    Thermo-programmed synthetic DNA-based receptors

    Herein, we present a generalizable and versatile strategy to engineer synthetic DNA ligand-binding devices that can be programmed to load and release a specific ligand at a defined temperature. We do so by re-engineering two model DNA-based receptors: a triplex-forming bivalent DNA-based receptor that recognizes a specific DNA sequence and an ATP-binding aptamer. The temperature at which these receptors load/release their ligands can be finely modulated by controlling the entropy associated with the linker connecting the two ligand-binding domains. The availability of a set of receptors with tunable and reversible temperature dependence allows achieving complex load/release behavior such as sustained ligand release over a wide temperature range. Similar programmable thermo-responsive synthetic ligand-binding devices can be of utility in applications such as drug delivery and production of smart materials

    Two seas for one great diversity: Checklist of the marine heterobranchia (Mollusca; Gastropoda) from the Salento Peninsula (South-East Italy)

    The Salento peninsula is a portion of the Italian mainland separating two distinct Mediterranean basins, the Ionian and the Adriatic seas. Several authors have studied the marine Heterobranchia (Mollusca, Gastropoda) fauna composition living in the Ionian Sea, but to date further knowledge regarding this interesting group of mollusks is still needed. Recent studies have corroborated the peculiarity of the Mediterranean Sea showing high levels of endemism and cryptic diversity. On the other hand, marine sea slugs have been revealed to be important indicators of the marine ecosystem's health, due to their species-specific diet that consist of a vast variety of sessile and benthic invertebrates. A baseline study of the marine Heterobranchia diversity is therefore a necessary step to reveal the hidden diversity and to monitor the possible presence of alien species. The present study shows results from approximately 600 scientific dives carried out during a nine-year period in all of the main submarine habitats of the studied area, while accounting for the marine Heterobranchia from both the Ionian and Adriatic Seas. With this contribution, the list of marine Heterobranchia inhabiting the Salento Peninsula rises to 160. Furthermore, it also reports, for the first time, the presence of one alien species and three new records for Italian waters. Ecological notes and geographical distribution for each added species are provided together with animal iconography, consisting mainly of in situ photographs, for species identification

    Inhibition of acetylpolyamine and spermine oxidases by the polyamine analogue chlorhexidine

    ""Acetylpolyamine and spermine oxidases are involved in the catabolism of polyamines. The discovery of selective. inhibitors of these enzymes represents an important tool for the development of novel anti-neoplastic drugs. Here, a. comparative study on acetylpolyamine and spermine oxidases inhibition by the polyamine analogue chlorhexidine. is reported. Chlorhexidine is an antiseptic diamide, commonly used as a bactericidal and bacteriostatic agent.. Docking simulations indicate that chlorhexidine binding to these enzymes is compatible with the stereochemical. properties of both acetylpolyamine oxidase and spermine oxidase active sites. In fact, chlorhexidine is predicted. to establish several polar and hydrophobic interactions with the active site residues of both enzymes, with binding. energy values ranging from −7.6 to −10.6 kcal\\\/mol. In agreement with this hypothesis, inhibition studies indicate that. chlorhexidine behaves as a strong competitive inhibitor of both enzymes, values of Ki being 0.10 ÎŒM and 0.55 ÎŒM for. acetylpolyamine oxidase and spermine oxidase, respectively."

    Switching the aptamer attachment geometry can dramatically alter the signalling and performance of electrochemical aptamer-based sensors

    Electrochemical aptamer-based (EAB) sensors, composed of an electrode-bound DNA aptamer with a redox reporter on the distal end, offer the promise of high-frequency, real-time molecular measurements in complex sample matrices and even in vivo. Here we assess the extent to which switching the aptamer terminus that is electrode-bound and the one that is redox-reporter-modified affects the performance of these sensors. Using sensors against doxorubicin, cocaine, and vancomycin as our test beds, we find that both signal gain (the relative signal change seen in the presence of a saturating target) and the frequency dependence of gain depend strongly on the attachment orientation, suggesting that this easily investigated variable is a worthwhile parameter to optimize in the design of new EAB sensors

    Excited State Destri - De Vega Equation for Sine-Gordon and Restricted Sine-Gordon Models

    We derive a generalization of the Destri - De Vega equation governing the scaling functions of some excited states in the Sine-Gordon theory. In particular configurations with an even number of holes and no strings are analyzed and their UV limits found to match some of the conformal dimensions of the corresponding compactified massless free boson. Quantum group reduction allows to interpret some of our results as scaling functions of excited states of Restricted Sine-Gordon theory, i.e. minimal models perturbed by phi_13 in their massive regime. In particular we are able to reconstruct the scaling functions of the off-critical deformations of all the scalar primary states on the diagonal of the Kac-table.Comment: Latex, 12 page

    Cardoon meal as alternative protein source to soybean meal for limousine bulls fattening period: Effects on growth performances and meat quality traits

    SIMPLE SUMMARY: Beef cattle feeding strategies are commonly based on soybean meal utilization as a fundamental protein source. This feed, though, might have negative environmental impacts on the major areas of production and is becoming very expensive. Cardoon (Cynara cardunculus L.) is a resilient crop which represents a good opportunity in reclaiming and remediating unutilized lands. Cardoon seeds are rich in oil, which is extracted for industrial purposes, and the related by-products (press cake and meal) are characterized by high protein content and essential fatty acids. The aim of this study was to evaluate cardoon meal as a protein source during the Limousine bulls’ fattening period, in order to study a suitable alternative to develop and create low-input and low-emission feeding strategies. The results obtained in terms of growth performances showed no statistical difference between bulls fed the by control diet (containing soybean meal as main protein source) and animals fed by the experimental diet, where soybean meal was partially replaced by one containing cardoon meal. Meat quality traits were measured, and no differences between the groups were found. Hence, these by-products could be considered as a valuable solution in Limousine bulls’ fattening periods and could be used to represent a key factor to improve cattle-feeding sustainability. ABSTRACT: Soybean meal is the most important protein source in beef cattle feeding. The research of alternative protein sources to replace soy use, avoiding negative effects on in vivo performance and on the product’s quality, is an important issue. In this context, cardoon represents a non-OGM resilient crop that can be cultivated in marginal lands for extracting its seed oil (utilized for biodiesel and biodegradable bioplastic production) and whose and the residual meal from its seed oil (utilized for biodiesel and biodegradable bioplastic production) could be a suitable by-product for animal feeding, due to its fairly high protein content. The aim of this study was to evaluate the feasibility of using cardoon meal as an innovative protein source during the Limousine bulls’ fattening period. Thirty-two bulls were divided into two groups and fed with a diet containing soybean meal (SG) or partially replacing soybean meal with cardoon meal as a protein source (CG), respectively. The feeding trial lasted about 11 months. Growth performances and meat physical–chemical traits were evaluated. No statistical differences in feed efficiency, average daily gain, or in the main meat quality indicators, as well as in fatty acid profiles were found among the groups. Therefore, cardoon meal could be considered as an alternative to soybean meal in fattening Limousine bulls in order to enhance the sustainability of the farming system

    A case study of the application of hand-held mobile laser scanning in the planning of an Italian forest (Alpe Di Catenaia, Tuscany)

    Precision forestry is becoming a key sector for forest planning because it allows complex analyses of forest data to be carried out simply and economically. It contributes to the integration between technicians and operators in the sector by guaranteeing the transparency of the forest management operations (Corona et al., 2017). In the context of the progressive development of technology, we investigated the feasibility of using the hand-held mobile laser scanner (HMLS) system in different types of forest sites and comparison of the characteristics of individual trees (tree height, diameters at breast height) with traditional surveys, applied with the aim to validate the performance of the system for a future alternative methodology for forest planning thanks to the collaboration with the forestry company “Dimensione Ricerca Ecologia Ambiente Italia” (D.R.E.Am. Italia). GEOSLAM ZEB HORIZON ℱ laser scanner is a hand-held mobile laser scanner containing SLAM technology that can be solved the problem of no GNSS signal or poor signal under the forest canopy making it more practical for forest investigations (Gollob et al., 2020). 15 forest sample plots are selected to reflect different stand conditions in Mediterranean forests taking into count the development stage and density of the sub-canopy vegetation, as well as the species composition in the forest stands. The aim of this study is to show the possible extrinsic circumstances that make the method fail by varying the ecological status of forest plots

    On-board and Ground Visual Pose Estimation Techniques for UAV Control

    In this paper, two techniques to control UAVs (Unmanned Aerial Vehicles), based on visual information are presented. The first one is based on the detection and tracking of planar structures from an on-board camera, while the second one is based on the detection and 3D reconstruction of the position of the UAV based on an external camera system. Both strategies are tested with a VTOL (Vertical take-off and landing) UAV, and results show good behavior of the visual systems (precision in the estimation and frame rate) when estimating the helicopterÂżs position and using the extracted information to control the UAV
