This study aimed to determine the best combination of lotus (Nelumbo nucifera) tempeh making and the composition of microbes that live in fermented lotus seeds. This study has used 2 stages of research design; the first stage was optimizing treatment of lotus tempeh processing using the Response Surface Methodology (RSM) Box Behnken design with Minitab software version 19, then the total fungi, total BAL (bacteria acid lactat), activity water (Aw) analyzed by descriptive. The best treatment were 0.5 g yeast, 24 h soaking time, 36 h fermentation time; and 0.5 g yeast, 6 h soaking time, 48 h fermentation time. The best treatments have resulted 6.58 log cfu/gr - 8.59 log cfu/gr of total BAL.  In addition, the total fungi ranged from 5.30 ± 0.64 log cfu/gr  to  5.34 ± 0.64 log cfu/gr; pH was 4.6 up to 5.7, and 0.86 of water activity. This study aimed to determine the best combination of lotus (Nelumbo nucifera) tempeh making and the composition of microbes that live in fermented lotus seeds.
