    Identification and nucleotide sequences of mxaA, mxaC, mxaK, mxaL, and mxaD genes from Methylobacterium extorquens AM1

    The DNA sequence for a 4.4-kb HindIII-XhoI Methylobacterium extorquens AM1 DNA fragment that is known to contain three genes (mxaAKL) involved in incorporation of calcium into methanol dehydrogenase (I. W. Richardson and C. Anthony, Biochem. J. 287:709-7115, 1992) was determined. Five complete open reading frames and two partial open reading frames were found, suggesting that this region contains previously unidentified genes. A combination of sequence analysis, mutant complementation data, and gene expression studies showed that these genes correspond to mxaSACKLDorf1. Of the three previously unidentified genes (mxaC, mxaD, and orf1), mutant complementation studies showed that mxaC is required for methanol oxidation, while the function of the other two genes is still unknown

    Rumen fluke in Irish sheep: prevalence, risk factors and molecular identification of two paramphistome species

    peer-reviewedBackground Rumen flukes are trematode parasites found globally; in tropical and sub-tropical climates, infection can result in paramphistomosis, which can have a deleterious impact on livestock. In Europe, rumen fluke is not regarded as a clinically significant parasite, recently however, the prevalence of rumen fluke has sharply increased and several outbreaks of clinical paramphistomosis have been reported. Gaining a better understanding of rumen fluke transmission and identification of risk factors is crucial to improve the control of this parasitic disease. In this regard, a national prevalence study of rumen fluke infection and an investigation of associated risk factors were conducted in Irish sheep flocks between November 2014 and January 2015. In addition, a molecular identification of the rumen fluke species present in Ireland was carried out using an isolation method of individual eggs from faecal material coupled with a PCR. After the DNA extraction of 54 individual eggs, the nuclear fragment ITS-2 was amplified and sequenced using the same primers. Results An apparent herd prevalence of 77.3 % was determined. Several risk factors were identified including type of pasture grazed, regional variation, and sharing of the paddocks with other livestock species. A novel relationship between the Suffolk breed and higher FEC was reported for the first time. The predominant rumen fluke species found was C. daubneyi. Nevertheless, P. leydeni was unexpectedly identified infecting sheep in Ireland for the first time. Conclusions An exceptionally high prevalence of rumen fluke among Irish sheep flocks has been highlighted in this study and a more thorough investigation is necessary to analyse its economic impact. The isolation of individual eggs coupled with the PCR technique used here has proven a reliable tool for discrimination of Paramphistomum spp. This technique may facilitate forthcoming studies of the effects of paramphistomosis on livestock production. The most noteworthy finding was the identification of P. leydeni affecting sheep in Ireland, however further studies are required to clarify its implications. Also, a significant relationship between Suffolk breed and a heavier infection was found, which can be used as a starting point for future research on control strategies of rumen fluke infection.This study was funded by Irish Department of Agriculture, Food and the Marine research stimulus funding; project reference 13/ S/405

    Congenital infections as contributors to the onset of diabetes in children: A longitudinal study in the United States, 2001‐2017

    BackgroundMaternal infections during pregnancy, particularly with rubella virus, were reported to increase the risk of diabetes in children. Widespread vaccination has decreased the number of infants with congenital rubella syndrome in the United States, although it remains a problem in developing countries. Because vaccine hesitancy has recently increased, we investigated the association between congenital infections with subsequent diabetes risk in children in the United States.MethodsUsing data from a nationwide private health insurer for years 2001‐2017, 1 475 587 infants were followed for an average of 3.9 years (maximum 16.5 years). Information was obtained regarding congenital infections (rubella, cytomegalovirus, other congenital infections) and perinatal infections, as well as for the development of diabetes mellitus and diabetic ketoacidosis.ResultsThere were 781 infants with congenital infections and 73 974 with perinatal infections. Diabetes developed in 3334 children. The odds of developing diabetes for infants with congenital rubella infection were 12‐fold greater (P = .013) and, for infants with congenital cytomegalovirus infection, were 4‐fold greater (P = .011) than infants without congenital or perinatal infection. Infants with other congenital infections had 3‐fold greater odds of developing diabetes (P = .044). Results were similar for diabetes ketoacidosis. Infants with other perinatal infections had 49% greater odds of developing diabetes during the follow‐up period (P < .001).ConclusionCongenital and other perinatal infections are associated with elevated risks of developing diabetes mellitus during childhood.     Recolonizing wolves and mesopredator suppression of coyotes:impacts on pronghorn population dynamics

    Food web theory predicts that the loss of large carnivores may contribute toelevated predation rates and, hence, declining prey populations, through the process ofmesopredator release. However, opportunities to test predictions of the mesopredator releasehypothesis are rare, and the extent to which changes in predation rates influence preypopulation dynamics may not be clear due to a lack of demographic information on the preypopulation of interest. We utilized spatial and seasonal heterogeneity in wolf distribution andabundance to evaluate whether mesopredator release of coyotes (Canis latrans), resulting fromthe extirpation of wolves (Canis lupus) throughout much of the United States, contributes tohigh rates of neonatal mortality in ungulates. To test this hypothesis, we contrasted causes ofmortality and survival rates of pronghorn (Antilocapra americana) neonates captured at wolf-free and wolf-abundant sites in western Wyoming, USA, between 2002 and 2004. We thenused these data to parameterize stochastic population models to heuristically assess the impactof wolves on pronghorn population dynamics due to changes in neonatal survival. Coyotepredation was the primary cause of mortality at all sites, but mortality due to coyotes was 34%lower in areas utilized by wolves (P , 0.001). Based on simulation modeling, the realizedpopulation growth rate was 0.92 based on fawn survival in the absence of wolves, and 1.06 atsites utilized by wolves. Thus, wolf restoration is predicted to shift the trajectory of thepronghorn population from a declining to an increasing trend. Our results suggest thatreintroductions of large carnivores may influence biodiversity through effects on preypopulations mediated by mesopredator suppression. In addition, our approach, whichcombines empirical data on the population of interest with information from other datasources, demonstrates the utility of using simulation modeling to more fully evaluateecological theories by moving beyond estimating changes in vital rates to analyses ofpopulation-level impacts

    Applying rotorcraft modelling technology to renewable energy research

    The perceived need to reduce mankind's impact on the global climate motivates towards a future society in which a significant proportion of its energy needs will be extracted from the winds and the tides of the planet. This paper shows several examples of the application of Brown's Vorticity Transport Model, originally developed to perform simulations of helicopter aeromechanics and wake dynamics, to the analysis of the performance of renewable energy devices and their possible impact on the environment. Prediction of the loading on wind turbines introduces significant additional challenges to such a model, including the need to account fully for the effects of radial flow on blade stall. The wake-mediated aerodynamic interactions that occur within a wind farm can reduce its power output significantly, but this problem is very similar to that where the aerodynamic unsteadiness of the coupled wake of the main and tail rotors of a helicopter can result in significantly increased pilot workload. The helicopter-related problem of brownout, encountered during operations in desert conditions, has its analogue in the entrainment of sediment into the wakes of tidal turbines. In both cases it may be possible to ameliorate the influence of the rotor on its environment by careful and well-informed design. Finally, calculations of the distortion and dispersal of the exhaust plumes of a helicopter by the wake of its rotor allow insight into how wind turbines might interfere with the dispersal of pollutants from nearby industrial sites. These examples show how cross-disciplinary information transfer between the rotorcraft field and the renewable energy community is helping to develop the technologies that will be required by our future society, as well as helping to understand the environmental issues that might need to be faced as these technologies become more prevalent

    CCR4-bearing T cells participate in autoimmune diabetes.

    Chemokine receptor expression is exquisitely regulated on T cell subsets during the course of their migration to inflammatory sites. In the present study we demonstrate that CCR4 expression marks a pathogenic population of autoimmune T cells. CCR4 was found exclusively on memory CD4(+) T cells during the progression of disease in NOD mice. Cells expressing the CCR4 ligand TARC (thymus- and activation-regulated chemokine) were detected within infiltrated islets from prediabetic mice. Interestingly, neutralization of macrophage-derived chemokine (MDC) with Ab caused a significant reduction of CCR4-positive T cells within the pancreatic infiltrates and inhibited the development of insulitis and diabetes. Furthermore, enhanced recruitment of CCR4-bearing cells in NOD mice resulting from transgenic expression of MDC resulted in acceleration of clinical disease. Cumulatively, the results demonstrate that CCR4-bearing T cells participate in the development of such tissue-driven autoimmune reactions
