    Evaluation of different rotary devices on bone repair in rabbits

    In oral surgery, the quality of bone repair may be influenced by several factors that can increase the morbidity of the procedure. The type of equipment used for ostectomy can directly affect bone healing. The aim of this study was to evaluate bone repair of mandible bone defects prepared in rabbits using three different rotary devices. Fifteen New Zealand rabbits were randomly assigned to 3 groups (n=5) according to type of rotary device used to create bone defects: I - pneumatic low-speed rotation engine, II - pneumatic high-speed rotation engine, and III - electric low-speed rotation engine. The anatomic pieces were surgically obtained after 2, 7 and 30 days and submitted to histological and morphometric analysis. The morphometric results were expressed as the total area of bone remodeling matrix using an image analysis system. Increases in the bone remodeling matrix were noticed with time along the course of the experiment. No statistically significant differences (p>0.05) were observed among the groups at the three sacrificing time points considering the total area of bone mineralized matrix, although the histological analysis showed a slightly advanced bone repair in group III compared to the other two groups. The findings of the present study suggest that the type of rotary device used in oral and maxillofacial surgery does not interfere with the bone repair process.A ostectomia é uma manobra cirúrgica fundamental que pode afetar a reparação tecidual de modo a aumentar a morbidade do procedimento. O tipo de equipamento e/ou instrumental utilizado para a ostectomia pode influenciar diretamente no reparo ósseo. O objetivo deste trabalho foi analisar o processo de reparação óssea em defeitos realizados em mandíbula de coelho com três diferentes equipamentos. Quinze coelhos foram aleatoriamente divididos em 3 grupos (n=5) de acordo com o equipamento usado para preparacão de 3 cavidades ósseas com brocas padronizadas: I) motor de baixa rotação pneumático, II) motor de alta rotação pneumático e III) motor de baixa rotação elétrico, todos com refrigeração constante. Após os períodos experimentais de 2, 7e 30 dias, as peças anatômicas foram removidas e submetidas a análises microscópica e histomorfométrica. Não houve diferença estatisticamente significante (p>0,05) entre os grupos nos 3 períodos experimentais, considerando-se a área total de matriz óssea mineralizada, embora a análise microscópica tenha revelado uma tendência a um processo de reparação óssea um pouco mais adiantado no grupo III, quando comparado aos demais grupos.     A Ecologia da Informação na Definição de Domínios

    Todo e qualquer sistema de informação existe dentro de um determinado contexto organizacional. As organizações têm os seus próprios objectivos estratégicos, recursos humanos, meios tecnológicos e financeiros que a tornam única entre os seus pares. A arquitectura de uma organização tem então de ser igualmente uma estrutura singular, adaptada à realidade que pretende representar num modelo de dados. É a base para a troca de informação entre os vários centros de produção da organização, pelo que o vocabulário organizacional tem que ser compreendido por todos os agentes informativos. Caso haja uma dissonância nos canais de comunicação a informação deixa de fluir num formato inteligível, tornando os agentes em antagonistas, em vez do pretendido trabalho cooperativo. A correcta definição dos domínios é assim um trabalho fundamental e que deve seguir uma perspectiva abrangente. Este artigo apresenta uma breve introdução teórica à aplicação da ecologia à construção de domínios mais contextualizados

    Lipoid proteinosis

    OBJETIVO: Apresentar as características clínicas, tratamento cirúrgico e achado histológico de um caso de lipoidoproteinose. DESCRIÇÃO DO CASO: Criança do sexo masculino, cinco anos de idade, branco, que procurou atendimento odontológico na Universidade. A mãe da criança relatou presença de intensa halitose e dificuldade na alimentação e higienização bucal, decorrentes de crescimento gengival generalizado nos arcos dentários superior e inferior. No exame clínico, verificaram-se comprometimento funcional e estético generalizado (rouquidão, artralgia bilateral no joelho e tornozelo, lesões tumorais nas orelhas, entre outros), além de extensa hiperplasia gengival em ambos os arcos dentários. Optou-se pelo tratamento cirúrgico, com remoção do tecido hiperplásico e exodontia de todos os dentes decíduos e de dois permanentes. O exame histopatológico da peça cirúrgica confirmou o diagnóstico de lipoidoproteinose. COMENTÁRIOS: A lipoidoproteinose é uma doença rara caracterizada pela deposição da substância hialina na pele, membranas mucosas e nos órgãos internos. Os sinais que podem surgir após o nascimento, são: rouquidão; lesões pápulo-nodulares na cabeça, pescoço e membros; lesões papulares amareladas nas margens das pálpebras. O curso desta doença é benigno e crônico.OBJECTIVE: Report clinical characteristics, histological findings and treatment of a patient with lipoid proteinosis. CASE DESCRIPTION: Five year-old male patient was assisted in the university clinic for dental treatment. The mother reported the presence of intense halitosis and difficulties for feeding and oral hygiene due to extensive gingival hyperplasia on the maxillary and mandibular arches. Clinical examination revealed generalized functional and esthetic involvement, including hoarseness; bilateral knee and ankle arthralgia; tumoral lesions on the ears and gingival hyperplasia throughout the extent of both dental arches. The surgical treatment removed the hyperplastic tissue and all deciduous teeth, as well as two permanents teeth. Histopathological examination of the specimen confirmed the diagnosis of lipoid proteinosis. COMMENTS: Lipoid proteinosis is a rare disorder characterized by progressive deposition of hyaline material in the skin, mucous membranes and organs. Clinical signs appear after birth and are characterized by hoarseness, papulonodular lesions on the head, neck and limbs and yellowish papules on the eyelid margins, among others. The disease has a benign and chronic evolution

    Physical, chemical, and biological properties of white MTA with additions of AlF<sub>3</sub>

No. of bitstreams: 0 Previous issue date: 2018-04-13Objectives: Addition of aluminum fluoride (AlF3) to MTA was tested to inhibit dental discoloration. Materials and methods: MTA Angelus with 0, 5, 15, and 45% AlF3 were tested. The set cements were characterized using scanning electron microscopy, energy-dispersive spectroscopy, and X-ray diffraction. Radiopacity and setting time were analyzed according to ANSI/ADA 57 and ASTM C266-08. Volume change was evaluated using volumetric micro-CT analysis. The pH and calcium ion release were assessed after 3 and 24 h and 28 days. Dental discoloration in contact with the cements was assessed after 24 h and 28 and 90 days of contact with bovine and human dentine. Tissue reaction to subcutaneous implantation in rats was examined after 30 and 60 days. Results: AlF3 altered the microstructure of MTA. The addition of 5% AlF3 did not significantly alter the radiopacity, setting time, and volume change (p > 0.05). pH and calcium ion release significantly increased with addition of AlF3 (p > 0.05). All the tested proportions of AlF3 prevented the dental darkening verified for MTA Angelus in bovine and human teeth. AlF3 did not interfere in inflammatory response of MTA in all periods of analysis; otherwise, lower amounts showed less intense inflammatory infiltrate. Clinical relevance: AlF3 prevents destabilization of bismuth oxide and consequent tooth darkening, frequently verified in clinical practice when using white MTA.     New Surgical Model for Bone–Muscle Injury Reveals Age and Gender-Related Healing Patterns in the 5 Lipoxygenase (5LO) Knockout Mouse

    Signaling lipid mediators released from 5 lipoxygenase (5LO) pathways influence both bone and muscle cells, interfering in their proliferation and differentiation capacities. A major limitation to studying inflammatory signaling pathways in bone and muscle healing is the inadequacy of available animal models. We developed a surgical injury model in the vastus lateralis (VL) muscle and femur in 129/SvEv littermates mice to study simultaneous musculoskeletal (MSK) healing in male and female, young (3 months) and aged (18 months) WT mice compared to mice lacking 5LO (5LOKO). MSK defects were surgically created using a 1-mm punch device in the VA muscle followed by a 0.5-mm round defect in the femur. After days 7 and 14 post-surgery, the specimens were removed for microtomography (microCT), histopathology, and immunohistochemistry analyses. In addition, non-injured control skeletal muscles along with femur and L5 vertebrae were analyzed. Bones were microCT phenotyped, revealing that aged female WT mice presented reduced BV/TV and trabecular parameters compared to aged males and aged female 5LOKO mice. Skeletal muscles underwent a customized targeted lipidomics investigation for profiling and quantification of lipid signaling mediators (LMs), evidencing age, and gender related-differences in aged female 5LOKO mice compared to matched WT. Histological analysis revealed a suitable bone-healing process with osteoid deposition at day 7 post-surgery, followed by woven bone at day 14 post-surgery, observed in all young mice. Aged WT females displayed increased inflammatory response at day 7 post-surgery, delayed bone matrix maturation, and increased TRAP immunolabeling at day 14 post-surgery compared to 5LOKO females. Skeletal muscles of aged animals showed higher levels of inflammation in comparison to young controls at day 14 post-surgery; however, inflammatory process was attenuated in aged 5LOKO mice compared to aged WT. In conclusion, this new model shows that MSK healing is influenced by age, gender, and the 5LO pathway, which might serve as a potential target to investigate therapeutic interventions and age-related MSK diseases. Our new model is suitable for bone–muscle crosstalk studies

    A instalação de implantes osseointegrados em rebordos atróficos reconstruídos com a utilização de enxertos ósseos autógenos vem sendo amplamente utilizada, principalmente com a adoção de dois tempos cirúrgicos . Neste estudo, buscou-se, à luz da microscopia óptica, avaliar o reparo e a qualidade óssea obtidos em rebordos maxilares que receberam enxertos de duas zonas doadoras, a crista ilíaca e o mento. Para tanto, dez pacientes submetidos à este tipo de reconstrução integraram a amostra. No período de quatro meses após este procedimento cirúrgico, no momento da colocação dos implantes, foram obtidos cinco espécimes de cada tipo de enxerto, por meio de biópsias realizadas com brocas trefinas. As peças cirúrgicas passaram por procedimento histotécnico de rotina e foram coradas pelas técnicas de H.E. e Tricrômico de Mallory. Como resultados, verificou-se tecido ósseo em franco processo de remodelação, evidenciado pelas numerosas áreas de osteogênese presentes em ambos os enxertos. Da análise das superfícies de matriz óssea mineralizada obteviveram-se valores significativamente maiores para as regiões que receberam enxerto de mento. Desta forma, concluiu-se que o período de quatro meses se mostrou suficiente para a incorporação dos enxertos, sendo viável a instalação dos implantes neste momento. Quanto à qualidade óssea, os enxertos de mento apresentaram-se superiores aos de ilíaco, possivelmente devido à sua arquitetura original.The insertion of endosseous implants in atrophic ridges reconstructed with autogenous bone grafts has been extensively used, most of the times using two-stage technique. In this study, bone graft repair and bone quality achieved in maxillary ridges reconstructed using iliac crest and chin bone grafts were analysed under light microscopy. Ten patients who underwent this procedure were observed in this study. After 4 months of the bone graft surgery, at the moment of the implants installation, 5 specimens were obtained from biopsies made in each bone graft group using trephine burs. The specimens underwent routine histotechnical procedure and stained by H.E. and Mallory triple stain. As a result, process of bone turnover was revealed by the areas of intense osteogenesis. From the analysis of mineralized bone tissue surface areas, significant greater values were verified in maxillary ridge regions reconstructed with chin graft. From these results, it was concluded that the period of 4 months was sufficient to the incorporation of the grafts. It was also concluded that chin grafts presented better bone quality than iliac crest grafts
