    Wiring a periscope--ocelli, retinula axons, visual neuropils and the ancestrality of sea spiders.

    The Pycnogonida or sea spiders are cryptic, eight-legged arthropods with four median ocelli in a 'periscope' or eye tubercle. In older attempts at reconstructing phylogeny they were Arthropoda incertae sedis, but recent molecular trees placed them as the sister group either to all other euchelicerates or even to all euarthropods. Thus, pycnogonids are among the oldest extant arthropods and hold a key position for the understanding of arthropod evolution. This has stimulated studies of new sets of characters conductive to cladistic analyses, e.g. of the chelifores and of the hox gene expression pattern. In contrast knowledge of the architecture of the visual system is cursory. A few studies have analysed the ocelli and the uncommon "pseudoinverted" retinula cells. Moreover, analyses of visual neuropils are still at the stage of Hanström's early comprehensive works. We have therefore used various techniques to analyse the visual fibre pathways and the structure of their interrelated neuropils in several species. We found that pycnogonid ocelli are innervated to first and second visual neuropils in close vicinity to an unpaired midline neuropil, i.e. possibly the arcuate body, in a way very similar to ancestral euarthropods like Euperipatoides rowelli (Onychophora) and Limulus polyphemus (Xiphosura). This supports the ancestrality of pycnogonids and sheds light on what eyes in the pycnogonid ground plan might have 'looked' like. Recently it was suggested that arthropod eyes originated from simple ocelli similar to larval eyes. Hence, pycnogonid eyes would be one of the early offshoots among the wealth of more sophisticated arthropod eyes

    Comparing simulated 26^{26}Al maps to gamma-ray measurements

The diffuse gamma-ray emission of 26Al^{26}{\rm Al} at 1.8 MeV reflects ongoing nucleosynthesis in the Milky Way, and traces massive-star feedback in the interstellar medium due to its 1 Myr radioactive lifetime. Interstellar-medium morphology and dynamics are investigated in astrophysics through 3D hydrodynamic simulations in fine detail, as only few suitable astronomical probes are available. Aims. We compare a galactic-scale hydrodynamic simulation of the Galaxy's interstellar medium, including feedback and nucleosynthesis, with gamma-ray data on 26Al^{26}{\rm Al} emission in the Milky Way extracting constraints that are only weakly dependent on the particular realisation of the simulation or Galaxy structure. Methods. Due to constraints and biases in both the simulations and the gamma-ray observations, such comparisons are not straightforward. For a direct comparison, we perform maximum likelihood fits of simulated sky maps as well as observation-based maximum entropy maps to measurements with INTEGRAL/SPI. To study general morphological properties, we compare the scale heights of 26Al^{26}{\rm Al} emission produced by the simulation to INTEGRAL/SPI measurements.} Results. The direct comparison shows that the simulation describes the observed inner Galaxy well, but differs significantly from the observed full-sky emission morphology. Comparing the scale height distribution, we see similarities for small scale height features and a mismatch at larger scale heights.     Dissecting a neuron network: FIB-SEM-based 3D-reconstruction of the visual neuropils in the sea spider Achelia langi (Dohrn, 1881) (Pycnogonida)

    Background: The research field of connectomics arose just recently with the development of new three-dimensional- electron microscopy (EM) techniques and increasing computing power. So far, only a few model species (for example, mouse, the nematode Caenorhabditis elegans, and the fruit fly Drosophila melanogaster) have been studied using this approach. Here, we present a first attempt to expand this circle to include pycnogonids, which hold a key position for the understanding of arthropod evolution. The visual neuropils in Achelia langi are studied using a focused ion beam-scanning electron microscope (FIB-SEM) crossbeam-workstation, and a three-dimensional serial reconstruction of the connectome is presented. Results: The two eyes of each hemisphere of the sea spider's eye tubercle are connected to a first and a second visual neuropil. The first visual neuropil is subdivided in two hemineuropils, each responsible for one eye and stratified into three layers. Six different neuron types postsynaptic to the retinula (R-cells) axons are characterized by their morphology: five types of descending unipolar neurons and one type of ascending neurons. These cell types are also identified by Golgi impregnations. Mapping of all identifiable chemical synapses indicates that the descending unipolar neurons are postsynaptic to the R-cells and, hence, are second-order neurons. The ascending neurons are predominantly presynaptic and sometimes postsynaptic to the R-cells and may play a feedback role. Conclusions: Comparing these results with the compound eye visual system of crustaceans and insects - the only arthropod visual system studied so far in such detail - we found striking similarities in the morphology and synaptic organization of the different neuron types. Hence, the visual system of pycnogonids shows features of both chelicerate median and mandibulate lateral eyes

    Akurasi Pengukuran Lebar Retak Permukaan Pada Beton Menggunakan Portable Scanner Dengan Variasi Resolusi Pemindaian

    Beton merupakaan salah satu bahan yang paling sering digunakan di dunia konstruksi. Dalam pelaksanaannya di lapangan tidak menutup kemungkinan terjadi kegagalan. Salah satu kegagalan yang terjadi adalah keretakan pada beton (crack). Metode Scanning Image Analysis dilakukan untuk mengevaluasi struktur beton dengan cara memindai permukaan beton yang mengalami keretakan menggunakan alat portable scanner dengan bantuan software pengolah gambar untuk memprosesnya. Pada portable scanner ini, dilakukan pengukuran dengan tiga variasi resolusi pemindaian yaitu 300 dpi, 600 dpi dan 900 dpi. Penelitian dilakukan dimana posisi retak berada di posisi bawah, karena posisi bawah inilah yang paling sulit dilakukan di lapangan. Kemudian hasilnya akan dibandingkan dengan lebar aktualnya. Tujuan penelitian ini adalah untuk mengetahui tingkat akurasi hasil pengukuran portable scanner dibandingkan dengan microcrack detector dan jangka sorong pada pengukuran lebar retak permukaan beton dan untuk mengetahui pengaruh variasi resolusi pemindaian pada portable scanner terhadap pengukuran lebar retak permukaan pada beton. Tingkat akurasi pengukuran menggunakan portable scanner ditunjukkan dengan nilai kesalahan relatif. Kesalahan relatif tertinggi pada hasil pengukuran lebar retak menggunakan portable scanner yaitu 3,350% terhadap microcrack detector dan 4,680% terhadap jangka sorong. Dari data tersebut, dapat dilihat bahwa nilai kesalahan relatif kurang dari 5% sehingga akurasi masih di dalam batas toleransi. Berdasarkan analisis varian dengan uji F untuk hasil pengukuran menggunakan portable scanner antar resolusi pemindaian terhadap hasil pengukuran lebar retak menunjukkan Fhitung < Ftabel artinya tidak terdapat pengaruh resolusi pemindaian portable scanner pada hasil pengukuran lebar retak

    Quantum rejection sampling

    Rejection sampling is a well-known method to sample from a target distribution, given the ability to sample from a given distribution. The method has been first formalized by von Neumann (1951) and has many applications in classical computing. We define a quantum analogue of rejection sampling: given a black box producing a coherent superposition of (possibly unknown) quantum states with some amplitudes, the problem is to prepare a coherent superposition of the same states, albeit with different target amplitudes. The main result of this paper is a tight characterization of the query complexity of this quantum state generation problem. We exhibit an algorithm, which we call quantum rejection sampling, and analyze its cost using semidefinite programming. Our proof of a matching lower bound is based on the automorphism principle which allows to symmetrize any algorithm over the automorphism group of the problem. Our main technical innovation is an extension of the automorphism principle to continuous groups that arise for quantum state generation problems where the oracle encodes unknown quantum states, instead of just classical data. Furthermore, we illustrate how quantum rejection sampling may be used as a primitive in designing quantum algorithms, by providing three different applications. We first show that it was implicitly used in the quantum algorithm for linear systems of equations by Harrow, Hassidim and Lloyd. Secondly, we show that it can be used to speed up the main step in the quantum Metropolis sampling algorithm by Temme et al.. Finally, we derive a new quantum algorithm for the hidden shift problem of an arbitrary Boolean function and relate its query complexity to "water-filling" of the Fourier spectrum.Comment: 19 pages, 5 figures, minor changes and a more compact style (to appear in proceedings of ITCS 2012

    GPAQ-R: development and psychometric properties of a version of the general practice assessment questionnaire for use for revalidation by general practitioners in the UK.

    BACKGROUND: The General Practice Assessment Questionnaire (GPAQ) has been widely used to assess patient experience in general practice in the UK since 2004. In 2013, new regulations were introduced by the General Medical Council (GMC) requiring UK doctors to undertake periodic revalidation, which includes assessment of patient experience for individual doctors. We describe the development of a new version of GPAQ - GPAQ-R which addresses the GMC's requirements for revalidation as well as additional NHS requirements for surveys that GPs may need to carry out in their own practices. METHODS: Questionnaires were given out by doctors or practice staff after routine consultations in line with the guidance given by the General Medical Council for surveys to be used for revalidation. Data analysis and practice reports were provided independently. RESULTS: Data were analysed for questionnaires from 7258 patients relating to 164 GPs in 29 general practices. Levels of missing data were generally low (typically 4.5-6%). The number of returned questionnaires required to achieve reliability of 0.7 were around 35 for individual doctor communication items and 29 for a composite score based on doctor communication items. This suggests that the responses to GPAQ-R had similar reliability to the GMC's own questionnaire and we recommend 30 completed GPAQ-R questionnaires are sufficient for revalidation purposes. However, where an initial screen raises concern, the survey might be repeated with 50 completed questionnaires in order to increase reliability. CONCLUSIONS: GPAQ-R is a development of a well-established patient experience questionnaire used in general practice in the UK since 2004. This new version can be recommended for use in order to meet the UK General Medical Council's requirements for surveys to be used in revalidation of doctors. It also meets the needs of GPs to ask about patient experience relating to aspects of practice care that are not specific to individual general practitioners (e.g. receptionists, telephone access) which meet other survey requirements of the National Health Service in England. Use of GPAQ-R has the potential to reduce the number of surveys that GPs need to carry out in their practices to meet the various regulatory requirements which they face

    Mimicking Asymptomatic Infection?

    The protozoan parasite Giardia duodenalis is responsible for more than 280 million cases of gastrointestinal complaints (“giardiasis”) every year, worldwide. Infections are acquired orally, mostly via uptake of cysts in contaminated drinking water. After transformation into the trophozoite stage, parasites start to colonize the duodenum and upper jejunum where they attach to the intestinal epithelium and replicate vegetatively. Outcome of Giardia infections vary between individuals, from self-limiting to chronic, and asymptomatic to severely symptomatic infection, with unspecific gastrointestinal complaints. One proposed mechanism for pathogenesis is the breakdown of intestinal barrier function. This has been studied by analyzing trans-epithelial electric resistances (TEER) or by indicators of epithelial permeability using labeled sugar compounds in in vitro cell culture systems, mouse models or human biopsies and epidemiological studies. Here, we discuss the results obtained mainly with epithelial cell models to highlight contradictory findings. We relate published studies to our own findings that suggest a lack of barrier compromising activities of recent G. duodenalis isolates of assemblage A, B, and E in a Caco-2 model system. We propose that this epithelial cell model be viewed as mimicking asymptomatic infection. This view will likely lead to a more informative use of the model if emphasis is shifted from aiming to identify Giardia virulence factors to defining non- parasite factors that arguably appear to be more decisive for disease
