes a basic intracellular protein of 148 amino acids that is homologous to MBF1 (multiprotein-bridgingfactor 1) of the silkworm Bombyx mori and to H7, which is implicated in the early developmental events of Dictyostelium discoideum. Interestingly, human immunodeficiency virus type 1 Tat, which affects endothelial functions, and the phorbol ester 12-O-tetradecanoylphorbol-13-acetate and culture on fibrin gels, which promote endothelial differentiationin vitro, all down-regulate EDF-1 expression both at the RNA and protein levels. In addition, the inhibition of EDF-1 translation by an antisense anti-EDF-1 construct results in the inhibition of endothelial cell growth and in the transition from a nonpolar cobblestone phenotype to a polar fibroblast-like phenotype. These data suggest that EDF-1 may play a role in the regulation of human endothelial cell differentiation

    Neurological, psychological, and cognitive disorders in patients with chronic kidney disease on conservative and replacement therapy

    Chronic kidney disease (CKD) is a highly prevalent condition in the world. Neurological, psychological, and cognitive disorders, related to CKD, could contribute to the morbidity, mortality, and poor quality of life of these patients. The aim of this study was to assess the neurological, psychological, and cognitive imbalance in patients with CKD on conservative and replacement therapy. Seventy-four clinically stable patients affected by CKD on conservative therapy, replacement therapy (hemodialysis (HD), peritoneal dialysis (PD)), or with kidney transplantation (KT) and 25 healthy controls (HC), matched for age and sex were enrolled. Clinical, laboratory, and instrumental examinations, as renal function, inflammation and mineral metabolism indexes, electroencephalogram (EEG), psychological (MMPI-2, Sat P), and cognitive tests (neuropsychological tests, NPZ5) were carried out. The results showed a significant differences in the absolute and relative power of delta band and relative power of theta band of EEG (P=0.008, P<0.001, P=0.051), a positive correlation between relative power of delta band and C-reactive protein (CRP) (P< 0.001) and a negative correlation between estimated glomerular filtration rate (eGFR) (P<0.001) and 1,25-dihydroxyvitamin D3 (1,25-(OH)2D3) (P<0.001), in all the samples. Qualitative analysis of EEG showed alterations of Grade 2 (according to Parsons-Smith classification) in patients on conservative therapy, and Grade 2-3 in KT patients. The scales of MMPI-2 hysteria and paranoia, are significantly correlated with creatinine, eGFR, serum nitrogen, CRP, 1,25-(OH)2D3, intact parathyroid hormone (iPTH), phosphorus, and cynical and hysterical personality, are correlated with higher relative power of delta (P=0.016) and theta band (P= 0.016). Moreover, all NPZ5 scores showed a significant difference between the means of nephropathic patients and the means of the HC, and a positive correlation with eGFR, serum nitrogen, CRP, iPTH, and vitamin D. In CKD patients, simple and noninvasive instruments, as EEG, and cognitive-psychological tests, should be performed and careful and constant monitoring of renal risk factors, probably involved in neuropsychological complications (inflammation, disorders of mineral metabolism, electrolyte disorders, etc.), should be carried out. Early identification and adequate therapy of neuropsychological, and cognitive disorders, might enable a better quality of life and a major compliance with a probable reduction in the healthcare costs

    Facies and biostratigraphic analyses of the Contrada Diesi succession, cropping out along the northern slope of Mt. Magaggiaro (Sciacca, SW Sicily), provided new data on the Middle Jurassic-Early Cretaceous pelagic sedimentation in the Saccense domain. The richness in ammonites allowed the identification of Bathonian-Kimmeridgian Biozones and Subzones, while the Tithonian-Valanginian interval was defined mainly by calpionellids and calcareous nannofossils. Facies and microbiofacies analyses of the Jurassic-Cretaceous pelagic sediments of the area, together with ammonite, calpionellid and calcareous nannofossil integrated biostratigraphy, were very effective tools for comparison of biostratigraphic events. Many gaps in sedimentation were recognized, the most important spanning the middle and late Berriasian and part of the early Berriasian. The Contrada Diesi succession provides new litho-biostratigraphic data on the Saccense Domain. It suggests a high degree of internal variability tied to the irregular paleotopography of the carbonate platform substrate (Inici Fm.), derived from Early Jurassic tectonics. Gaps in sedimentation in the Contrada Diesi sections indicate that the environment of the Saccense Domain was characterized by a variable rate of sedimentation and energy changes

    Human natural killer cells and other innate lymphoid cells in cancer: Friends or foes?

    Innate lymphoid cells (ILC) including NK cells (cytotoxic) and the recently identified "helper" ILC1, ILC2 and ILC3, play an important role in innate defenses against pathogens. Notably, they mirror analogous T cell subsets, regarding the pattern of cytokine produced, while the timing of their intervention is few hours vs days required for T cell-mediated adaptive responses. On the other hand, the effectiveness of ILC in anti-tumor defenses is controversial. The relevance of NK cells in the control of tumor growth and metastasis has been well documented and they have been exploited in the therapy of high risk leukemia in the haploidentical hematopoietic stem cell transplantation setting. In contrast, the actual involvement of helper ILCs remains contradictory. Thus, while certain functional capabilities of ILC1 and ILC3 may favor anti-tumor responses, other functions could rather favor tumor growth, neo-angiogenesis, epithelial-mesenchymal transition and metastasis. In addition, ILC2, by secreting type-2 cytokines, are thought to induce a prevalent pro-tumorigenic effect. Finally, the function of both NK cells and helper ILCs may be inhibited by the tumor microenvironment, thus adding further complexity to the interplay between ILC and tumors

    Cretaceous tetrapod tracks from Italy: a treasure trove of exceptional biodiversity

    After about thirty years of investigation, the Cretaceous tetrapod track record from Italy has proved to be a ‘Rosetta Stone’ for improving understanding of the palaeogeographical and palaeoenvironmental evolution of the peri-Adriatic area. In the present contribution, we summarize current knowledge and diferent interpretations proposed on the basis of twelve ichnosites from northern, central and southern Italy. The tetrapod track record is represented by few ichnosites in the earliest Cretaceous, with the bulk of the record reported from carbonate platform deposits of the Aptian-Cenomanian interval and, in the Late Cretaceous, from a extensive-tracksite in Apulia preserving thousands of dinosaur footprints. On the whole, the ichnological diversity documented by the material indicates a high diversity of trackmakers, among which are sauropods, diferent kinds of theropods, ankylosaurs and hadrosaurs. The persistent occurrence of dinosaur footprints at diferent stratigraphic levels produced significant questions and constituted a dramatic constraint for the understanding of palaeogeographical and geodynamical evolution of the Mediterranean area during the Mesozoic, suggesting new and diferent interpretations that challenged previous reconstructions.Fil: Petti, Fabio Massimo.     We study dynamical supersymmetry breaking in vector-like superconformal N=1 gauge theories. We find appropriate deformations of the superpotential to overcome the problem of the instability of the non supersymmetric vacuum.
