Acute Malnutrition and Under-5 Mortality, Northeastern Part of India.
We assessed the prevalence of childhood acute malnutrition and under-five mortality rate (U5MR) in Darbhanga district, India, using a two-stage 49-cluster household survey. A total of 1379 households comprising 8473 people were interviewed. During a 90-day recall period, U5MR was 0.5 [95% confidence interval (CI), 0.2-1.4] per 10 000 per day. The prevalence of global acute malnutrition among 1405 children aged 6-59 months was 15.4% (NCHS) and 19.4% (2006 WHO references). This survey suggests that in Darbhanga district, the population is in a borderline food crisis with few food resources. Appropriate strategies should be developed to improve the overall nutritional and health status of children
Influence of hesperidin on the systemic and intestinal rat immune response
Polyphenols, widely found in edible plants, influence the immune system. Nevertheless, the immunomodulatory properties of hesperidin, the predominant flavanone in oranges, have not been deeply studied. To establish the effect of hesperidin on in vivo immune response, two different conditions of immune system stimulations in Lewis rats were applied. In the first experimental design, rats were intraperitoneally immunized with ovalbumin (OVA) plus Bordetella pertussis toxin and alum as the adjuvants, and orally given 100 or 200 mg/kg hesperidin. In the second experimental design, rats were orally sensitized with OVA together with cholera toxin and fed a diet containing 0.5% hesperidin. In the first approach, hesperidin administration changed mesenteric lymph node lymphocyte (MLNL) composition, increasing the TCRαβ+ cell percentage and decreasing that of B lymphocytes. Furthermore, hesperidin enhanced the interferon (IFN)-γ production in stimulated MLNL. In the second approach, hesperidin intake modified the lymphocyte composition in the intestinal epithelium (TCRγδ+ cells) and the lamina propria (TCRγδ+, CD45RA+, natural killer, natural killer T, TCRαβ+CD4+, and TCRαβ+CD8+ cells). Nevertheless, hesperidin did not modify the level of serum anti-OVA antibodies in either study. In conclusion, hesperidin does possess immunoregulatory properties in the intestinal immune response, but this effect is not able to influence the synthesis of specific antibodies
Development and characterization of an effective food allergy model in Brown Norway rats
Background: Food allergy (FA) is an adverse health effect produced by the exposure to a given food. Currently, there is no optimal animal model of FA for the screening of immunotherapies or for testing the allergenicity of new foods. Objective: The aim of the present study was to develop an effective and rapid model of FA in Brown Norway rats. In order to establish biomarkers of FA in rat, we compared the immune response and the anaphylactic shock obtained in this model with those achieved with only intraperitoneal immunization. Methods: Rats received an intraperitoneal injection of ovalbumin (OVA) with alum and toxin from Bordetella pertussis, and 14 days later, OVA by oral route daily for three weeks (FA group). A group of rats receiving only the i.p. injection (IP group) were also tested. Serum anti-OVA IgE, IgG1, IgG2a, IgG2b and IgA antibodies were quantified throughout the study. After an oral challenge, body temperature, intestinal permeability, motor activity, and mast cell protease II (RMCP-II) levels were determined. At the end of the study, anti-OVA intestinal IgA, spleen cytokine production, lymphocyte composition of Peyer's patches and mesenteric lymph nodes, and gene expression in the small intestine were quantified. Results: Serum OVA-specific IgG1, IgG2a and IgG2b concentrations rose with the i.p. immunization but were highly augmented after the oral OVA administration. Anti-OVA IgE increased twofold during the first week of oral OVA gavage. The anaphylaxis in both IP and FA groups decreased body temperature and motor activity, whereas intestinal permeability increased.Interestingly, the FA group showed a much higher RMCP II serum protein and intestinal mRNA expression. Conclusions: These results show both an effective and relatively rapid model of FA assessed by means of specific antibody titres and the high production of RMCP-II and its intestinal gene expression
Preclinical immunomodulation by the probiotic Bifidobacterium breve M-16V in early life
This study aimed to investigate the effect of supplementation with the probiotic Bifidobacterium breve M-16V on the maturation of the intestinal and circulating immune system during suckling. In order to achieve this purpose, neonatal Lewis rats were supplemented with the probiotic strain from the 6th to the 18th day of life. The animals were weighed during the study, and faecal samples were obtained and evaluated daily. On day 19, rats were euthanized and intestinal wash samples, mesenteric lymph node (MLN) cells, splenocytes and intraepithelial lymphocytes (IEL) were obtained. The probiotic supplementation in early life did not modify the growth curve and did not enhance the systemic immune maturation. However, it increased the proportion of cells bearing TLR4 in the MLN and IEL, and enhanced the percentage of the integrin αEβ7+ and CD62L+ cells in the MLN and that of the integrin αEβ7+ cells in the IEL, suggesting an enhancement of the homing process of naïve T lymphocytes to the MLN, and the retention of activated lymphocytes in the intraepithelial compartment. Interestingly, B. breve M-16V enhanced the intestinal IgA synthesis. In conclusion, supplementation with the probiotic strain B. breve M-16V during suckling improves the development of mucosal immunity in early lif
Second International Congress on Chocolate and Cocoa in Medicine Held in Barcelona, Spain, 25-26th September 2015.
In order to further our understanding of, and disseminate the latest findings on the healthy properties of cocoa and chocolate, the International Society of Chocolate and Cocoa in Medicine (ISCHOM) was founded in 2010 in Florence (http://ischom.com/ischom/). This Society aims to gather information and become a forum of discussion and debate on cocoa and chocolate, not only among researchers from around the world, but also to introduce the science involved and the latest findings to the public. Cultural and educational promotion of the benefits of cocoa and chocolate on human health is another of the Society's major concerns. Finally, ISCHOM provides information on developing healthy habits regarding the inclusion of cocoa and chocolate in our diet. In this context, after the first congress in Florence in 2014, ISCHOM held its second meeting in Barcelona, Spain, on 25 and 26 September 2015 (https://www.eiseverywhere.com/ehome/117409/263536/). By means of these annual meetings, the Society pursues the constant sharing and updating of current knowledge concerning the health properties of cocoa and chocolate. The event not only brings together international researchers, but also diverse companies in order to strengthen the knowledge in this field. About 90 delegates from 12 different countries attended the Society's second congress. It was organized into five scientific sessions with two lectures each. The sessions that took place in Barcelona were focused on different topics in order to shed some light on a wide range of valuable effects of chocolate and cocoa on our health. Some of the issues discussed were the role of chocolate and cocoa as cardioprotective agents and its health claims, their effects on metabolism and their own metabolism; the possible role of cocoa as a preventive therapy for diabetes and allergies; its influence on microbiota; and the beneficial effects of cocoa on the nervous system. The opening session outlined the scientific thought on this matter spiced with wit. The congress also included a special session focused on heritage and innovation in chocolate that delighted the audience. Furthermore, two discussion sessions of oral communications also took place. The first session related to assessing the intake of cocoa and its effect, the relationship between cocoa flavanols, cognitive performance and cerebral blood flow; and the effect of other bioactive compounds of cocoa, methylxanthines such as theobromine, and their outcomes on rat lymphoid tissues were ascertained. The second oral communication discussion session concerned the assessment of the antioxidant capacity and polyphenol content of different varieties of cocoa, how cocoa intake improves hepatic lipid metabolism in rats, and how flavanols present in cocoa may confer benefits by diminishing brain damage caused by strokes in mice. In addition, the poster session was exhibited during the entire congress showing state-of-the-art research about chocolate and cocoa in medicine for all the attendants

Effects of a cocoa diet on an intestinal inflammation model in rats
Cocoa is a rich source of fiber and flavonoids with recognized antioxidant and anti-inflammatory potential. The aim of this study was to evaluate the effects of a cocoa-enriched diet on rats with dextran sulphate sodium (DSS)-induced colitis. Wistar rats were fed with either a 5% cocoa diet or standard diet. Colon inflammation was induced by DSS in the drinking water: 5% for 6 days and 2% over the following 9 days. Colitis was assessed by body weight loss, stool consistency and blood presence in stools. A group of animals fed standard diet was treated with quercitrin (1 mg/kg) after colitis establishment. After 2 weeks of DSS treatment, the colon oxidative and inflammatory status and lymphocyte composition from blood and mesenteric lymph nodes (MLN) were assessed. The cocoa-fed group did not exhibit amelioration of clinical colitis but displayed higher antioxidant activity than the colitic reference group by the restoration of colon glutathione content and prevention of lipid peroxidation. The cocoa diet showed anti-inflammatory potential because it down-regulated serum TNF-alpha, colon iNOS activity and decreased colon cell infiltration. Lymphocyte composition in MLN was not modified by drinking DSS, but there was an increase in the proportion of NK and regulatory T cells in the blood. These changes were not modified by cocoa. In conclusion, cocoa intake may help to inhibit the negative oxidative effects consequent to colitis, although this action is not enough to abrogate the intestinal inflammation significantly
