A p38MAPK/MK2 signaling pathway leading to redox stress, cell death and ischemia/reperfusion injury
Background
Many diseases and pathological conditions are characterized by transient or constitutive overproduction of reactive oxygen species (ROS). ROS are causal for ischemia/reperfusion (IR)-associated tissue injury (IRI), a major contributor to organ dysfunction or failure. Preventing IRI with antioxidants failed in the clinic, most likely due to the difficulty to timely and efficiently target them to the site of ROS production and action. IR is also characterized by changes in the activity of intracellular signaling molecules including the stress kinase p38MAPK. While ROS can cause the activation of p38MAPK, we recently obtained in vitro evidence that p38MAPK activation is responsible for elevated mitochondrial ROS levels, thus suggesting a role for p38MAPK upstream of ROS and their damaging effects.<p></p>
Results
Here we identified p38MAPKα as the predominantly expressed isoform in HL-1 cardiomyocytes and siRNA-mediated knockdown demonstrated the pro-oxidant role of p38MAPKα signaling. Moreover, the knockout of the p38MAPK effector MAPKAP kinase 2 (MK2) reproduced the effect of inhibiting or knocking down p38MAPK. To translate these findings into a setting closer to the clinic a stringent kidney clamping model was used. p38MAPK activity increased upon reperfusion and p38MAPK inhibition by the inhibitor BIRB796 almost completely prevented severe functional impairment caused by IR. Histological and molecular analyses showed that protection resulted from decreased redox stress and apoptotic cell death.<p></p>
Conclusions
These data highlight a novel and important mechanism for p38MAPK to cause IRI and suggest it as a potential therapeutic target for prevention of tissue injury.<p></p>
A pathway for Parkinson's Disease LRRK2 kinase to block primary cilia and Sonic hedgehog signaling in the brain
Parkinson’s disease-associated LRRK2 kinase phosphorylates multiple Rab GTPases, including Rab8A and Rab10. We show here that LRRK2 kinase interferes with primary cilia formation in cultured cells, human LRRK2 G2019S iPS cells and in the cortex of LRRK2 R1441C mice. Rab10 phosphorylation strengthens its intrinsic ability to block ciliogenesis by enhancing binding to RILPL1. Importantly, the ability of LRRK2 to interfere with ciliogenesis requires both Rab10 and RILPL1 proteins. Pathogenic LRRK2 influences the ability of cells to respond to cilia-dependent, Hedgehog signaling as monitored by Gli1 transcriptional activation. Moreover, cholinergic neurons in the striatum of LRRK2 R1441C mice show decreased ciliation, which will decrease their ability to sense Sonic hedgehog in a neuro-protective circuit that supports dopaminergic neurons. These data reveal a molecular pathway for regulating cilia function that likely contributes to Parkinson’s disease-specific pathology
TGF-b2 induction regulates invasiveness of theileria-transformed leukocytes and disease susceptibility
Theileria parasites invade and transform bovine leukocytes causing either East Coast fever (T. parva), or tropical theileriosis (T. annulata). Susceptible animals usually die within weeks of infection, but indigenous infected cattle show markedly reduced pathology, suggesting that host genetic factors may cause disease susceptibility. Attenuated live vaccines are widely used to control tropical theileriosis and attenuation is associated with reduced invasiveness of infected macrophages in vitro. Disease pathogenesis is therefore linked to aggressive invasiveness, rather than uncontrolled proliferation of Theileria-infected leukocytes. We show that the invasive potential of Theileria-transformed leukocytes involves TGF-b signalling. Attenuated live vaccine lines express reduced TGF-b2 and their invasiveness can be rescued with exogenous TGF-b. Importantly, infected macrophages from disease susceptible Holstein-Friesian (HF) cows express more TGF-b2 and traverse Matrigel with great efficiency compared to those from disease-resistant Sahiwal cattle. Thus, TGF-b2 levels correlate with disease susceptibility. Using fluorescence and time-lapse video microscopy we show that Theileria-infected, disease-susceptible HF macrophages exhibit increased actin dynamics in their lamellipodia and podosomal adhesion structures and develop more membrane blebs. TGF-b2-associated invasiveness in HF macrophages has a transcription-independent element that relies on cytoskeleton remodelling via activation of Rho kinase (ROCK). We propose that a TGF-b autocrine loop confers an amoeboid-like motility on Theileria-infected leukocytes, which combines with MMP-dependent motility to drive invasiveness and virulence
Systematic proteomic analysis of LRRK2-mediated Rab GTPase phosphorylation establishes a connection to ciliogenesis
We previously reported that Parkinson's disease (PD) kinase LRRK2 phosphorylates a subset of Rab GTPases on a conserved residue in their switch-II domains (Steger et al., 2016) (PMID: 26824392). Here, we systematically analyzed the Rab protein family and found 14 of them (Rab3A/B/C/D, Rab5A/B/C, Rab8A/B, Rab10, Rab12, Rab29, Rab35 and Rab43) to be specifically phosphorylated by LRRK2, with evidence for endogenous phosphorylation for ten of them (Rab3A/B/C/D, Rab8A/B, Rab10, Rab12, Rab35 and Rab43). Affinity enrichment mass spectrometry revealed that the primary ciliogenesis regulator, RILPL1 specifically interacts with the LRRK2-phosphorylated forms of Rab8A and Rab10, whereas RILPL2 binds to phosphorylated Rab8A, Rab10, and Rab12. Induction of primary cilia formation by serum starvation led to a two-fold reduction in ciliogenesis in fibroblasts derived from pathogenic LRRK2-R1441G knock-in mice. These results implicate LRRK2 in primary ciliogenesis and suggest that Rab-mediated protein transport and/or signaling defects at cilia may contribute to LRRK2-dependent pathologies
Outcome after resection of Adrenocortical Carcinoma liver metastases: a retrospective study
Background: Metastatic Adrenocortical Carcinoma (ACC) is a rare malignancy with a poor 5-year-survival rate (<15%). A surgical approach is recommended in selected patients if complete resection of distant metastasis can be achieved. To date there are only limited data on the outcome after surgical resection of hepatic metastases of ACC. Methods: A retrospective analysis of the German Adrenocortical Carcinoma Registry was conducted. Patients with liver metastases of ACC but without extrahepatic metastases or incomplete tumour resection were included. Results: Seventy-seven patients fulfilled these criteria. Forty-three patients underwent resection of liver metastases of ACC. Complete tumour resection (R0) could be achieved in 30 (69.8%). Median overall survival after liver resection was 76.1 months in comparison to 10.1 months in the 34 remaining patients with unresected liver metastases (p < 0.001). However, disease free survival after liver resection was only 9.1 months. Neither resection status (R0/R1) nor extent of liver resection were significant predictive factors for overall survival. Patients with a time interval to the first metastasis/recurrence (TTFR) of greater than 12 months or solitary liver metastases showed significantly prolonged survival. Conclusions: Liver resection in the case of ACC liver metastases can achieve long term survival with a median overall survival of more than 5 years, but disease free survival is short despite metastasectomy. Time to recurrence and single versus multiple metastases are predictive factors for the outcome
Photoelectric Emission from Interstellar Dust: Grain Charging and Gas Heating
We model the photoelectric emission from and charging of interstellar dust
and obtain photoelectric gas heating efficiencies as a function of grain size
and the relevant ambient conditions. Using realistic grain size distributions,
we evaluate the net gas heating rate for various interstellar environments, and
find less heating for dense regions characterized by R_V=5.5 than for diffuse
regions with R_V=3.1. We provide fitting functions which reproduce our
numerical results for photoelectric heating and recombination cooling for a
wide range of interstellar conditions. In a separate paper we will examine the
implications of these results for the thermal structure of the interstellar
medium. Finally, we investigate the potential importance of photoelectric
heating in H II regions, including the warm ionized medium. We find that
photoelectric heating could be comparable to or exceed heating due to
photoionization of H for high ratios of the radiation intensity to the gas
density. We also find that photoelectric heating by dust can account for the
observed variation of temperature with distance from the galactic midplane in

Fallstudien zur Adaption und Antizipation im österreichischen Fachhochschulsektor
"Mit der Etablierung des Fachhochschulwesens wurde im internationalen Vergleich nicht nur eine Ausbildungslücke des österreichischen Bildungssystems geschlossen, sondern zugleich auch ein neues Kapitel hinsichtlich der Abstimmung von Ausbildung und beruflicher Praxis eröffnet. Neben dem Hochschulniveau bildet vor allem die Praxisorientierung einen wesentlichen Eckpfeiler des Fachhochschulwesens. Um diese Praxisorientierung zu gewährleisten wurden innovative - dem österreichischen Bildungswesen bislang unbekannte - Instrumente wie z.B. die Bedarfs- und Akzeptanzstudien implementiert. Die Dynamik der wirtschaftlichen Entwicklung erfordert aber eine immer schnellere Anpassung an Veränderungen, weshalb sich die Frage stellt, auf welche Weise die einzelnen FH-Studiengänge abseits der obligatorischen Instrumente den Praxisbezug ihrer Ausbildung sicherstellen. Um diese Prozesse zu untersuchen werden in dieser Studie zwei ausgewählte Fachhochschulstudiengänge (Medientechnik und Design in Hagenberg, OÖ sowie Industrielle Elektronik in Kapfenberg, Steiermark) im Rahmen von Fallstudien vor allem hinsichtlich ihrer Praxis der Anpassung des Curriculums an den Qualifikationsbedarf der Wirtschaft bzw. ihres Systems zur Antizipation wirtschaftlicher Veränderungen analysiert und verglichen." As the ongoing economical development speeds up the adaptation to and anticipation of industrial change becomes an every-day necessity for educational institutions. It is the aim of this study to analyse these processes of adaptation/anticipation in their every-day practice by doing case studies. We choose two single Fachhochschul-programs ('Media Engineering and Design' in Hagenberg, Upper Austria and 'Electronic Engineering' in Kapfenberg, Styria) and analysed their practice of adapting the curriculum to qualification needs and anticipating changes in their professional field."
