Telomeres cluster de novo before the initiation of synapsis: a three-dimensional spatial analysis of telomere positions before and during meiotic prophase.
We have analyzed the progressive changes in the spatial distribution of telomeres during meiosis using three-dimensional, high resolution fluorescence microscopy. Fixed meiotic cells of maize (Zea mays L.) were subjected to in situ hybridization under conditions that preserved chromosome structure, allowing identification of stage-dependent changes in telomere arrangements. We found that nuclei at the last somatic prophase before meiosis exhibit a nonrandom, polarized chromosome organization resulting in a loose grouping of telomeres. Quantitative measurements on the spatial arrangements of telomeres revealed that, as cells passed through premeiotic interphase and into leptotene, there was an increase in the frequency of large telomere-to-telomere distances and a decrease in the bias toward peripheral localization of telomeres. By leptotene, there was no obvious evidence of telomere grouping, and the large, singular nucleolus was internally located, nearly concentric with the nucleus. At the end of leptotene, telomeres clustered de novo at the nuclear periphery, coincident with a displacement of the nucleolus to one side. The telomere cluster persisted throughout zygotene and into early pachytene. The nucleolus was adjacent to the cluster at zygotene. At the pachytene stage, telomeres rearranged again by dispersing throughout the nuclear periphery. The stage-dependent changes in telomere arrangements are suggestive of specific, active telomere-associated motility processes with meiotic functions. Thus, the formation of the cluster itself is an early event in the nuclear reorganizations associated with meiosis and may reflect a control point in the initiation of synapsis or crossing over
Paradoxes of Property: Piracy and Sharing in Information Capitalism
All societies both suffer and benefit from levels of what is perceived as disorder, and the guiding principles of the society may be contradictory, or paradoxical, in that their ordering systems create disorder. Our aim in this text is explore the disorders and vagaries of property that seem essential to its continuance, construction and destruction, and then demonstrate how these paradoxes play out in the information economy in particular within the domain of peer-to-peer (P2P) file-sharing. We do not wish to reduce these paradoxes and contradictions to a temporary error or to a future ordered synthesis, but to take them as they are in all their splintered fury. Much contemporary social action stems from these incoherencies, and the disputes, displays of power, and innovations which circle around them. In the P2P field the disorder generated by the order of property provides opportunities for new productive and adaptive social and technical forms of life to emerge. By contrasting order and disorder we are not implying the necessary existence of a binary distinction between the two, or that those definitions of order and disorder will not change depending on the social position of the definers. Disorder is not always and everywhere the same. It resists definition, which adds to its effect

We shall argue that property is constituted within a paradoxical field of vague boundaries, personal relations, poetry and violence. It is not constituted by a set of universal rules (although rules may grow around it), but by ongoing culturally negotiated, psychologically based and enforced categorisations, disorders and persuasions. Consequently, there are many different types of ‘property’ and relations we can define as ‘ownership’ across different cultures. We use anarchism because it notices the violence of property, anthropology because it notices the strangeness and variety of property, and alchemy as a way of thinking about transformations, that make or undermine property
Attachment styles and personal growth following romantic breakups: The mediating roles of distress, rumination, and tendency to rebound

Study 1 (N = 411) and Study 2 (N = 465) measured attachment style, breakup distress, and personal growth; Study 2 additionally measured ruminative reflection, brooding, and proclivity to rebound with new partners. Structural equation modelling revealed in both studies that anxiety was indirectly associated with greater personal growth through heightened breakup distress, whereas avoidance was indirectly associated with lower personal growth through inhibited breakup distress. Study 2 further showed that the positive association of breakup distress with personal growth was accounted for by enhanced reflection and brooding, and that anxious individuals’ greater personal growth was also explained by their proclivity to rebound. These findings suggest that anxious individuals’ hyperactivated breakup distress may act as a catalyst for personal growth by promoting the cognitive processing of breakup-related thoughts and emotions, whereas avoidant individuals’ deactivated distress may inhibit personal growth by suppressing this cognitive work
Multiple measures are needed to quantify training loads in professional rugby league
To investigate the effect of training mode (conditioning and skills) on multivariate training load relationships in professional rugby league via principal component analysis. Four measures of training load (internal: heart rate exertion index, session rating of perceived exertion; external: PlayerLoadâ„¢, individualised high-speed distance) were collected from 23 professional male rugby league players over the course of one 12-wk preseason period. Training was categorised by mode (skills or conditioning) and then subjected to a principal component analysis. Extraction criteria were set at an eigenvalue of greater than 1. Modes that extracted more than 1 principal component were subject to a Varimax rotation. Skills extracted 1 principal component, explaining 57% of the variance. Conditioning extracted 2 principal components (1st: internal; 2nd: external), explaining 85% of the variance. The presence of multiple training load dimensions (principal components) during conditioning training provides further evidence of the influence of training mode on the ability of individual measures of external or internal training load to capture training variance. Consequently, a combination of internal- and external- training load measures is required during certain training modes
Presynaptic actions of 4-Aminopyridine and γ-aminobutyric acid on rat sympathetic ganglia in vitro
Responses to bath-applications of 4-aminopyridine (4-AP) and -aminobutyric acid (GABA) were recorded intracellularly from neurones in the rat isolated superior cervical ganglion.
4-aminopyridine (0.1–1.0 mmol/l) usually induced spontaneous action potentials and excitatory postsynaptic potentials (EPSPs), which were blocked by hexamethonium. Membrane potential was unchanged; spike duration was slightly increased. Vagus nerve B-and C-fibre potentials were prolonged.
In 4-AP solution (0.1–0.3 mmol/l), GABA (0.1 mmol/l), 3-aminopropanesulphonic acid or muscimol evoked bursts of spikes and EPSPs in addition to a neuronal depolarization. These bursts, which were not elicited by glycine, glutamate, taurine or (±)-baclofen, were completely antagonised by hexamethonium, tetrodotoxin or bicuculline methochloride.
It is concluded that: (a) 4-AP has a potent presynaptic action on sympathetic ganglia; (b) presynaptic actions of GABA can be recorded postsynaptically in the presence of 4-AP; and (c) the presynaptic GABA-receptors revealed in this condition are similar to those on the postsynaptic membrane
Increasing the satisfaction of general practitioners with continuing medical education programs: A method for quality improvement through increasing teacher-learner interaction
BACKGROUND: Continuing medical education (CME) for general practitioners relies on specialist-based teaching methods in many settings. Formal lectures by specialists may not meet the learning needs of practitioners and may cause dissatisfaction with traditional CME. Increasing learner involvement in teaching programs may improve learner satisfaction. METHODS: A quality improvement program for CME for 18 general practitioners in the Tel Aviv region was designed as a result of dissatisfaction with traditional CME activities. A two-step strategy for change was developed. The CME participants first selected the study topics relevant to them from a needs assessment and prepared background material on the topics. In the second step, specialist teachers were invited to answer questions arising from the preparation of selected topics. Satisfaction with the traditional lecture program and the new participatory program were assessed by a questionnaire. The quality criteria included the relevance, importance and applicability of the CME topic chosen to the participant's practice, the clarity of the presentation and the effective use of teaching aids by the lecturer and the potential of the lecturer to serve as a consultant to the participant. RESULTS: The participatory model of CME significantly increased satisfaction with relevance, applicability and interest in CME topics compared to the traditional lecture format. CONCLUSIONS: Increased learner participation in the selection and preparation of CME topics, and increased interaction between CME teachers and learners results in increased satisfaction with teaching programs. Future study of the effect of this model on physician performance is required
Multi-Scale Simulation Modeling for Prevention and Public Health Management of Diabetes in Pregnancy and Sequelae
Diabetes in pregnancy (DIP) is an increasing public health priority in the
Australian Capital Territory, particularly due to its impact on risk for
developing Type 2 diabetes. While earlier diagnostic screening results in
greater capacity for early detection and treatment, such benefits must be
balanced with the greater demands this imposes on public health services. To
address such planning challenges, a multi-scale hybrid simulation model of DIP
was built to explore the interaction of risk factors and capture the dynamics
underlying the development of DIP. The impact of interventions on health
outcomes at the physiological, health service and population level is measured.
Of particular central significance in the model is a compartmental model
representing the underlying physiological regulation of glycemic status based
on beta-cell dynamics and insulin resistance. The model also simulated the
dynamics of continuous BMI evolution, glycemic status change during pregnancy
and diabetes classification driven by the individual-level physiological model.
We further modeled public health service pathways providing diagnosis and care
for DIP to explore the optimization of resource use during service delivery.
